Marine Turtle Conservation on the Caribbean Coast of Nicaragua
The purpose of this initial phase was to expand our previous work on marine turtles on
the Caribbean coast of Nicaragua. This was done through the establishment of a collaborative
program with the Miskitu Indians to reduce the uncontrolled take of marine turtles in this region.
We initiated activities for a multi-year program that will include research, training, and
educational activities involving three Miskitu Indian Communities located in the Indigenous
Communities and Miskito Cays Biosphere Reserve (ICMCBR). (5 page document
Influence of Nitrate and Nitrite on Thyroid Hormone Responsive and Stress-Associated Gene Expression in Cultured Rana catesbeiana Tadpole Tail Fin Tissue
Nitrate and nitrite are common aqueous pollutants that are known to disrupt the thyroid axis. In amphibians, thyroid hormone (TH)-dependent metamorphosis is affected, although whether the effect is acceleration or deceleration of this developmental process varies from study to study. One mechanism of action of these nitrogenous compounds is through alteration of TH synthesis. However, direct target tissue effects on TH signaling are hypothesized. The present study uses the recently developed cultured tail fin biopsy (C-fin) assay to study possible direct tissue effects of nitrate and nitrite. Tail biopsies obtained from premetamorphic Rana catesbeiana tadpoles were exposed to 5 and 50 mg/L nitrate (NO3–N) and 0.5 and 5 mg/L nitrite (NO2–N) in the absence and presence of 10 nM T3. Thyroid hormone receptor β (TRβ) and Rana larval keratin type I (RLKI), both of which are TH-responsive gene transcripts, were measured using quantitative real time polymerase chain reaction. To assess cellular stress which could affect TH signaling and metamorphosis, heat shock protein 30, and catalase (CAT) transcript levels were also measured. We found that nitrate and nitrite did not significantly change the level of any of the four transcripts tested. However, nitrate exposure significantly increased the heteroscedasticity in response of TRβ and RLKI transcripts to T3. Alteration in population variation in such a way could contribute to the previously observed alterations of metamorphosis in frog tadpoles, but may not represent a major mechanism of action

Endocrine Disrupting Contaminants—Beyond the Dogma
Descriptions of endocrine disruption have largely been associated with wildlife and driven by observations documenting estrogenic, androgenic, antiandrogenic, and antithyroid actions. These actions, in response to exposure to ecologically relevant concentrations of various environmental contaminants, have now been established in numerous vertebrate species. However, many potential mechanisms and endocrine actions have not been studied. For example, the DDT [1,1,1-trichloro-2,2-bis(p-chlorophenyl)ethane] metabolite, p,p′-DDE [1,1-dichloro-2,2-bis(p-chlorophenyl)ethylene] is known to disrupt prostaglandin synthesis in the uterus of birds, providing part of the explanation for DDT-induced egg shell thinning. Few studies have examined prostaglandin synthesis as a target for endocrine disruption, yet these hormones are active in reproduction, immune responses, and cardiovascular physiology. Future studies must broaden the basic science approach to endocrine disruption, thereby expanding the mechanisms and endocrine end points examined. This goal should be accomplished even if the primary influence and funding continue to emphasize a narrower approach based on regulatory needs. Without this broader approach, research into endocrine disruption will become dominated by a narrow dogma, focusing on a few end points and mechanisms
Altered Breast Development in Young Girls from an Agricultural Environment
In several human populations, the age at which female breast development begins is reported to have declined over the last five decades. Much debate has occurred over whether this reported decline has actually occurred and what factors contribute to it. However, geographical patterns reflecting earlier developmental onset in some human populations suggest environmental factors influence this phenomenon. These factors include interactions between genetic makeup, nutrition, and possible cumulative exposure to estrogens, both endogenous as well as environmental beginning during in utero development. We examined the onset of breast development in a group of peripubertal girls from the Yaqui Valley of Sonora, Mexico. We observed that girls from valley towns, areas using modern agricultural practices, exhibited larger breast fields than those of girls living in the foothills who exhibited similar stature [e.g., weight, height, body mass index (BMI)], and genetic background. Further, girls from valley towns displayed a poorly defined relationship between breast size and mammary gland development, whereas girls from the Yaqui foothills, where traditional ranching occurs, show a robust positive relationship between breast size and mammary size. The differences noted were obtained by a medically based exam involving morphometric analysis and palpation of tissues, in contrast to visual staging alone. In fact, use of the Tanner scale, involving visual staging of breast development for puberty, detected no differences between the study populations. Mammary tissue, determined by palpation, was absent in 18.5% of the girls living in agricultural areas, although palpable breast adipose tissue was present. No relationship was seen between mammary diameter and weight or BMI in either population. These data suggest that future in-depth studies examining mammary tissue growth and fat deposition in breast tissue are required if we are to understand environmental influences on these phenomena
Variation in Nest Temperatures of the American Alligator Found on the Kennedy Space Center Merritt Island National Wildlife Refuge
Information on nest temperatures of the American Alligator (Alligator mississippiensis) constructed in the wild is limited. Nesting temperatures during a critical thermal sensitive period determine the sex of alligators and are therefore critical in establishing the sex biases in recruitment efforts of alligators within a given community. Nest components, varying environmental conditions, and global warming could have a significant impact on nest temperatures, thus affecting future generations of a given population. One hundred and seventy four programmable thermistors were inserted into fifty eight nests from 2010 through 2015 nesting cycles. Three thermistors were placed inside each nest cavity (one on top of the eggs, one in the middle of the eggs, and one at the bottom of the clutch of the eggs) to collect temperature profiles in the incubation chamber and throughout the entire incubation period. One thermistor was also placed near or above these nests to obtain an ambient air temperature profile. Once retrieved, data from these thermistors were downloaded to examine temperature profiles throughout the incubation period as well as during the period of sexual determination. These data would help establish survival rates related to nest temperature and predict sex ratio of recruited neonates at the Kennedy Space Center. Over three million temperatures have been recorded since 2010 for the alligator thermistor study giving us insight to the recruitment efforts found here. Precipitation was the largest influence on nesting temperatures outside of daily photoperiod, with immediate changes of up to eight degrees Celsius
Perfluorinated Alkyl Acids in Plasma of American Alligators (Alligator Mississippiensis) from Florida and South Carolina
This study aimed to quantitate fourteen perfluoroalkyl acids (PFAAs) in 125 adult American alligators at twelve sites across the southeastern US. Of those fourteen PFAAs, nine were detected in 65% - 100% of the samples: PFOA, PFNA, PFDA, PFUnA, PFDoA, PFTriA, PFTA, PFHxS, and PFOS. Males (across all sites) showed significantly higher concentrations of four PFAAs: PFOS (p = 0.01), PFDA (p = 0.0003), PFUnA (p = 0.021), and PFTriA (p = 0.021). Concentrations of PFOS, PFHxS, and PFDA in plasma were significantly different among the sites in each sex. Alligators at Merritt Island National Wildlife Refuge and Kiawah Nature Conservancy both exhibited some of the highest PFOS concentrations (medians 99.5 ng/g and 55.8 ng/g respectively) in plasma measured to date in a crocodilian species. A number of positive correlations between PFAAs and snout-vent length (SVL) were observed in both sexes suggesting PFAA body burdens increase with increasing size. In addition, several significant correlations among PFAAs in alligator plasma may suggest conserved sources of PFAAs at each site throughout the greater study area. This study is the first to report PFAAs in American alligators, reveals potential PFAA hot spots in Florida and South Carolina, and provides and additional contaminant of concern when assessing anthropogenic impacts on ecosystem health
