    Explaining Political Violence Against Civilians in Northern Ireland: A Contention-Oriented Approach

    In contrast to prevalent theories of terrorism, this study develops a contention-oriented approach where levels and forms of political violence against civilians depend upon: (1) the strategies of combatants; (2) the means of contention; (3) the locations of allies and opponents; (4) the collective identities of combatants; and (5) the dynamics of contention, including whether or not representatives of paramilitary organizations are included in formal peace processes. Quantitative analyses of a multi-source database of civilian deaths taking place in Northern Ireland between 1966 and 2006 offer preliminary support for this approach. The study underscores the insights provided by theories and methods used in the fields of social movements research and peace and conflict studies

    Sequential Assembly of Centromeric Proteins in Male Mouse Meiosis

    The assembly of the mitotic centromere has been extensively studied in recent years, revealing the sequence and regulation of protein loading to this chromosome domain. However, few studies have analyzed centromere assembly during mammalian meiosis. This study specifically targets this approach on mouse spermatocytes. We have found that during prophase I, the proteins of the chromosomal passenger complex Borealin, INCENP, and Aurora-B load sequentially to the inner centromere before Shugoshin 2 and MCAK. The last proteins to be assembled are the outer kinetochore proteins BubR1 and CENP-E. All these proteins are not detected at the centromere during anaphase/telophase I and are then reloaded during interkinesis. The loading sequence of the analyzed proteins is similar during prophase I and interkinesis. These findings demonstrate that the interkinesis stage, regularly overlooked, is essential for centromere and kinetochore maturation and reorganization previous to the second meiotic division. We also demonstrate that Shugoshin 2 is necessary for the loading of MCAK at the inner centromere, but is dispensable for the loading of the outer kinetochore proteins BubR1 and CENP-E

    A Typology of Oppositional Knowledge: Democracy and the U.S. Peace Movement

    Institutionally privileged political discourses not only legitimate the policy agendas of power-holders, but also de-legitimate dissent. Oppositional discourses are social movement responses to these cultural obstacles to mass mobilization. Integrating discourse analysis and framing theory, we argue that the production of oppositional knowledge constitutes a long-term, counter-hegemonic project that connects macro-level discourses with meso and micro-level efforts at political persuasion, mobilization, and change. Drawing examples from statements issued by U.S. peace movement organizations (PMOs) over fifteen years, we map the production of oppositional discourses across five conflict periods. Using qualitative data analysis and both inductive and deductive theorizing, we develop a typology of the U.S. peace movement\'s discourses on democracy. We show that four forms of oppositional knowledge were generated by PMOs to facilitate policy dialogue and accountability. Through their statements, peace movement organizations crafted a shared conception of democracy that is antithetical to military intervention abroad and political repression at home.Discourse, Peace and Social Movements, War, Hegemony, Democracy, Symbolic Culture, Political Persuasion

    Decoding Polo-like kinase 1 signaling along the kinetochore–centromere axis

    Protein kinase signaling along the kinetochore-centromere axis is crucial to assure mitotic fidelity, yet its spatial coordination is obscure. Here, we examined how pools of human Polo-like kinase 1 (Plk1) within this axis control signaling events to elicit mitotic functions. To do this, we restricted active Plk1 to discrete subcompartments within the kinetochore-centromere axis using chemical genetics and decoded functional and phosphoproteomic signatures of each. We observe distinct phosphoproteomic and functional roles, suggesting that Plk1 exists and functions in discrete pools along this axis. Deep within the centromere, Plk1 operates to assure proper chromosome alignment and segregation. Thus, Plk1 at the kinetochore is a conglomerate of an observable bulk pool coupled with additional functional pools below the threshold of microscopic detection/resolution. Although complex, this multiplicity of locales provides an opportunity to decouple functional and phosphoproteomic signatures for a comprehensive understanding of Plk1’s kinetochore functions
