    LRS-II: A Specialized Knowledge System For Launch Resource Scheduling

    This research used the Level 5 expert system software to develop a specialized knowledge system called the Launch Resource Scheduling system (LRS-II). LRS-II will be used as a decision aid by USSPACECOM to determine if there is sufficient launch capability to meet future satellite requirements and to quickly assess the impact of contingencies such as launch or on-orbit failures. LRS-II uses multiple knowledge bases to match satellite launch requirements to available launch vehicles, launch pads, and upper stages. Specialized knowledge about resources are stored in dBase III files, specific fields of the satellite record records to schedule the earliest launch requirement. During manifesting, the constraints of satellite and resource availability, site processing time, shuttle mission duration, and satellite on-orbit checkout time are used to insure the selected launch date is accurate

    Associations between Sleep Problems and Anxiety in Youth

    Most of us are able to recall sleepless nights during periods of heightened anxiety, and the associations between sleep and anxiety are sufficiently well established to be acknowledged in the DSM-IV (American Psychiatric Association, 1994). Indeed, sleeprelated difficulties are included in the diagnostic criteria for posttraumatic stress disorder, acute stress disorder and generalized anxiety disorder. In contrast to the wealth of literature highlighting associations between sleep problems and anxiety in adults, less is known about these associations in children and adolescents. It is important to understand associations between disorders as there is evidence to suggest that co-occurring difficulties may result in greater impairment than those occurring alone. For example, a study focusing upon depression found that individuals with co-occurring disorders were more likely to demonstrate suicidal behaviour as compared to those with pure depression (Rohde et al., 1991). Furthermore, focusing upon children is important as various difficulties, including anxiety, may appear early in life and persist into adulthood (Kim- Cohen et al., 2003). This article summarises some key findings with regards to the associations between sleep problems and anxiety in youth and proposes possible clinical implications of this research

    Genome-wide identification of regulatory elements in Sertoli cells

    A current goal of molecular biology is to identify transcriptional networks that regulate cell differentiation. However, identifying functional gene regulatory elements has been challenging in the context of developing tissues where material is limited and cell types are mixed. To identify regulatory sites during sex determination, we subjected Sertoli cells from mouse fetal testes to DNaseI-seq and ChIP-seq for H3K27ac. DNaseI-seq identified putative regulatory sites around genes enriched in Sertoli and pregranulosa cells; however, active enhancers marked by H3K27ac were enriched proximal to only Sertoli-enriched genes. Sequence analysis identified putative binding sites of known and novel transcription factors likely controlling Sertoli cell differentiation. As a validation of this approach, we identified a novel Sertoli cell enhancer upstream of Wt1, and used it to drive expression of a transgenic reporter in Sertoli cells. This work furthers our understanding of the complex genetic network that underlies sex determination and identifies regions that potentially harbor non-coding mutations underlying disorders of sexual development

    Microbiota regulate intestinal epithelial gene expression by suppressing the transcription factor Hepatocyte nuclear factor 4 alpha

    Microbiota influence diverse aspects of intestinal physiology and disease in part by controlling tissue-specific transcription of host genes. However, host genomic mechanisms mediating microbial control of intestinal gene expression are poorly understood. Hepatocyte nuclear factor 4 (HNF4) is the most ancient family of nuclear receptor transcription factors with important roles in human metabolic and inflammatory bowel diseases, but a role in host response to microbes is unknown. Using an unbiased screening strategy, we found that zebrafish Hnf4a specifically binds and activates a microbiota-suppressed intestinal epithelial transcriptional enhancer. Genetic analysis revealed that zebrafish hnf4a activates nearly half of the genes that are suppressed by microbiota, suggesting microbiota negatively regulate Hnf4a. In support, analysis of genomic architecture in mouse intestinal epithelial cells disclosed that microbiota colonization leads to activation or inactivation of hundreds of enhancers along with drastic genome-wide reduction of HNF4A and HNF4G occupancy. Interspecies meta-analysis suggested interactions between HNF4A and microbiota promote gene expression patterns associated with human inflammatory bowel diseases. These results indicate a critical and conserved role for HNF4A in maintaining intestinal homeostasis in response to microbiota

    Optomechanical Properties of Stretched Polymer Dispersed Liquid Crystal Films for Scattering Polarizer Applications

    A scattering polarizer is created by subjecting a polymer dispersed liquid crystal (PDLC) film to tensile strain. The optomechanical properties of the film are investigated by simultaneously measuring the stress-strain and polarization dependent optical transmission characteristics. The correlation between transmittances of two orthogonal polarizations and the stress-strain curve reveals that the polymer orientation as well as the droplet shape anisotropy influences the liquid crystal alignment within the droplets. A Monte-Carlo simulation based on the Lebwohl-Lasher model is used to explain the subtle influence of polymer orientation on liquid crystal alignment.Comment: 22 pages, pd

    Thrombospondin-1 Is a Major Activator of TGF-β1 In Vivo

    AbstractThe activity of TGF-β1 is regulated primarily extracellularly where the secreted latent form must be modified to expose the active molecule. Here we show that thrombospondin-1 is responsible for a significant proportion of the activation of TGF-β1 in vivo. Histological abnormalities in young TGF-β1 null and thrombospondin-1 null mice were strikingly similar in nine organ systems. Lung and pancreas pathologies similar to those observed in TGF-β1 null animals could be induced in wild-type pups by systemic treatment with a peptide that blocked the activation of TGF-β1 by thrombospondin-1. Although these organs produced little active TGF-β1 in thrombospondin null mice, when pups were treated with a peptide derived from thrombospondin-1 that could activate TGF-β1, active cytokine was detected in situ, and the lung and pancreatic abnormalities reverted toward wild type
