Costs and benefits of improving quality management in a multicriteria framework

This paper presents the 2003 results of empirical research carried into effect by using the questionnaire with interview method in one of the leading enterprises of the nutritive sector in Poland. The aim of this exploration was to define an influence of chosen factors, which results from initiating and certifying systems of quality management on the costs and benefits of improvement quality management. Besides, indication which from accepted variants that present adequate quality systems, best satisfy all analyzed factors, simultaneously supplies the biggest benefits to enterprise. To solve this problem, the Analytic Hierarchy Process (AHP) method with cost - benefit analysis was applied allowing for complex formulation of a given problem. The author’s intention was to show the utility of these methods in the solution of this problem and in similar multicriteria decision problems

Evaluation of the neuroprotective properties of group IIII metabotropic glutamate receptors ligands in the cellular model of Parkinson's disease

Ze względu na brak leku neuroprotekcyjnego w klinice, bardzo istotnym zagadnieniem w badaniach przedklinicznych pozostaje poszukiwanie nowych strategii farmakologicznych do leczenia chorób o podłożu neurodegeneracyjnym, w tym choroby Parkinsona (PD). Interesującym punktem uchwytu dla nowych leków neuroprotekcyjnych wydają się być metabotropowe receptory glutaminergiczne, a w szczególności grupa III tych receptorów (mGluR III). Dotychczasowe wyniki badań na komórkowych i zwierzęcych modelach PD sugerują korzystny wpływ pobudzenia mGluR III, który może wiązać się zarówno z hamowaniem objawów motorycznych, jak i spowalnianiem degeneracji neuronów istoty czarnej. Jak do tej pory, aktywacja mGluR III badana była głównie w kontekście hamowania nadmiernej transmisji glutaminianergicznej, która wiąże się z bezpośrednim mechanizmem pobudzenia tych receptorów na zakończeniach presynaptycznych i hamowanie uwalniania neuroprzekaźnika. Celem tej pracy było zbadanie potencjału neuroprotekcyjnego aktywatorów mGluR III w modelu in vitro PD z użyciem toksyny mitochondrialnej MPP(+), który to nie jest bezpośrednio związany z indukcją procesów ekscytotoksyczności. Po pierwsze, wykazano że wszystkie badane ligandy mGluR III, agonista ortosteryczny mGluR III (ACPT-I), agonista ortosteryczny mGluR8 ((S)-3,4-DCPG), jak i pozytywny allosteryczny modulator mGluR8 (AZ12216052) częściowo chroniły komórki niezróżnicowane (UN-SH-SY5Y) przed neurotoksycznością MPP(+), natomiast w komórkach neuronalnie zróżnicowanych (RA-SH-SY5Y) tylko specyficzna aktywacja receptora mGluR8 okazała się być skuteczna w ochronie komórek. Po drugie, badając możliwe mechanizmy odpowiedzialne za działanie neuroprotekcyjne wykluczyliśmy wpływ badanych aktywatorów mGluR III na aktywność kaspazy-3, enzymu kluczowego dla procesu apoptozy. Obserwowano natomiast ich ochronny efekt na indukowaną MPP(+) fragmentację DNA mierzoną metodą TUNEL. Co więcej, działanie neuroprotekcyjne badanych związków było hamowane podaniem inhibitora nekroptozy, nekrostatyny-1, a barwienie z użyciem jodku propidyny wykazało hamowanie przez testowane ligandy mGluR III zmian nekrotycznych wywołanych MPP(+). Otrzymane wyniki z modelu komórkowego wskazują na neuroprotekcyjny potencjał aktywacji mGluR III w modelu uszkodzeń komórek wywołanych MPP(+), a ochronne działanie badanych aktywatorów mGluR III może polegać na modulacji mechanizmów wewnątrzkomórkowej ścieżki nekrotyczną. Uzyskane rezultaty dają przesłanki do prowadzenia dalszych badań nad aktywatorami mGluR III w kierunku ich działania neuroprotekcyjnego w różnych chorobach neurodegeneracyjnych, z chorobą Parkinsona włącznie.The lack of the neuroprotective drug in clinic underlines the importance of searching for new pharmacological approaches for treatment of various neurodegenerative diseases, including Parkinson’s disease (PD) in the preclinical settings. The metabotropic glutamate receptors, especially group III (mGluR III) seems to be an interesting target for new putative neuroprotectants. The existing data from in vitro and in vivo studies on PD suggest that stimulation of mGluR III could be beneficial in inhibition of motor symptoms of PD as well could delay the degeneration of dopaminergic neurons in substantia nigra. The activation of mGluR III has been studied predominantly in relation to its inhibitory action on glutamatergic overactivity, which is the main mechanism of stimulation of presynaptic receptors leading to attenuation of neurotransmitter release. The present study was devoted to investigate the neuroprotective effects of mGluR III activators in in vitro model of PD engaging the mitochondrial inhibitor, MPP(+) which mechanism of toxic action is not directly connected with glutamatergic overactivation. We demonstrated that all tested activators of mGluR III: mGluRIII orthosteric agonist, ACPT-I; mGluR8 orthosteric agonist, (S)-3,4-DCPG and mGluR8 positive allosteric modulator, AZ12216052 partially inhibited undifferentiated SH-SY5Y cells against MPP(+) neurotoxicity. In neuronally differentiated (RA-SH-SY5Y) cells we found protection mediated only by mGluR8 activators. Next, we showed that in the mechanism of neuroprotection mediated by mGluR III is not involved regulation of caspase-3 activity, a crucial enzyme in apoptotic execution. However we found the decrease in MPP(+)-induced DNA fragmentation evoked by all tested mGluR III agents as was confirmed by TUNEL labeling. Moreover, we demonstrated the attenuation of necrotic cells after treatment with ACPT-I, (S)-3,4-DCPG and AZ12216052 and the protection mediated by these agents was blocked by inhibitor of necroptosis, necrostatin-1. The obtained data indicate that activation of mGluR III by orthosteric and allosteric ligands is neuroprotective against the MPP+-evoked cell damage and the mechanism probably engage the modulation of necrotic pathway. These results give a rationale for further studies on mGluR III as a putative target for new drugs to combat neurodegenerative diseases, including Parkinson’s disease

The impact of Sacz Lachs’ culture on tourism development (on the example of Limanowa, Podegrodzie and Nowy Sacz)

Praca została poświęcona wpływowi kultury Lachów Sądeckich na rozwój turystyki w Limanowej, Podegrodziu i Nowym Sączu. Obejmuje ona również charakterystykę geograficzną tych trzech miejscowości oraz opis kultury Lachów. Celem pracy jest ocena tego wpływu oraz ustalenie na podstawie badań ankietowych czy kultura ta utrwaliła się w świadomości turystów odwiedzających Beskid Wyspowy i Kotlinę Sądecką.The following study describes the impact of Sacz Lachs’ culture on tourism development in Limanowa, Podegrodzie and Nowy Sacz. It includes geographical characteristics of these three locations as well as a description of Lachs’ culture. The aim of this study is to assess that impact and to evaluate, on the basis of the questionnaires and surveys, whether this culture has its influence on the awareness of the tourists visiting Beskid Wyspowy and Sadecka Basin

Establishment of a Wisent (Bison bonasus) Germplasm Bank

The wisent, or European bison (Bison bonasus), belongs to the same family (Bovidae) as the American bison and domestic cattle. The wisent is the largest mammal in Europe, and is called the “Forest Emperor”. The wisent is listed as “Vulnerable” on the IUCN Red List, and is protected by international law. Achievements in reproductive biotechnology have opened new possibilities for the cryoconservation of the wisent germplasm. Therefore, this research aimed to improve a strategy for the protection and preservation of the European bison through the creation of a wisent germplasm bank, based on the following procedures: isolation and in vitro maturation (IVM) of oocytes, in vitro fertilization (IVF) of matured oocytes, in vitro embryo culture (IVC), and embryo cryopreservation. Wisent ovaries were isolated from females outside the reproductive season, and eliminated from breeding for reasons other than infertility. Cumulus–oocyte complexes (COCs) were isolated from follicles greater than 2 mm in diameter and matured for 24 h and 30 h. After IVM, COCs were fertilized in vitro with wisent sperm. The obtained wisent zygotes, based on oocytes matured for 24 h and 30 h, were cultured for 216 h. Embryos at the morula and early blastocyst stages were vitrified and then warmed and transferred to interspecies recipients (Bos taurus). USG and biochemical tests were used to monitor pregnancies. This study obtained embryos in the morula and early blastocyst stages only after oocytes were fertilized and matured for 30 h. On average, per oocyte donor, 12.33 ± 0.5 COCs were isolated, and only 9.33 ± 0.61 COCs were qualified for in vitro maturation (75.68%), while 9.16 ± 0.48 COCs were matured (84.32%). On average, per donor, 5.5 ± 0.34 embryos were cleaved (59.96%) after 48 h post-fertilization (hpf), and 3.33 ± 0.21 achieved the eight-cell stage (36.52%) after 96 hpf, while 1 ± 0.21 morula and early blastocyst stages (10.71%) were achieved after 216 hpf. A total of six embryos (one morula and five early blastocysts) were obtained and vitrified; after warming, five of them were interspecies transferred to cattle (Bos taurus). On day 41 after fertilization, 3 out of 5 pregnancies were detected based on USG, P4, and PAG tests. However, no pregnancy was observed on day 86 after fertilization, indicating embryo resorption. This study shows that obtaining wisent embryos in vitro, and subsequent cryopreservation to create a wisent embryo bank, can be applied and implemented for the wisent protection program
