    Neuropsychiatric systemic lupus erythematosus: where are we now?

    When dealing with neuropsychiatric Systemic Lupus Erythematosus (NPSLE) there are still many controversial topics. In 1999 the American College of Rheumatology gave classification criteria for 19 clinical syndromes. However major problems are still related to low specificity of some of them such as headache, cognitive impairment or mood disorders. Even though a frequency of CNS involvement from 14 to 75% has been described, depending on both the population studied and the methodology of assessment, a lower frequency ranging from 21 to 28 % derived by larger case series seems more realistic. The introduction of the concept of "borderline cases", proposed by Italian Study Group for NP-SLE, is based both on clinical and instrumental evaluation and could represent a useful tool when dealing with conditions which do not fulfil ACR classification. Also the relationship between SLE activity and NP involvement is a debated issue. Concerning pathogenesis, it seems reasonable to consider multifactorial mechanisms related to antibody-mediated damage, antiphospholipid pro-thrombotic effect, non-inflammatory vasculopathy and cytokines mediated cytotoxycity. However, direct and unequivocal evidence for the implication of any of the above-mentioned mechanisms is still lacking. Although a wide range of neuroimaging tools have been used to evaluate CNS involvement, no single technique has proven to be definitive and, when dealing with a patient with suspected NPSLE, it is important to combine different diagnostic techniques. Due to the lack of effective imaging along with limitation in knowledge of underlying pathogenetic mechanisms and paucity of histopathologic findings, therapeutic approach in NPSLE remains a difficult issue and is currently based on personal experience. Italian Study Group for NP-SLE proposes the creation of a national registry on NPSLE to validate ACR classification criteria. Furthermore, the possibility to collect large series and stratifying them for each of the included neuro-psychiatric syndromes seems a good strategy for planning multicentric controlled therapeutic trials in the near future

    Recent advances in Smart City infrastructures and the Internet of Things represent a significant opportunity to improve people's quality of life. Corresponding research often focuses on Cloud-centric network architectures where sensor devices transfer collected data to the Cloud for processing. However, the formidable traffic generated by countless IoT devices and the need for low-latency services raise the need to move away from centralized architectures and bring the computation closer to the data sources. To this end, this paper discusses SPF, a middleware solution that supports IoT application development, deployment, and management. SPF runs IoT services on capable devices located at the network edge and proposes an information-centric programming model that takes advantage of decentralized computation resources located in the proximity of application users and data sources. SPF also adopts Value-of-Information based methods to prioritize the transmission of essential information

    5'-deiodinase activity and circulating thyronines in lactating cows.

    To investigate the correlation between lactation and thyroid hormone metabolism, the authors studied concentrations of total and free thyroxine (T4 and fT4), triiodothyronine (T3 and fT3), and reverse triiodothyronine (rT3) in plasma and milk, as well as liver and mammary gland 5'-deiodinase (5'D) activity in dry, early, middle, and late lactating dairy cows. Cows in early lactation show lower plasma levels of T4 and rT3 than dry, middle, and late lactating animals, whereas T3 shows the lowest plasma levels in the dry period; free T4 and T3 show a similar pattern. In early lactation there is a clear decrease in liver 5'D associated with a notable increase in mammary 5'D. Concentrations of T4 and T3 in milk drop significantly in the first few days after delivery, whereas rT3 increases up to the fourth month. The findings suggest a relationship between the hypothyroid status of lactating cows and the rearrangement of organ-specific 5'-deiodinase activity related to the maintenance of the udder's function

    The Evolution of the Stellar Hosts of Radio Galaxies

    We present new near-infrared images of z>0.8 radio galaxies from the flux-limited 7C-III sample of radio sources for which we have recently obtained almost complete spectroscopic redshifts. The 7C objects have radio luminosities about 20 times fainter than 3C radio galaxies at a given redshift. The absolute magnitudes of the underlying host galaxies and their scale sizes are only weakly dependent on radio luminosity. Radio galaxy hosts at z~2 are significantly brighter than the hosts of radio-quiet quasars at similar redshifts and the model AGN hosts of Kauffmann & Haehnelt (2000). There is no evidence for strong evolution in scale size, which shows a large scatter at all redshifts. The hosts brighten significantly with redshift, consistent with the passive evolution of a stellar population that formed at z>~3. This scenario is consistent with studies of host galaxy morphology and submillimeter continuum emission, both of which show strong evolution at z>~2.5. The lack of a strong ``redshift cutoff'' in the radio luminosity function to z>4 suggests that the formation epoch of the radio galaxy host population lasts >~1Gyr from z>~5 to z~3. We suggest these facts are best explained by models in which the most massive galaxies and their associated AGN form early due to high baryon densities in the centres of their dark matter haloes.Comment: To appear in A

    Uncovering the physiological mechanisms underlying the roe deer (Capreolus capreolus) testicular cycle: Analyses of gelatinases and VEGF patterns and correlation with testes weight and testosterone

    The roe deer (Capreolus capreolus) represents a spontaneous model of testicular inactivation: During winter, bucks show a suspension of spermatogenesis that starts again in spring and peaks during the breeding season (July\u2013August). The underlying mechanisms to the regulation of the cyclic testicular changes are still not fully clear but seem to be imputable to the spermatogenic cell line since other testicular cell populations remain stable without apoptotic phenomena. The aim of the study was to investigate apoptosis, gelatinases (MMP2 and 9), their inhibiting factors (TIMP 1-2), and two isoforms of vascular endothelial growth factor (VEGF121 and 165) with its receptors (VEGFR1-2) in testes collected during pre-and post-rut periods, and to correlate them with testicular weight (TW) and testosterone (TEST). Testes from 18 adult sexually mature bucks were collected in Bologna Apennines (Italy). Samples were weighed and parenchyma collected. Radioimmunoassay, real-time PCR, and zymography were performed. The results showed a post-rut decrease in TW and TEST and an increase in proMMP2, also highlighting a correlation between the gelatinases and the testicular functionality. The VEGF pattern did not show modifications nor correlation with TW and TEST. Overall, gelatinases and their inhibitors, described herein for the first time in roe deer testes, seem to play an important role in the testicular cycle

    Spinal muscular atrophy with respiratory distress type 1: Clinical phenotypes, molecular pathogenesis and therapeutic insights

    Spinal muscular atrophy with respiratory distress type 1 (SMARD1) is a rare autosomal recessive neuromuscular disorder caused by mutations in the IGHMBP2 gene, which encodes immunoglobulin \u3bc-binding protein 2, leading to progressive spinal motor neuron degeneration. We review the data available in the literature about SMARD1. The vast majority of patients show an onset of typical symptoms in the first year of life. The main clinical features are distal muscular atrophy and diaphragmatic palsy, for which permanent supportive ventilation is required. No effective treatment is available yet, but novel therapeutic approaches, such as gene therapy, have shown encouraging results in preclinical settings and thus represent possible methods for treating SMARD1. Significant advancements in the understanding of both the SMARD1 clinical spectrum and its molecular mechanisms have allowed the rapid translation of preclinical therapeutic strategies to human patients to improve the poor prognosis of this devastating disease

    Testicular melatonin and its pathway in roe deer bucks (Capreolus capreolus) during pre-and post-rut periods: Correlation with testicular involution

    open8noRoe deer are seasonal breeders with a complete yearly testicular cycle. The peak in reproductive activity is recorded during summer, the rutting period, with the highest levels of androgens and testicular weight. Melatonin plays a pivotal role in seasonal breeders by stimulating the hypothalamus–pituitary–gonads axis and acting locally; in different species, its synthesis within testes has been reported. The aim of this study was to evaluate the physiological melatonin pattern within roe deer testes by comparing data obtained from animals sampled during pre-and post-rut periods. Melatonin was quantified in testicular parenchyma, along with the genetic expression of enzymes involved in its local synthesis (AANAT and ASMT) and function (UCP1). Melatonin receptors, MT1-2, were quantified both at protein and gene expression levels. Finally, to assess changes in reproductive hormonal profiles, testicular dehydroepiandrosterone (DHEA) was quantified and used for a correlation analysis. Melatonin and AANAT were detected in all samples, without significant differences between pre-and post-rut periods. Despite DHEA levels confirming testicular involution during the post-rut period, no correlations appeared between such involution and melatonin pathways.     Is kinesiophobia a predictor of early functional performance after total hip replacement? A prospective prognostic cohort study

    Background: Considerable attention has been paid to the role of kinesiophobia with respect to knee prosthesis but it has not yet been studied as a prognostic factor of short-term functional performance following total hip replacement. The main purpose of the present study is to examine the possible predictors of early functional performance of patients undergoing total primary hip arthroplasty, including demographics as age, sex and body mass index, preoperative functional ability, type of anaesthesia, level of haemoglobin, pain and level of kinesiophobia before surgery. Secondly, we want to describe the main characteristics of the population with the highest levels of kinesiophobia. Methods: A prospective, prognostic cohort study was carried out. Patients undergoing primary hip replacement were recruited consecutively. The main outcome is the early functional performance achieved by patients after surgery and measured using the Iowa Level of Assistance (ILOA) scale on the fifth postoperative day. Preoperative kinesiophobia was measured by the Tampa Scale and the preoperative functional ability by the Western Ontario and McMaster Osteoarthritis Index (WOMAC). The multivariate analysis was performed by the General Linear Model. The analysis of the population with high levels of kinesiophobia was conducted by identifying a cut-off of 40 compared to the Tampa Scale. Results: Statistical analysis was performed on 269 patients. The average ILOA score recorded was 19.5 (DS 8.3). The levels of kinesiophobia, showed an average score of 35.1 (7.8) and it was not associated with early functional performance. The independent predictive factors include age, sex and body mass index. Kinesiophobia high levels were recorded in 30% of the population and this population had a higher level of pre-operative WOMAC score. Conclusions: Early functional performance after hip replacement surgery was not correlated with the level of kinesiophobia. Three significant factors that describe a population most at risk of not achieving optimal functional performance are increased age, being female and increase in body mass index. In the preoperative phase, high levels of kinesiophobia were associated with more impaired preoperative functional ability. Trial registration: Current Controlled Trials NCT02786121, May 2016.     Massive Ellipticals at High Redshift: NICMOS Imaging of Z~1 Radio Galaxies

    We present deep, continuum images of eleven high-redshift (0.811 < z < 1.875) 3CR radio galaxies observed with NICMOS. Our images probe the rest-frame optical light where stars are expected to dominate the galaxy luminosity. The rest-frame UV light of eight of these galaxies demonstrates the well-known ``alignment effect''. Most of the radio galaxies have rounder, more symmetric morphologies at rest-frame optical wavelengths. Here we show the most direct evidence that in most cases the stellar hosts are normal elliptical galaxies with de Vaucouleurs law light profiles. For a few galaxies very faint traces of the UV-bright aligned component are also visible in the infrared images. We derive both the effective radius and surface-brightness for nine of eleven sample galaxies by fitting surface-brightness models to them. We find their sizes are similar to those of local FRII radio source hosts and are in general larger than other local galaxies. The derived host galaxy luminosities are very high and lie at the bright end of luminosity functions constructed at similar redshifts. The galaxies in our sample are also brighter than the rest-frame size--surface-brightness locus defined by the low-redshift sources. Passive evolution roughly aligns the z ~ 1 galaxies with the low-redshift samples. The optical host is sometimes centered on a local minimum in the rest-frame UV emission, suggesting the presence of substantial dust obscuration. We also see good evidence of nuclear point sources in three galaxies. Overall, our results are consistent with the hypothesis that these galaxies have already formed the bulk of their stars at redshifts greater than z >~ 2, and that the AGN phenomenon takes place within otherwise normal, perhaps passively evolving, galaxies.     Sardinia Radio Telescope wide-band spectral-polarimetric observations of the galaxy cluster 3C 129

    We present new observations of the galaxy cluster 3C 129 obtained with the Sardinia Radio Telescope in the frequency range 6000-7200 MHz, with the aim to image the large-angular-scale emission at high-frequency of the radio sources located in this cluster of galaxies. The data were acquired using the recently-commissioned ROACH2-based backend to produce full-Stokes image cubes of an area of 1 deg x 1 deg centered on the radio source 3C 129. We modeled and deconvolved the telescope beam pattern from the data. We also measured the instrumental polarization beam patterns to correct the polarization images for off-axis instrumental polarization. Total intensity images at an angular resolution of 2.9 arcmin were obtained for the tailed radio galaxy 3C 129 and for 13 more sources in the field, including 3C 129.1 at the galaxy cluster center. These data were used, in combination with literature data at lower frequencies, to derive the variation of the synchrotron spectrum of 3C 129 along the tail of the radio source. If the magnetic field is at the equipartition value, we showed that the lifetimes of radiating electrons result in a radiative age for 3C 129 of t_syn = 267 +/- 26 Myrs. Assuming a linear projected length of 488 kpc for the tail, we deduced that 3C 129 is moving supersonically with a Mach number of M=v_gal/c_s=1.47. Linearly polarized emission was clearly detected for both 3C 129 and 3C 129.1. The linear polarization measured for 3C 129 reaches levels as high as 70% in the faintest region of the source where the magnetic field is aligned with the direction of the tail.Comment: 19 pages, 17 figures, accepted for publication in MNRA
