    Meteorological satellite products support for project COHMEX

    The first year effort focussed on real-time support and satellite data collection during the field phase of COHMEX. Work efforts following the field phase of COHMEX concentrated on post-processing of the real-time data sets, and generation of enhanced, research-quality satellite data sets for selected COHMEX core days. These satellite-derived data sets will augment the special COHMEX conventional data base with high horizontal and temporal resolution information. The data sets will be examined for their usefulness in delineating important elements in the meteorological environment leading to convective activity. In addition, a limited research effort was conducted using the Cooperative Institute for Meteorological Satellite Studies (CIMSS) 4-d data assimilation system in conjunction with evaluating VISSR Atmospheric Sounder (VAS) and His-resolution Interferometer Sounder (HIS) data. The need to address the characteristics of the data types, and the problems they introduce into 4-d assimilation procedures is evident. The HIS instrument was flown aboard an ER-2 aircraft on several occasions during COHMEX. One of the flights was chosen for further study. Processed VAS soundings and COHMEX radiosonde data were also collected for this day. The case study included an evaluation of the HIS and VAS data and an impact study of the data on the assimilation system analysis

    Dynein Achieves Processive Motion Using both Stochastic and Coordinated Stepping

    Processivity, the ability of single molecules to move continuously along a track, is a fundamental requirement of cargo-transporting molecular motors. Here, we investigate how cytoplasmic dynein, a homodimeric, microtubule-based motor, achieves processive motion. To do this, we developed a versatile method for assembling Saccharomyces cerevisiae dynein heterodimers, using complementary DNA oligonucleotides covalently linked to dynein monomers labeled with different organic fluorophores. Using two-color, single-molecule microscopy and high-precision, two-dimensional tracking, we find that dynein has a highly variable stepping pattern that is distinct from all other processive cytoskeletal motors, which use \u27hand-over-hand\u27 mechanisms. Uniquely, dynein stepping is stochastic when its two motor domains are close together. However, coordination emerges as the distance between motor domains increases, implying that a tension-based mechanism governs these steps. This plasticity may allow tuning of dynein for its diverse cellular functions

    In Vivo Capture and Label-Free Detection of Early Metastatic Cells

    Breast cancer is a leading cause of death for women, with mortality resulting from metastasis. Metastases are often detected once tumour cells affect the function of solid organs, with a high disease burden limiting effective treatment. Here we report a method for the early detection of metastasis using an implanted scaffold to recruit and capture metastatic cells in vivo, which achieves high cell densities and reduces the tumour burden within solid organs 10-fold. Recruitment is associated with infiltration of immune cells, which include Gr1hiCD11b+cells. We identify metastatic cells in the scaffold through a label-free detection system using inverse spectroscopic optical coherence tomography, which identifies changes to nanoscale tissue architecture associated with the presence of tumour cells. For patients at risk of recurrence, scaffold implantation following completion of primary therapy has the potential to identify metastatic disease at the earliest stage, enabling initiation of therapy while the disease burden is low

    Supplier-induced demand for psychiatric admissions in Northern New England

    The development of hospital service areas (HSAs) using small area analysis has been useful in examining variation in medical and surgical care; however, the techniques of small area analysis are underdeveloped in understanding psychiatric admission rates. We sought to develop these techniques in order to understand the relationship between psychiatric bed supply and admission rates in Northern New England. Our primary hypotheses were that there would be substantial variation in psychiatric admission across geographic settings and that bed availability would be positively correlated with admission rates, reflecting a supplier-induced demand phenomenon. Our secondary hypothesis was that the construction of psychiatric HSAs (PHSAs) would yield more meaningful results than the use of existing general medical hospital service areas

    Dark Matter from Minimal Flavor Violation

    We consider theories of flavored dark matter, in which the dark matter particle is part of a multiplet transforming nontrivially under the flavor group of the Standard Model in a manner consistent with the principle of Minimal Flavor Violation (MFV). MFV automatically leads to the stability of the lightest state for a large number of flavor multiplets. If neutral, this particle is an excellent dark matter candidate. Furthermore, MFV implies specific patterns of mass splittings among the flavors of dark matter and governs the structure of the couplings between dark matter and ordinary particles, leading to a rich and predictive cosmology and phenomenology.
