Scanning from heating: 3D shape estimation of transparent objects from local surface heating
Today, with quality becoming increasingly important, each product requires three-dimensional in-line quality control. On the other hand, the 3D reconstruction of transparent objects is a very difficult problem in computer vision due to transparency and specularity of the surface. This paper proposes a new method, called Scanning From Heating (SFH), to determine the surface shape of transparent objects using laser surface heating and thermal imaging. Furthermore, the application to transparent glass is discussed and results on different surface shapes are presented
Normal Levels of Sox9 Expression in the Developing Mouse Testis Depend on the TES/TESCO Enhancer, but This Does Not Act Alone
During mouse sex determination, transient expression of the Y-linked gene Sry up-regulates its direct target gene Sox9, via a 3.2 kb testis specific enhancer of Sox9 (TES), which includes a core 1.4 kb element, TESCO. SOX9 activity leads to differentiation of Sertoli cells, rather than granulosa cells from the bipotential supporting cell precursor lineage. Here, we present functional analysis of TES/TESCO, using CRISPR/Cas9 genome editing in mice. Deletion of TESCO or TES reduced Sox9 expression levels in XY fetal gonads to 60 or 45% respectively relative to wild type gonads, and reduced expression of the SOX9 target Amh. Although human patients heterozygous for null mutations in SOX9, which are assumed to have 50% of normal expression, often show XY female sex reversal, mice deleted for one copy of Sox9 do not. Consistent with this, we did not observe sex reversal in either TESCO-/- or TES-/- XY embryos or adult mice. However, embryos carrying both a conditional Sox9 null allele and the TES deletion developed ovotestes. Quantitative analysis of these revealed levels of 23% expression of Sox9 compared to wild type, and a significant increase in the expression of the granulosa cell marker Foxl2. This indicates that the threshold in mice where sex reversal begins to be seen is about half that of the ~50% levels predicted in humans. Our results demonstrate that TES/TESCO is a crucial enhancer regulating Sox9 expression in the gonad, but point to the existence of additional enhancers that act redundantly

Structure of amyloid-β (20-34) with Alzheimer's-associated isomerization at Asp23 reveals a distinct protofilament interface.
Amyloid-β (Aβ) harbors numerous posttranslational modifications (PTMs) that may affect Alzheimer's disease (AD) pathogenesis. Here we present the 1.1 Å resolution MicroED structure of an Aβ 20-34 fibril with and without the disease-associated PTM, L-isoaspartate, at position 23 (L-isoAsp23). Both wild-type and L-isoAsp23 protofilaments adopt β-helix-like folds with tightly packed cores, resembling the cores of full-length fibrillar Aβ structures, and both self-associate through two distinct interfaces. One of these is a unique Aβ interface strengthened by the isoaspartyl modification. Powder diffraction patterns suggest a similar structure may be adopted by protofilaments of an analogous segment containing the heritable Iowa mutation, Asp23Asn. Consistent with its early onset phenotype in patients, Asp23Asn accelerates aggregation of Aβ 20-34, as does the L-isoAsp23 modification. These structures suggest that the enhanced amyloidogenicity of the modified Aβ segments may also reduce the concentration required to achieve nucleation and therefore help spur the pathogenesis of AD
That was the last straw, we need more: Are Translation Systems Sensitive to Disambiguating Context?
The translation of ambiguous text presents a challenge for translation
systems, as it requires using the surrounding context to disambiguate the
intended meaning as much as possible. While prior work has studied ambiguities
that result from different grammatical features of the source and target
language, we study semantic ambiguities that exist in the source (English in
this work) itself. In particular, we focus on idioms that are open to both
literal and figurative interpretations (e.g., goose egg), and collect TIDE, a
dataset of 512 pairs of English sentences containing idioms with disambiguating
context such that one is literal (it laid a goose egg) and another is
figurative (they scored a goose egg, as in a score of zero). In experiments, we
compare MT-specific models and language models for (i) their preference when
given an ambiguous subsentence, (ii) their sensitivity to disambiguating
context, and (iii) the performance disparity between figurative and literal
source sentences. We find that current MT models consistently translate English
idioms literally, even when the context suggests a figurative interpretation.
On the other hand, LMs are far more context-aware, although there remain
disparities across target languages. Our findings underline the potential of

Structure-based inhibitors of amyloid beta core suggest a common interface with tau.
Alzheimer's disease (AD) pathology is characterized by plaques of amyloid beta (Aβ) and neurofibrillary tangles of tau. Aβ aggregation is thought to occur at early stages of the disease, and ultimately gives way to the formation of tau tangles which track with cognitive decline in humans. Here, we report the crystal structure of an Aβ core segment determined by MicroED and in it, note characteristics of both fibrillar and oligomeric structure. Using this structure, we designed peptide-based inhibitors that reduce Aβ aggregation and toxicity of already-aggregated species. Unexpectedly, we also found that these inhibitors reduce the efficiency of Aβ-mediated tau aggregation, and moreover reduce aggregation and self-seeding of tau fibrils. The ability of these inhibitors to interfere with both Aβ and tau seeds suggests these fibrils share a common epitope, and supports the hypothesis that cross-seeding is one mechanism by which amyloid is linked to tau aggregation and could promote cognitive decline
Sublinear-Time Algorithms for Monomer-Dimer Systems on Bounded Degree Graphs
For a graph , let be the partition function of the

and develop a sublinear-time algorithm for estimating at an
arbitrary value within additive error with high
probability. The query complexity of our algorithm does not depend on the size
of and is polynomial in , and we also provide a lower bound
quadratic in for this problem. This is the first analysis of a
sublinear-time approximation algorithm for a # P-complete problem. Our
approach is based on the correlation decay of the Gibbs distribution associated
with . We show that our algorithm approximates the probability
for a vertex to be covered by a matching, sampled according to this Gibbs
distribution, in a near-optimal sublinear time. We extend our results to
approximate the average size and the entropy of such a matching within an
additive error with high probability, where again the query complexity is
polynomial in and the lower bound is quadratic in .
Our algorithms are simple to implement and of practical use when dealing with
massive datasets. Our results extend to other systems where the correlation
decay is known to hold as for the independent set problem up to the critical
activity
