    Performance of sweet cherry cultivars grafted on Colt rootstock

    In this paper growing characteristics and fruit bearing parameters of ‘Lapins’, ‘Kordia’, ‘Sweetheart’ and ‘Regina’ sweet cherry cultivars grafted on vigorous Colt rootstock were evaluated at the University of Debrecen, Pallag Experimental Station. Based on our data five years old trees can be described with homogenous strong growing, but very week yielding (2.1-3.1 kg/tree), as while fruit size varies between 26.2 and 27.2 mm. Producing the examined cultivars on Colt rootstock with high plant density requires higher attention and more interventions (root pruning, sawing the trunk) during the technology

    Comparison of Genotoxic Damage in Monolayer Cell Cultures and Three-Dimensional Tissue-Like Cell Assemblies

    Assessing the biological risks associated with exposure to the high-energy charged particles encountered in space is essential for the success of long-term space exploration. Although prokaryotic and eukaryotic cell models developed in our laboratory and others have advanced our understanding of many aspects of genotoxicity, in vitro models are needed to assess the risk to humans from space radiation insults. Such models must be representative of the cellular interactions present in tissues and capable of quantifying I genotoxic damage. Toward this overall goal, the objectives of this study were to examine the effect of the localized microenvironment of cells, cultured as either 2-dimensional (2D) monolayers or 3-dimensional (3D) aggregates, on the rate and type of genotoxic damage resulting from exposure to iron charged particles, a significant portion of space radiation. We used rodent transgenic cell lines containing 50-70 copies of a LacI transgene to provide the enhanced sensitivity required to quantify mutational frequency and type in the 1,100-bp LacI target as well as assessment of DNA,damage to the entire 45-kbp construct. Cultured cells were exposed to high-ener~ir on charged particles at Brookhaven National Laboratory s Alternating Gradient Synchrotron facility for a total dose of 0, 0.1, 0.25,0.5, 1.0, or 2.0 Gy and allowed to recover for 0, 1, or 7 days, after which mutational type and frequency were evaluated. The mutational frequency was found to be higher in 3D samples than in 2D samples at all radiation doses. Mutational frequency also was higher at 7 days after irradiation than immediately after exposure. DNA sequencing of the mutant targets revealed that deletional mutations contributed an increasingly high percentage (up to 27%) of all mutations in cells as the dose was increased from 0.5 to 2 Gy. Several mutants also showed large and complex deletions in multiple locations within the Lac1 target. However, no differences in mutational type were found between the 2D and the 3D samples. These 3D tissue-like model systems can reduce the uncertainty involved in extrapolating risk between in vitro cellular and in vivo models

The bioreactors of this invention are ideally suited for studying short, moderate and long term studies of cell cultures and the response of cell cultures to one or more stressors such as pharmaceuticals, hypoxia, pathogens, or any other stressor.     Miniature Bioreactor System for Long-Term Cell Culture

    A prototype miniature bioreactor system is designed to serve as a laboratory benchtop cell-culturing system that minimizes the need for relatively expensive equipment and reagents and can be operated under computer control, thereby reducing the time and effort required of human investigators and reducing uncertainty in results. The system includes a bioreactor, a fluid-handling subsystem, a chamber wherein the bioreactor is maintained in a controlled atmosphere at a controlled temperature, and associated control subsystems. The system can be used to culture both anchorage-dependent and suspension cells, which can be either prokaryotic or eukaryotic. Cells can be cultured for extended periods of time in this system, and samples of cells can be extracted and analyzed at specified intervals. By integrating this system with one or more microanalytical instrument(s), one can construct a complete automated analytical system that can be tailored to perform one or more of a large variety of assays

    Humor appreciation of captionless cartoons in obsessive-compulsive disorder

    Background: It seems that the core neural regions and cognitive processes implicated in obsessive-compulsive disorder (OCD) pathophysiology may overlap with those involved in humor appreciation. However, to date, there have been no studies that have explored humor appreciation in OCD. The purpose of the present work was to investigate humor appreciation in a group of patients with OCD.Methods: We examined 25 patients with OCD and 25 healthy controls, matched by age, education, and gender. We administered Penn's Humor Appreciation Test (PHAT), a computerized test comprising captionless cartoons by Mordillo. Each set of stimuli consisted of two almost identical drawings, one of which was funny due to the alteration of a detail in the cartoon, whereas the other was not funny. Severity of psychopathology was evaluated with the Yale-Brown Obsessive Compulsive Scale (Y-BOCS).Results: No significant effect for group, gender or group × gender interaction was found on the PHAT scores. In OCD patients, humor appreciation was not significantly associated with age of onset, duration of illness, and obsessions, but correlated significantly with compulsions.Conclusions: Humor appreciation, based on captionless cartoons in OCD, does not seem to be deficient compared to healthy subjects but may be related to illness characteristics. © 2011 Bozikas et al; licensee BioMed Central Ltd

    Estimating the Effects of Weather, Dry Matter Intake, and Body Weight on Daily Water Intake in Weaned Calves

    Objective The purpose of this study was to study the effect of weather, dry matter intake, and body weight on the water requirements of weaned calves and estimating the requirements in a model.Study Description Weaned steers (n=48) were selected to study the effects of the weather, body weight, and dry matter intake on water intake in the winter (n=24) and summer (n=24) months. Calves were provided with ad libitum access to feed and water at the SDSU Cow-Calf Education and Research Facility (CCERF) and measured utilizing an automated feed and water system (Insentec, The Hague, Netherlands). Temperature, humidity, precipitation, wind speed, solar radiation, and air pressure were recorded at a Mesonet automated weather station in Brookings, SD (located 2.4 miles from the SDSU CCERF). Effects of climate data, body weight, and daily dry matter intake on daily water intake will be analyzed utilizing a mixed-effects model

    Sweet cherry is currently considered as a fruit with high interests. The amount of the produced yield is well saleable in the world and also in Hungary year by year, moreover often there is a shortage with the high quality fruits. Researches with the sweet cherry production focus on the intensity growing all over the world, namely the realization of producing with small trees ensuring high quality and quantity. In our examinations ’Lapins’ sweet cherry cultivar was evaluated grafted on rootstocks with different vigor (Gisela 5, Gisela 6, Colt) at the University of Debrecen, Pallag Experimental Station. According to our results trees grafted on Colt rootstock can be described with very low yields due to the excessive high vigor. Trees with Gisela 5 and Gisela 6 rootstocks showed excessive high productivity, as size of the fruits did not reach the required values
