    Implementing the teaching personal and social responsibility model within preschool education: strengths, challenges and strategies

    The teaching personal and social responsibility (TPSR) model has been considered a valuable tool for children to reach their full potential in life. However, little research has been conducted with the TPSR model in preschool contexts. The purpose of the present study was to understand the experiences of a program leader while implementing a TPSR-based program within a preschool setting. The participants were 24 children with 5 years of age attending a school located in the north of Portugal, and the program leader. Data sources included reflexive journaling, field notes and participant observations. Findings suggest the TPSR model could be adapted and implemented with preschool children. However, there is a need to consider specific adaptations to develop TPSR-based interventions and facilitate responsibility outcomes.     Ambivalence in emotion-focused therapy for depression: the maintenance of problematically dominant self-narratives

    Objective: Ambivalence can be understood as a cyclical movement between an emerging narrative novelty—an Innovative Moment (IM)—and a return to a problematically dominant self-narrative. The return implies that the IM, with its potential for change is devalued right after its emergence. Our goal is to test the hypothesis that the probability of the client expressing such form of ambivalence decreases across treatment in good-outcome cases but not in poor-outcome cases. Method: Return-to-the-Problem Markers (RPMs) signaling moments of devaluation of IMs were coded in passages containing IMs in six clients with major depression treated with emotion-focused therapy: three good-outcome cases and three poor-outcome cases. Results: The percentage of IMs with RPMs decreased across therapy in good-outcome cases, whereas it remained unchanged and high in the poor-outcome cases. Conclusions: These results were consistent with the theoretical suggestion that therapeutic failure may be associated with this form of ambivalence.Obiettivo: L'ambivalenza può essere considerata come un movimento ciclico tra un nuovo elemento narrativo emergente - Innovative Moment (IM) - e il ritorno ad una auto-narrazione problematica dominante. Il ritorno implica che il IM perde il proprio potenziale di favorire un cambiamento nel momento in cui emerge. Il nostro obiettivo è quello di verificare l'ipotesi che la probabilità che il paziente possa esprimere tale forma di ambivalenza diminuisce nel corso di un trattamento che ha un buon esito, ma non nei casi con esito negativo. Metodo: in sei pazienti affetti da depressione maggiore trattati con terapia focalizzata sulle emozioni, sono stati codificati i passaggi contenenti IM utilizzando il Return-to-the-Problem Markers (RPMs) che segnala i passaggi in cui IM vengono svalutati: tre casi con buon esito e tre con esito negativo. Risultati: La percentuale di IM con gli RPM diminuiva nel corso delle terapie dei casi con buon esito, mentre è rimasto invariato ed elevato nei casi con scarso esito. Conclusioni: Questi risultati sono coerenti con il presupposto teorico che il fallimento terapeutico possa essere associato a questa forma di ambivalenza.Objetivo: A ambivalência pode ser entendida como um movimento cíclico entre uma novidade narrativa emergente – um Momento de Inovação (MI) – e um retorno à narrativa problemática dominante. O retorno implica que o MI, com o seu potencial para a mudança, seja desvalorizado logo após a sua emergência. O nosso objetivo é testar a hipótese de que a probabilidade do cliente expressar tal forma de ambivalência diminui ao longo do tratamento em casos de sucesso mas não em casos de insucesso terapêutico. Método: Marcadores de Retorno ao Problema (MRP) que assinalam momentos de desvalorização dos MIs foram codificados em passagens contendo MIs em seis casos de clientes com depressão major tratados com terapia focada nas emoções: três casos de sucesso e três casos de insucesso. Conclusões: Estes resultados foram consistentes com as sugestões teóricas de que o insucesso terapêutico poderá estar associado a esta forma de ambivalência.Ziel: Ambivalenz kann als zyklische Bewegung zwischen einer emergenten narrativen Neuheit – einem Innovative Moment (IM) – und einer Rückkehr zu einer problematischen dominanten Selbst-Erzählung verstanden werden. Diese Rückkehr impliziert, dass der IM mit seinem Potential für Veränderung direkt nach seinem Auftauchen abgewertet wird. Unser Ziel ist die Hypothese zu testen, dass die Wahrscheinlichkeit, dass der Klient eine solche Form der Ambivalenz zeigt, bei Fällen mit gutem Therapieergebnis über die Behandlung abnimmt, aber nicht in Fällen mit schlechtem Ergebnis. Methode: Rückkehr-zu-dem-Problem Marker (RPMs), die Momente signalisieren, in denen IMs abgewertet werden, wurden in Passagen, die IMs beinhalten, von sechs Klienten mit Major Depression, die mit emotionsfokussierter Therapie behandelt wurden, kodiert: 3 Fälle mit gutem und 3 Fälle mit schlechtem Therapieergebnis. Ergebnisse: Der Prozentsatz von IMs mit RPMs verringerte sich bei Fällen mit gutem Ergebnis über die Therapie hinweg, wohingegen er in den Fällen mit schlechtem Ergebnis unverändert und hoch blieb. Schlussfolgerung: Die Ergebnisse waren mit dem theoretischen Vorschlag konsistent, dass therapeutischer Misserfolg mit dieser Form der Ambivalenz assoziiert ist.目標:矛盾可以視為是種擺盪在新產生的新穎敘說—創意時刻(IM)—和回復到一種有問題的支配性自我敘說之間的循環動作。回復意味著潛藏改變契機的IM,其價值在出現後即被摒棄。我們的目標是檢視以下假設的可能性:「在具良好成效的案例中,案主在治療歷程中表達這種形式的矛盾會降低;而在不良成效的案例中則否」。方法:在六位具有嚴重憂鬱症患者的IM段落中,找到顯示IM價值降低時刻的故態復萌標記(RPMs)並加以編碼。這六位患者均接受情緒焦點治療,其中三位為具良好成效案例,另外三位為不良成效案例。結果:在具良好成效案例中,IM當中具有RPM的比率降低;而在不良成效的案例中,RPM的比率維持不變或更高。結論:此結果與理論假設一致,即假設治療的失敗可能與這種形式的矛盾有關

    Narrative change in emotion-focused psychotherapy: a study on the evolution of reflection and protest innovative moments

    Innovative moments (IMs) are exceptions to a client’s problematic self-narrative in the therapeutic dialogue. The innovative moments coding system is a tool which tracks five different types of IMs*action, reflection, protest, reconceptualization and performing change. An in-depth qualitative analysis of six therapeutic cases of emotion-focused therapy (EFT) investigated the role of two of the most common IMs*reflection and protest*in both good and poor outcome cases. Through this analysis two subtypes (I and II) of reflection and protest IMs were identified, revealing different evolution patterns. Subtype II of both reflection and protest IMs is significantly higher in the good outcome group, while subtype I of both IMs types does not present statistically significant differences between groups. The evolution from subtype I to subtype II across the therapeutic process seems to reflect a relevant developmental progression in the change process

    Innovative moments in grief therapy: reconstructing meaning following perinatal death

    This article presents an intensive analysis of a good outcome case of constructivist grief therapy with a bereaved mother, using the Innovative Moments Coding System (IMCS). Inspired by M. White and D. Epston’s narrative therapy, the IMCS conceptualizes therapeutic change as resulting from the elaboration and expansion of unique outcomes (or as we prefer, innovative moments), referring to experiences not predicted by the problematic or dominant self-narrative. The IMCS identifies and tracks the occurrence of 5 different types of innovative moments: action, reflection, protest, re-conceptualization, and performing change. Results documented the process of meaning reconstruction over the 6 sessions of treatment, and demonstrated the feasibility and reliability of analyzing narrative change in this form of grief therapy, opening it to comparison with other approaches

    Probiotic potential of fructo-oligosaccharides produced by Aspergillus ibericus

    The gastrointestinal tract harbours a diverse and dynamic microbial community that directly impacts human health. Prebiotics, such as fructo-oligosaccharides (FOS), play a crucial role in the modulation of colonic microbiota, reducing pathophysiological disorders and associated chronic diseases. The prebiotic potential of FOS produced by a newly isolated strain Aspergillus ibericus was studied. FOS fermentability by the probiotic Lactobacillus rhamnosus was evaluated. L. rhamnosus was grown in de Man-Rogosa-Sharpe (MRS) broth, with different carbon sources: glucose (positive control), no sugar (negative control), microbial-derived FOS from A. ibericus and Raftilose®P95, a non-microbial commercial FOS sample (from Beneo-Orafti, Belgium). A final concentration of 2 % (w/v) in sugar was used. Fermentation was carried out in a 96-well microplate and a shake flask, for 24 h, at 37 ºC, with an agitation of 120 rpm. Biomass growth was analysed by optical density at 620 nm. The consumption of sugars and the production of short chain fatty acids (SCFA) and lactate was quantified by HPLC. Maximum cell growth was reached at approximately 12 h, for all carbon sources. The highest growth was achieved for glucose samples, followed by the microbial-derived FOS, then Raflitose and finally the negative control. Although the microbial-derived FOS promoted great cellular growth, only kestose (GF2), together with residual amounts of glucose and sucrose presented in the sample, were consumed. This may explain the two different slopes exhibited during the exponential phase growth. Most likely hypothesis is that probiotic bacteria was cleaving GF2 in the first hours of fermentation, using only the smallest sugars present for growing. And Nystose (GF3) and fructofuranosylnystose (GF4) were not consumed, even when prolonging the fermentation up to 48 h. SCFA identified were valerate and propionate, as well as succinate, formate, acetate, iso-butyrate and nbutirate, although in lower amount. Higher amount of SCFA and lactate were determined while growing in the microbial-derived FOS, as compared to the commercial sample. Overall, lactate was the main metabolite produced during the fermentations. In conclusion, the prebiotic potential of microbial-derived FOS synthesized by A. ibericus was demonstrated, providing promising indication of its usability as food ingredient with strong prebiotic features.info:eu-repo/semantics/publishedVersio

As Marine Biotechnology is gaining momentum as one of the main pillars of the ‘Brue Growth’ revolution, the ability to screen for novel compounds of interest in species with little or no genomic resources is paramount. With this respect, proteins, which are easily metabolised, can be synthetised using convenient DNA recombinant methods and can easily be modified to better meet the needs of human society, making them prized targets. Evidently, proteins that hold natural bioactivity and specificity such as toxins and other venom components, have long captured the focus of biotechnologists, leading to the merger between environmental omics and toxinology termed as ‘venomics’. Indeed, bioactive proteins such as conopeptides, conotoxins, turripeptides and others are long deemed important subjects of research. Even though current mainstream paradigms set the focus on secondary metabolites from marine organisms, transcriptomics and proteomics approaches and their combination are rising strategies for screening for thousands of proteins and peptides in non-conventional biological models, emphasising, but not limited to, marine invertebrate animals due to their abundance, biodiversity and uncanny biochemical strategies to cope with selective pressure in literally every known marine habitat. Untargeted approaches, such as RNA-Seq - based transcriptomics and tandem mass spectrometry - based proteomics, can circumvent limitations related with absent or reduced genomic annotation. The present review will outline the main contributions of ‘omics’ and computational approaches for bioprospecting for proteinaceous marine bioactives. Despite the relatively low number of ‘omics’ studies with the main purpose of discover novel compounds, there is already important literature showcasing pipelines and approaches for revolutionising the exploration of the ocean.publishersversionpublishe

    Grape and Wine Metabolites: Biotechnological Approaches to Improve Wine Quality

    Grape metabolites can be affected by many extrinsic and intrinsic factors, such as grape variety, ripening stage, growing regions, vineyard management practices, and edaphoclimatic conditions. However, there is still much about the in vivo formation of grape metabolites that need to be investigated. The winemaking process also can create distinct wines. Nowadays, wine fermentations are driven mostly by single-strain inoculations, allowing greater control of fermentation. Pure cultures of selected yeast strains, mostly Saccharomyces cerevisiae, are added to grape must, leading to more predictable outcomes and decreasing the risk of spoilage. Besides yeasts, lactic acid bacteria also play an important role, in the final wine quality. Thus, this chapter attempts to present an overview of grape berry physiology and metabolome to provide a deep understanding of the primary and secondary metabolites accumulated in the grape berries and their potential impact in wine quality. In addition, biotechnological approaches for wine quality practiced during wine alcoholic and malolactic fermentation will also be discussed

    Dynamics of sense-making and development of the narrative in the clinical exchange

    The present work is aimed at exploring the relationship between the dynamics of sense-making carried out by the clinical exchange and the content of the patient’s narrative. To this end the relationship between the formal and functional mapping of a psychotherapy carried out by the Discourse Flow Analysis (DFA) and the analysis of the patient’s narrative provided by the Innovative Moment Coding System (IMCS) have been compared. The comparison concerns a 15-session good outcome Emotion- Focused Therapy (Lisa's case). Findings highlight the association between the formal and functional characteristics of the clinical dialogue and the content of the narrative. More in particular, an association between the U-shape trajectory of the super-ordered meaning depicted by DFA and the evolution of the innovative content of the narrative enucleated by the IMCS were found

    Spectral Optical Properties of Rabbit Brain Cortex between 200 and 1000 nm

    The knowledge of the optical properties of biological tissues in a wide spectral range is highly important for the development of noninvasive diagnostic or treatment procedures. The absorption coefficient is one of those properties, from which various information about tissue components can be retrieved. Using transmittance and reflectance spectral measurements acquired from ex vivo rabbit brain cortex samples allowed to calculate its optical properties in the ultraviolet to the near infrared spectral range. Melanin and lipofuscin, the two pigments that are related to the aging of tissues and cells were identified in the cortex absorption. By subtracting the absorption of these pigments from the absorption of the brain cortex, it was possible to evaluate the true ratios for the DNA/RNA and hemoglobin bands in the cortex—12.33-fold (at 260 nm), 12.02-fold (at 411 nm) and 4.47-fold (at 555 nm). Since melanin and lipofuscin accumulation increases with the aging of the brain tissues and are related to the degeneration of neurons and their death, further studies should be performed to evaluate the evolution of pigment accumulation in the brain, so that new optical methods can be developed to aid in the diagnosis and monitoring of brain diseases.The work of L.M.O. was supported by the Portuguese Science Foundation, Grant No. FCT-UIDB/04730/2020.
