    Multilateralism and Democracy: A Dissent Regarding Keohane, Macedo, and Moravcsik

    In an article printed last year in International Organization, Keohane, Macedo, and Moravcsik argued that multilateral organizations (MLO) could actually be good for democracy. We argue that KMM discount the prospect that MLO influence can be detrimental to democracies not because MLOs are “distant, elitist, and technocratic” but precisely because MLOs are highly political. International organizations have much to offer in improving the welfare of citizens and facilitating coordinations among states. They are not likely to improve procedural functions of democracies without a cost that itself is problematic for democracy

    Microdamage repair and remodeling requires mechanical loading

    Bone remodeling is necessary to avoid microdamage accumulation, which could lead to whole-bone failure. Previous studies have shown that this bone-repair mechanism is triggered by osteocyte apoptosis. Through the use of a rodent hindlimb suspension model and tibial four-point bending model, the effects of disuse on microdamage remodeling was examined. At day 0, male rats were assigned to one of three groups: weight bearing (WB), hindlimb suspension (HS), or hindlimb suspension with daily intermittent weight bearing following damage-inducing loading (HW). Within each group, the rats were further divided into subgroups corresponding to three sacrifice time points [day 14 (WB and HS only), day 18, or day 35]. At day 14, animals were anesthetized, and their left tibiae underwent cyclic four-point bending to produce fatigue-induced microdamage. At sacrifice, the tibiae were examined using 3D micro-computed tomography (”CT), flow cytometry, and histologic and immunohistochemical stains. The results indicate that only the WB and HW groups had a significant increase in intracortical TRAP-positive resorption pits following damage induction, which was paralleled by a significant decrease in microdamage over time in combination with a shift in the osteoclast lineage owing to a decrease in monocytes. These results demonstrate that osteocyte apoptosis may be insufficient for repair of microdamage without the stimulation provided through physiologic loading. Pardee Center for the Study of the Longer-Range Future. This was the Center's Inaugural Conference that took place during November 8, 9, and 10, 2001. Organized by David Fromkin, Director Frederick S. Pardee Center for the Study of the Longer-Range Future. Co-Sponsored by Boston University and Carnegie Council on Ethics and International Affairs.This conference brought together a discussion of different perspectives on what future paradigm shifts will look like – in government, in foreign policy, in what constitutes “classics,” in economic and religious modes, and changes in the interaction between these values. The conference agreed that today’s Western society values democracy, constitutionalism, liberalism, rule of law, open society, and market economy. These are not contingent upon one another and may change. But the “needs and aspirations” of humanity will at their most essential core remain the same. The amount and type of power given to governments is not a fixed thing, and developments in the meaning of democracy and how it is achieved may illustrate this

    Hartle-Hawking Wave-Function for Flux Compactifications

    We argue that the topological string partition function, which has been known to correspond to a wave-function, can be interpreted as an exact ``wave-function of the universe'' in the mini-superspace sector of physical superstring theory. This realizes the idea of Hartle and Hawking in the context of string theory, including all loop quantum corrections. The mini-superspace approximation is justified as an exact description of BPS quantities. Moreover this proposal leads to a conceptual explanation of the recent observation that the black hole entropy is the square of the topological string wave-function. This wave-function can be interpreted in the context of flux compactification of all spatial dimensions as providing a physical probability distribution on the moduli space of string compactification.     A generalized quantum microcanonical ensemble

    We discuss a generalized quantum microcanonical ensemble. It describes isolated systems that are not necessarily in an eigenstate of the Hamilton operator. Statistical averages are obtained by a combination of a time average and a maximum entropy argument to resolve the lack of knowledge about initial conditions. As a result, statistical averages of linear observables coincide with values obtained in the canonical ensemble. This was the Center's 2nd annual Conference that took place during November 14, 15, and 16, 2002.The conference brought together some 30 experts from various disciplines to discuss whether the trajectories of the future will be ‘global’ or ‘regional’ in nature.     Modeling multiple ecosystem services, biodiversity conservation, commodity production, and tradeoffs at landscape scales

    Nature provides a wide range of benefits to people. There is increasing consensus about the importance of incorporating these ecosystem services into resource management decisions, but quantifying the levels and values of these services has proven difficult. We use a spatially explicit modeling tool, Integrated Valuation of Ecosystem Services and Tradeoffs (InVEST), to predict changes in ecosystem services, biodiversity conservation, and commodity production levels. We apply InVEST to stakeholder-defined scenarios of land-use/land-cover change in the Willamette Basin, Oregon. We found that scenarios that received high scores for a variety of ecosystem services also had high scores for biodiversity, suggesting there is little tradeoff between biodiversity conservation and ecosystem services. Scenarios involving more development had higher commodity production values, but lower levels of biodiversity conservation and ecosystem services. However, including payments for carbon sequestration alleviates this tradeoff. Quantifying ecosystem services in a spatially explicit manner, and analyzing tradeoffs between them, can help to make natural resource decisions more effective, efficient, and defensible. © The Ecological Society of America

    Drugs with prolonged on-target residence times often show superior efficacy, yet general strategies for optimizing drug-target residence time are lacking. Here we made progress toward this elusive goal by targeting a noncatalytic cysteine in Bruton's tyrosine kinase (BTK) with reversible covalent inhibitors. Using an inverted orientation of the cysteine-reactive cyanoacrylamide electrophile, we identified potent and selective BTK inhibitors that demonstrated biochemical residence times spanning from minutes to 7 d. An inverted cyanoacrylamide with prolonged residence time in vivo remained bound to BTK for more than 18 h after clearance from the circulation. The inverted cyanoacrylamide strategy was further used to discover fibroblast growth factor receptor (FGFR) kinase inhibitors with residence times of several days, demonstrating the generalizability of the approach. Targeting of noncatalytic cysteines with inverted cyanoacrylamides may serve as a broadly applicable platform that facilitates 'residence time by design', the ability to modulate and improve the duration of target engagement in vivo

    Analysis of continuously rotating quadrupole focusing channels using generalized Courant-Snyder theory

    By extending the recently developed generalized Courant-Snyder theory for coupled transverse beam dynamics, we have constructed the Gaussian beam distribution and its projections with arbitrary mode emittance ratios. The new formulation has been applied to a continuously rotating quadrupole focusing channel because the basic properties of this channel are known theoretically and could also be investigated experimentally in a compact setup such as the linear Paul trap configuration. The new formulation retains a remarkably similar mathematical structure to the original Courant-Snyder theory, and thus, provides a powerful theoretical tool to investigate coupled transverse beam dynamics in general and more complex linear focusing channels.close
