Pembuatan Sistem Informasi Pada Bengkel Sepeda Motor X Berbasis Web
Bengkel “X” adalah usaha dagang dalam bidang penyedia sparepart dan servis motor. Proses transaksi jual beli yang dilakukan oleh bengkel ini masih konvesional, akibatnya sering terjadi kehabisan stok sparepart karena sulit untuk mengetahui stok sparepart secara up to date. Reservasi pada bengkel “X” masih bersifat manual, sehingga konsumen harus datang terlebih dahulu untuk melihat ketersediaan montir, lalu melakukan servis. Dari hasil analisis dibutuhkan website dinamis yang menyediakan informasi produk yang sedang promo dan website memberikan alternatif reservasi secara online dimana dari website konsumen dapat memilih tanggan dan waktu reservasi sehingga tidak perlu mengantri untuk melakukan servis. Untuk mengatasi permasalahan tersebut maka dibuatlah sistem informasi sehingga memudahkan dalam melakukan pencatatan proses pembelian dan penjualan, mengetahui stok sparepart, website dapat menangani reservasi secara online dan website yang menyediakan informasi tentang promo sparepart. Dari hasil uji coba sistem informasi ini memudahkan karyawan dalam melakukan pencatatan proses pembelian dan penjualan, mengetahui stok sparepart yang akan habis. Masyarakat mudah untuk memperoleh informasi tentang promo produk yang sedang berlaku, dan memudahkan masyarakat untuk melakukan reservasi online
Exploring the Effectiveness of a Retreat Method for Extension Staff
The California 4-H Association hosted two retreats to support its members with goals of balancing professional development with intentional relationship building. Evaluations demonstrated that staff found the intentional balance of time spent in unstructured, semi-structured, and structured time offered opportunities to grow professionally while building relationships with peers. Follow-up surveys found that 4-H professionals strengthened their network of peers to rely on in their work. Future work for professional development may benefit from a social capital lens
770-5 Chamber Specific Regulation of the Sarcoplasmic Reticulum Calcium ATPase Pump In Human Heart Failure
Alterations in the expression of Ca2+ channels have been described in failing human left ventricle, including down regulation of the ryanodine receptor (RyR)/Ca2+ release channel and the sarcoplasmic reticulum Ca2+ ATPase pump (SERCA) which are involved in excitation-contraction coupling and relaxation (Cir Res 71: 18, 1992). We previously reported chamber specific regulation of the RyR during end-stage human heart failure (Clin Res 42(2):166A. 1994). We investigated whether SERCA is also regulated in the other cardiac chambers during human heart failure. Total RNA and protein homogenates were isolated from the left and right atria (LA, RA) and left and right ventricles (LV, RV) obtained prospectively from 32 cardiac transplant patients and 4 normal controls. Messenger RNA (mRNA) levels of SERCA were quantified using Northern and slot blot hybridizations with a 1.6kb rat cardiac SERCA cDNA probe and normalized to 28S ribosomal levels. Protein levels of SERCA were quantified using enzyme-linked immunosorbent assays with monoclonal antibodies directed against dog cardiac SERCA. Northern analyses detected a single ≈4 kb mRNA in all regions. Compared to controls. SERCA mRNA expression in failing hearts was decreased in LV by 39% (p<0.005), unchanged in RV, and increased in LA by 255% (p<0.005) and in RA by 338% (p<0.025). Consistent with the mRNA data. immunodetectable levels of SERCA were also reduced in LV by 30% (p<0.05) and unchanged in RV; however, protein levels appeared unchanged or reduced in both atria in contrast to the mRNA. This is the first study reporting simultaneous measurements of SERCA mRNA and protein levels in the human heart. We conclude that chamber specific regulation of SERCA mRNA occurs during end-stage heart failure. corroborated by protein expression in the ventricles. Down regulations of SERCA may contribute to impaired relaxation and increased diastolic tone during heart failure
Nonconcordance between Clinical and Head CT Findings: The Specter of Overdiagnosis
Background:. It is unclear whether history and physical examination findings can predict abnormalities on head computed tomography (CT) believed to indicate increased risk of lumbar-puncture- (LP-) induced brain herniation. The objectives of this study were to (1) identify head CT findings felt to be associated with increased risk of brain herniation and (2) to assess the ability of history and physical examination to predict those findings. Methods:. Using a modified Delphi survey technique, an expert panel defined CT abnormalities felt to predict increased risk of LP-induced brain herniation. Presence of such findings on CT was compared with history and physical examination (H&P) variables in 47 patients. Results:. No H&P variable predicted “high-risk” CT; combining H&P variables to improve sensitivity led to extremely low specificity and still failed to identify all patients with high-risk CT. Conclusions:. “High-risk” CT is not uncommon in patients with clinical characteristics known to predict an absence of actual risk from LP, and thus it may not be clinically relevant. “Overdiagnosis” will be increasingly problematic as technological advances identify increasingly subtle deviations from “normal.
Global Diversity of Microbial Communities in Marine Sediment
Microbial life in marine sediment contributes substantially to global biomass and is a crucial component of the Earth system. Subseafloor sediment includes both aerobic and anaerobic microbial ecosystems, which persist on very low fluxes of bioavailable energy over geologic time. However, the taxonomic diversity of the marine sedimentary microbial biome and the spatial distribution of that diversity have been poorly constrained on a global scale. We investigated 299 globally distributed sediment core samples from 40 different sites at depths of 0.1 to 678 m below the seafloor. We obtained ∼47 million 16S ribosomal RNA (rRNA) gene sequences using consistent clean subsampling and experimental procedures, which enabled accurate and unbiased comparison of all samples. Statistical analysis reveals significant correlations between taxonomic composition, sedimentary organic carbon concentration, and presence or absence of dissolved oxygen. Extrapolation with two fitted species–area relationship models indicates taxonomic richness in marine sediment to be 7.85 × 103 to 6.10 × 105 and 3.28 × 104 to 2.46 × 106 amplicon sequence variants for Archaea and Bacteria, respectively. This richness is comparable to the richness in topsoil and the richness in seawater, indicating that Bacteria are more diverse than Archaea in Earth’s global biosphere
Recurrent patterns of DNA copy number alterations in tumors reflect metabolic selection pressures.
Copy number alteration (CNA) profiling of human tumors has revealed recurrent patterns of DNA amplifications and deletions across diverse cancer types. These patterns are suggestive of conserved selection pressures during tumor evolution but cannot be fully explained by known oncogenes and tumor suppressor genes. Using a pan-cancer analysis of CNA data from patient tumors and experimental systems, here we show that principal component analysis-defined CNA signatures are predictive of glycolytic phenotypes, including 18F-fluorodeoxy-glucose (FDG) avidity of patient tumors, and increased proliferation. The primary CNA signature is enriched for p53 mutations and is associated with glycolysis through coordinate amplification of glycolytic genes and other cancer-linked metabolic enzymes. A pan-cancer and cross-species comparison of CNAs highlighted 26 consistently altered DNA regions, containing 11 enzymes in the glycolysis pathway in addition to known cancer-driving genes. Furthermore, exogenous expression of hexokinase and enolase enzymes in an experimental immortalization system altered the subsequent copy number status of the corresponding endogenous loci, supporting the hypothesis that these metabolic genes act as drivers within the conserved CNA amplification regions. Taken together, these results demonstrate that metabolic stress acts as a selective pressure underlying the recurrent CNAs observed in human tumors, and further cast genomic instability as an enabling event in tumorigenesis and metabolic evolution
Quality and correlates of medical record documentation in the ambulatory care setting
BACKGROUND: Documentation in the medical record facilitates the diagnosis and treatment of patients. Few studies have assessed the quality of outpatient medical record documentation, and to the authors' knowledge, none has conclusively determined the correlates of chart documentation. We therefore undertook the present study to measure the rates of documentation of quality of care measures in an outpatient primary care practice setting that utilizes an electronic medical record. METHODS: We reviewed electronic medical records from 834 patients receiving care from 167 physicians (117 internists and 50 pediatricians) at 14 sites of a multi-specialty medical group in Massachusetts. We abstracted information for five measures of medical record documentation quality: smoking history, medications, drug allergies, compliance with screening guidelines, and immunizations. From other sources we determined physicians' specialty, gender, year of medical school graduation, and self-reported time spent teaching and in patient care. RESULTS: Among internists, unadjusted rates of documentation were 96.2% for immunizations, 91.6% for medications, 88% for compliance with screening guidelines, 61.6% for drug allergies, 37.8% for smoking history. Among pediatricians, rates were 100% for immunizations, 84.8% for medications, 90.8% for compliance with screening guidelines, 50.4% for drug allergies, and 20.4% for smoking history. While certain physician and patient characteristics correlated with some measures of documentation quality, documentation varied depending on the measure. For example, female internists were more likely than male internists to document smoking history (odds ratio [OR], 1.90; 95% confidence interval [CI], 1.27 – 2.83) but were less likely to document drug allergies (OR, 0.51; 95% CI, 0.35 – 0.75). CONCLUSIONS: Medical record documentation varied depending on the measure, with room for improvement in most domains. A variety of characteristics correlated with medical record documentation, but no pattern emerged. Further study could lead to targeted interventions to improve documentation
Volunteer Outcomes and Impact: The Contributions and Consequences of Volunteering in 4-H
Youth development volunteers are relied upon by many youth organizations to deliver educational programming, and consequently, fulfill organizations’ missions. Our study explored the consequences of volunteering in youth services by focusing on volunteer outcomes (consequences of volunteering on the volunteer) and volunteer impacts (volunteers’ contributions to the organization). We collected 1,245 open-ended responses to 2 questions from adults who served as volunteers in the University of California 4-H Youth Development Program. We applied iterative inductive thematic analysis without a pre-existing coding scheme to each question independently. Volunteers described their impacts on 4-H as expanding and maintaining programs for children, providing support to and teaching youth, and helping to realize youth outcomes.
