Protective effect of topical Cordia verbenacea in a rat periodontitis model: immune-inflammatory, antibacterial and morphometric assays
BACKGROUND: This study evaluated the effects of C. verbenacea essential oil topically administered in a rat periodontitis model. METHODS: Periodontitis was induced on rats in one of the mandibular first molars assigned to receive a ligature. Animals were randomly divided into two groups: a) non-treatment group (NT) (n = 18): animals received 1mL of vehicle; b) C. verbenacea group (C.v.) (n = 18): animals received 5mg/Kg of essential oils isolated from C. verbenacea. The therapies were administered topically 3 times daily for 11 days. Then, the specimens were processed for morphometric analysis of bone loss. The ligatures were used for microbiological assessment of the presence of Aggregatibacter actinomycetemcomitans, Tannerella forsythia and Porphyromonas gingivalis using PCR. The gingival tissue was collected to Elisa assay of interleukin (IL)-1α and IL-10 levels. RESULTS: Bone loss was inhibited by C. verbenacea when compared to the NT group (p < 0.05). A decrease in the levels of IL-1α and increase in the IL-10 amounts was observed in the C.v. group as compared to NT group (p < 0.05). A lower frequency of P. gingivalis was found in C.v. group (p < 0.05). CONCLUSION: C. verbenacea essential oil topically administered diminished alveolar bone resorption, promoting a positive local imbalance in the pro/anti-inflammatory system and reducing the frequency of detection of P. gingivalis
Biomassa e composição quÃmica de genótipos melhorados de espécies medicinais cultivadas em quatro municÃpios paulistas
The objective of this work was to evaluate the production and chemical constituents of selected genotypes of Artemisia annua, Cordia verbenaceae, Phyllanthus amarus and Mikania laevigata cultivated in Altinópolis, Campinas, Jales and São Carlos. Biomass variations, and qualitative and quantitative differences in plants chemical composition are observed among the locations, with higher values of interesting substances being detected in Jales.O objetivo deste trabalho foi avaliar a produção de biomassa e a composição quÃmica de genótipos selecionados de Artemisia annua (artemisia), Cordia verbenacea (erva-baleeira), Phyllanthus amarus (quebra-pedra) e Mikania laevigata (guaco), nos municÃpios de Altinópolis, Campinas, Jales e São Carlos, no Estado de São Paulo. The Effect of Essential Oils and Bioactive Fractions on Streptococcus mutans
The essential oils (EO) and bioactive fractions (BF) from Aloysia gratissima, Baccharis dracunculifolia, Coriandrum sativum, Cyperus articulatus, and Lippia sidoides were proven to have strong antimicrobial activity on planktonic microorganisms; however, little is known about their effects on the morphology or viability of oral biofilms. Previously, we determined the EO/fractions with the best antimicrobial activity against Streptococcus mutans and Candida spp. In this report, we used a confocal analysis to investigate the effect of these EO and BF on the morphology of S. mutans biofilms (thickness, biovolume, and architecture) and on the metabolic viability of C. albicans biofilms. The analysis of intact treated S. mutans biofilms showed no statistical difference for thickness in all groups compared to the control. However, a significant reduction in the biovolume of extracellular polysaccharides and bacteria was observed for A. gratissima and L. sidoides groups, indicating that these BF disrupt biofilm integrity and may have created porosity in the biofilm. This phenomenon could potentially result in a weakened structure and affect biofilm dynamics. Finally, C. sativum EO drastically affected C. albicans viability when compared to the control. These results highlight the promising antimicrobial activity of these plant species and support future translational research on the treatment of dental caries and oral candidiasis
Antimicrobial Activity of Essential Oils against Streptococcus mutans and their Antiproliferative Effects
This study aimed to evaluate the activity of essential oils (EOs) against Streptococcus mutans biofilm by chemically characterizing their fractions responsible for biological and antiproliferative activity. Twenty EO were obtained by hydrodistillation and submitted to the antimicrobial assay (minimum inhibitory (MIC) and bactericidal (MBC) concentrations) against S. mutans UA159. Thin-layer chromatography and gas chromatography/mass spectrometry were used for phytochemical analyses. EOs were selected according to predetermined criteria and fractionated using dry column; the resulting fractions were assessed by MIC and MBC, selected as active fractions, and evaluated against S. mutans biofilm. Biofilms formed were examined using scanning electron microscopy. Selected EOs and their selected active fractions were evaluated for their antiproliferative activity against keratinocytes and seven human tumor cell lines. MIC and MBC values obtained for EO and their active fractions showed strong antimicrobial activity. Chemical analyses mainly showed the presence of terpenes. The selected active fractions inhibited S. mutans biofilm formation (P < 0.05) did not affect glycolytic pH drop and were inactive against keratinocytes, normal cell line. A OMS estima que a ocorrência global de malária atinge 210 milhões de casos/ano. Responsável por 2 milhões de mortes/ano. A Artemisia annua utilizada a mais de 15 séculos pelos chineses, possui uma substância denominada artemisinina com atividade antimalárica. Devido a falta de informação sobre a nutrição mineral dessa planta, foi proposto o trabalho com o objetivo de conhecer as exigências nutricionais, caracterizar sintomatologia de deficiências e estudar as correlações entre o estado nutricional, a produção de material seco e a produção de artemisinina, foram realizados experimentos de cultivo hidropônico em casa de vegetação. Os tratamentos utilizados foram solução nutritiva completa, com omissão de macronutrientes e com nÃveis de nitrogênio. Os experimentos foram realizados por dois anos consecutivos, com algumas modificações entre eles. Os resultados permitiram concluir-se que: o teor de artemisinina obtido no segundo experimento, para o tratamento completo, pode ser considerado alto nas condições estudadas; o método analÃtico mostrou-se eficiente com a vantagem de quantificar também o ácido artemisinÃnico, que pode ser convertido em artemisinina; as omissões de N e P afetaram a produção.
