    Local reference ranges for full blood count and CD4 lymphocyte count testing

    Objective. Recent advances in full blood count and CD4 technology, coupled with the changing population demographics of the Gauteng region, have necessitated reevaluation of the reference ranges currently in use.Methods. A cross-sectional study of 631 female and 88 male HIV-negative participants from the Gauteng region was performed. Full blood count, automated differential and CD4 count analyses were done using the latest internationally accepted technology. Reference ranges were compiled from the 2.5th and 97.5th percentiles for both male and female participant groups, and gender and ethnic comparisonscalculated by non-parametric tests.Results. Results of 41 females were removed from the statistical analysis because their results were suggestive of possible anaemia. Full blood count reference interval comparison confirmed gender-specific differences in red blood cell and platelet parameters. Ethnic-specific differences were found for some red blood cell parameters in the black female cohort. In addition, black males and females bothgenerally had lower neutrophil and higher lymphocyte counts than a combined Asian/Caucasian/coloured ethnic group.Conclusion. Comparison of the currently calculated referenceranges with published data and reference values in use indicated that a separate ethnic-specific reference range should be introduced for the percentage/absolute neutrophil count and percentage lymphocytes. In addition, locallyerived reference ranges for red cell distribution width(RDW) and CD4 percentage of lymphocytes should be implemented for routine diagnostic testing

    Tree-Based Methods for Discovery of Association between Flow Cytometry Data and Clinical Endpoints

    We demonstrate the application and comparative interpretations of three tree-based algorithms for the analysis of data arising from flow cytometry: classification and regression trees (CARTs), random forests (RFs), and logic regression (LR). Specifically, we consider the question of what best predicts CD4 T-cell recovery in HIV-1 infected persons starting antiretroviral therapy with CD4 count between 200 and 350 cell/μL. A comparison to a more standard contingency table analysis is provided. While contingency table analysis and RFs provide information on the importance of each potential predictor variable, CART and LR offer additional insight into the combinations of variables that together are predictive of the outcome. In all cases considered, baseline CD3-DR-CD56+CD16+ emerges as an important predictor variable, while the tree-based approaches identify additional variables as potentially informative. Application of tree-based methods to our data suggests that a combination of baseline immune activation states, with emphasis on CD8 T-cell activation, may be a better predictor than any single T-cell/innate cell subset analyzed. Taken together, we show that tree-based methods can be successfully applied to flow cytometry data to better inform and discover associations that may not emerge in the context of a univariate analysis

    The impact of dietary protein: lipid ratio on growth performance, fatty acid metabolism, product quality and waste output in Atlantic salmon (Salmo salar)

    A common strategy for aquafeed manufacturers has been the utilisation of relatively large amounts of terrestrial, both animal and plant, oil sources to produce diets with a high energy content. The provision of high fat diets is aimed at promoting the utilisation of energy from lipid, thus increasing the amount of dietary protein used for tissue synthesis. However, in recent years the cost of marine sourced dietary lipids has risen, at the same time, farming operations are under increasing pressure to limit environmental degradation associated with nitrogenous waste effluent. Currently there is limited information available regarding the environmental and economic impacts of an altered dietary protein: lipid ratio in diets for large Atlantic salmon (Salmo salar) reared in seawater, presenting a potential impediment for nutritional based solutions. Accordingly the present study compared two isoenergetic diets with varied protein: lipid ratios via an assessment of growth, fatty acid utilisation, human nutritional quality, nitrogenous waste output and economic considerations. The trial diets were fed to the fish for the final 150 days of an on-farm grow-out period and resulted in minimal differences in fish growth, fatty acid utilisation and fillet quality. A decreased dietary protein: lipid ratio resulted in a more efficient protein utilisation both in terms of digestibility and assimilation into fish and, therefore, nitrogenous waste output was reduced. However, due to small differences in feed utilisation, the cost of fish production was numerically higher

    Digestibility in selected rainbow trout families and modelling of growth from the specific intake of digestible protein

    The experiments aimed to clarify variations in digestibility of dietary nutrients in rainbow trout. Furthermore, the objective was to study how differences in digestibility might be related to growth and feed utilisation at various growth rates. When comparing the results from the experiments it appeared that particularly protein digestibility was closely related to specific growth rate and feed conversion ratio at high growth rates. As a tool to visualise the relationship between protein digestibility and growth of rainbow trout a growth model was developed based on the specific intake of digestible protein, and general assumptions on protein content and protein retention efficiency in rainbow trout. The model indicated that increased protein digestibility only partly explained growth increase and that additional factors were important for growth increment

    Altered levels of shorter vs long-chain omega-3 fatty acids in commercial diets for market-sized Atlantic salmon reared in seawater – Effects on fatty acid composition, metabolism and product quality

    There is a growing trend of ‘replacing’ long-chain omega-3 polyunsaturated fatty acid (n-3 LC PUFA) rich oils with C18 shorter-chain omega-3 polyunsaturated fatty acid rich oils in Atlantic salmon aquafeed formulations. n-3 LC PUFA, including 20:5n-3 and 22:6n-3, play contrasting physiological roles and are metabolised differently in comparison to C18 PUFA. Accordingly, the present study recorded the effect of replacing n-3 LC PUFA rich dietary fish oil with C18 n-3 PUFA rich camelina oil at two inclusion levels in commercial-like diets fed to market-sized Atlantic salmon. This assessment was achieved by an analysis of industry relevant production parameters including growth performance, fatty acid composition and metabolism, nutrient digestibility and consumer acceptance (liking and attribute analysis of fillet). The trial was conducted over the final 150 days of an on-farm grow-out period in seawater. The dietary replacement of n-3 LC PUFA with C18 n-3 PUFA resulted in a significant decrease in fillet n-3 LC PUFA and a poorer growth performance. However, in the absence of fish oil, the inclusion of camelina oil at high levels (40%) contributed to an improved n-6/n-3 ratio and partially ameliorated low dietary n-3 LC PUFA by providing added substrate for endogenous n-3 LC PUFA synthesis in comparison to a 20% camelina oil inclusion. Furthermore, consumer acceptance of Atlantic salmon was unaffected by the dietary addition of camelina oil

    Interactions between dietary docosahexaenoic acid and other long-chain polyunsaturated fatty acids on performance and fatty acid retention in post-smolt Atlantic salmon (Salmo salar)

    A study with varying dietary inclusion levels (1, 5, 10, 15 and 20 g kg-1) of docosahexaenoic acid (DHA; 22:6n-3) was conducted with post-smolt (111 ± 2.6 g; mean ± S.) Atlantic salmon (Salmo salar) over a 9-week period. In addition to the series of DHA inclusion levels, the study included further diets that had DHA at 10 g kg-1 in combination with either eicosapentaenoic acid (EPA; 20:5n-3) or arachidonic acid (ARA; 20:4n-6), both also included at 10 g kg-1. An additional treatment with both EPA and DHA included at 5 g kg-1 (total of 10 g kg-1 long-chain polyunsaturated fatty acids, LC-PUFA) was also included. After a 9-week feeding period, fish were weighed, and carcass, blood and tissue samples collected. A minor improvement in growth was seen with increasing inclusion of DHA. However, the addition of EPA further improved growth response while addition of ARA had no effect on growth. As with most lipid studies, the fatty acid composition of the whole body lipids generally reflected that of the diets. However, there were notable exceptions to this, and these implicate some interactions among the different LC-PUFA in terms of the fatty acid bio- chemistry in this species. At very low inclusion levels, DHA retention was substantially higher (*250 %) than that at all other inclusion levels (31–58 %). The inclusion of EPA in the diet also had a positive effect on the retention efficiency of DHA. However, EPA retention was highly variable and at low DHA inclusion levels there was a net loss of EPA as this fatty acid was most likely elongated to produce DHA, consistent with increased DHA retention with addi- tional EPA in the diet. Retention of DPA (22:5n-3) was high at low levels of DHA, but diminished with increasing DHA inclusion, similar to that seen with DHA retention. The addition of EPA to the diet resulted in a substantial increase in the efficiency of DPA retention; the inclusion of ARA had the opposite effect. Retention of ARA was unaffected by DHA inclusion, but the addition of either EPA or ARA to the diet resulted in a substantial reduction in the efficiency of ARA retention. No effects of dietary treatment were noted on the retention of either linolenic (18:3n-3) or linoleic (18:2n-6) acids. When the total n-3 LC-PUFA content of the diet was the same but consisted of either DHA alone or as a combination of EPA plus DHA, the performance effects were similar
