    Focal Cerebral Infarction in Newborn: Description of Three Cases:

    We observed 3 full-term newborns with focal ischemic injury of the middle cerebral artery (MCA), in which diagnosis of MCA stroke was suspected by US and confirmed by CT scan and MRI. A four-year follow-up was carried out to study the effect of neonatal stroke on neurodevelopmental outcome. All children had a history of pre-perinatal risk factors: neonatal cerebral infarction in term infants, in fact, has many possible causes, including bacterial meningitis, inherited or acquired coagulopathies, trauma and hypoxia-ischemia. The prognosis of neonatal MCA infarction depends on early diagnosis, on the CNS plasticity mechanism and, finally, on medical therapy and neuropsychological rehabilitation

    A safe transoral surgical approach to parapharyngeal tumor arising from deep lobe of parotid gland

    The management of parapharyngeal tumor is surgical, but the approach remains a challenge. Attention should be paid to avoidance intra-operative bleeding or cranial nerves damage. We report a case of a 67-year-old male complaining of left-ear fullness. A submucosal mass arising from the lateral wall of oropharynx on the left side was observed. Magnetic resonance imaging detected a mass arising from the parotid gland, in particular from the deep lobe, and a fine needle biopsy was compatible with "Warthin tumor." We performed a mini-invasive transoral approach under magnification, previous isolation of homolateral vessels. The decision on which surgical approach to be used is determined by site, size vascularity, and histology of the tumor. A literature review of the main surgical approaches was performed. We performed a combined transoral dissection under magnification with cervicotomic exposure of the neck vascular bundle allowing to dissect the tumor and manage any intra-operative complications

    Hyaluronic acid: perspectives in upper aero-digestive tract. A systematic review

    Background: To date, topical therapies guarantee a better delivery of high concentrations of pharmacologic agents to the mucosa of the upper aerodigestive tract (UADT). The use of topical drugs, which are able to reduce mucosal inflammation and to improve healing tissues, can represent a relevant therapeutic advance. Topical sodium hyaluronate (SH) has recently been recognized as adjuvant treatment in the chronic inflammatory disease of the UADT. Aims: The aim of our work was to review the published literature regarding all the potential therapeutic effects of SH in the chronic inflammatory disease of UADT. Methods: Relevant published studies were searched in Pubmed, Google Scholar, Ovid using ("sodium hyaluronate" and "upper airways") or Medical Subject Headings. Results: At the end of our selection process, sixteen publications have been included. Six of them in the post-operative period of nasal-sinus surgery, 2 of them in pediatric patients affected by recurrent upper respiratory tract infections, 4 of them in reducing symptoms and preventing exacerbations of chronic upper airways in adult population, 4 of them in patients with chronic inflammatory disease of UADT, including gastro-esophageal reflux disease (GERD). Conclusions: Topical administration of SH plays a pivotkey role in the postoperative phase of patients undergoing FESS and nasal surgery, and positive results are generally observed in all the patients suffering from UADT chronic inflammatory disease

    Hybrids and allied species as potential rootstocks for eggplant: Effect of grafting on vigour, yield and overall fruit quality traits

    Grafting of fruiting vegetables is an effective technique to overcome pests and diseases in modern cropping systems and it is often used to improve yield and fruit quality. Eggplant is an important vegetable crop that benefits significantly from grafting. In this regards, the exploitation, valorization and breeding of new rootstock genotypes as possible substitute to those commonly used (Solanum torvum and tomato hybrids) would permit an intensive eggplant crop system in those situations where a rootstock rotation is required. In the present article, we study the effects of several potential rootstocks including both wild/allied species of eggplant [S. torvum (STO), S. macrocarpon (SMA), S. aethiopicum (accession SASI), S. aethiopicum (accession SASa2), S. paniculatum (jurubeba) (SPA) and S. indicum (SIN)] and Msa 2/2 E7 and 460 CAL. eggplant hybrids on plant vigor, yield and fruit characteristics of eggplant F1 hybrid (‘Birgah’), in two spring-summer growing seasons (2014 and 2015). SPA and the hybrids Msa 2/2 E7 and 460 CAL. displayed a high percentage of grafting success. ‘Birgah’ scion grafted onto the two above-mentioned rootstocks showed a notable vigour and yield. Both rootstocks did not promote any unfavorable effects on apparent fruit quality traits and overall fruit composition. Furthermore, the concentration of glycoalkaloids in the fruit remained below the recommended safety value (200 mg/100 g of dw). These results suggest that SPA and Msa 2/2 E7 and 460 CAL. eggplant hybrids might represent a potential rootstock alternative to S. torvum

    Solanum aethiopicum gr. gilo and Its Interspecific Hybrid with S. melongena as Alternative Rootstocks for Eggplant: Effects on Vigor, Yield, and Fruit Physicochemical Properties of Cultivar 'Scarlatti'

    Grafting is generally considered eective in ameliorating vegetable crop tolerance to bioticand abiotic stresses. The use of interspecific hybrid as rootstock for eggplant may represent a valid alternative approach to enhance eggplant performance. However, studies on the eects of different rootstocks on eggplant plant vigor, yield, and fruit quality traits often show conflicting results. Thus, an experiment was performed in two spring\u2013summer growing seasons (2014 and 2015) by grafting eggplant 'Scarlatti' F1 hybrid on two accessions of S. aethiopicum gr. gilo and on the interspecific hybrid S. melongena x S. aehtiopicum gr. gilo in comparison to the most common eggplant rootstock S. torvum. Results indicate that S. melongena x S. aethiopicum gr. gilo interspecific hybrid and S. torvum improved grafting success, plant vigor, early flowering and yield in 'Scarlatti' F1 scion. All rootstocks tested did not negatively influence fruit apparent quality traits and fruit quality composition. Moreover, fruit glycoalkaloids content remained below the recommended threshold value. These findings suggest that the use of S. melongena x S. aethiopicum gr. gilo interspecific hybrid as rootstock may be a good alternative to the most commonly used S. torvum

    Geological and geophysical characterization of the Brindisi di Montagna Scalo landslide (Basilicata, Southern Italy)

    The Brindisi di Montagna Scalo Landslide in Southern Italy is an active complex mass movement, which affects the left slope of the Basento River. In the last few decades, this landslide has been continuously monitored, as it directly threatened some of the most important communication routes in the Basilicata Region. Nevertheless, little progresses have been made to prevent further landslide advancement, and continuous maintenance is required. With the aims of better understanding, the main factors controlling the evolution of this landslide, and suggesting the most appropriate countermeasures, a multidisciplinary study, based on the integration of direct and indirect techniques, was carried out. Direct techniques included multi-temporal geomorphological analysis of the slope, alongside geological and structural field observations. Indirect techniques consisted of electrical resistivity tomography acquisition. The combined analyses of the geological and geophysical data showed that Quaternary tectonic processes played a fundamental role as a predisposing factor, whereas seasonal rainfall, and the perpetual undercutting by erosional processes caused by the Basento River at the toe of the landslide are the main triggering mechanisms. The Brindisi di Montagna Scalo Landslide represents an outstanding case-study, concerning the interaction between a flow-like complex landslide and essential linear infrastructure, such as motorways and railways

    Mechanisms of HIV-1 Nucleocapsid Protein Inhibition by Lysyl-Peptidyl-Anthraquinone Conjugates

    The Nucleocapsid protein NCp7 (NC) is a nucleic acid chaperone responsible for essential steps of the HIV-1 life cycle and an attractive candidate for drug development. NC destabilizes nucleic acid structures and promotes the formation of annealed substrates for HIV-1 reverse transcription elongation. Short helical nucleic acid segments bordered by bulges and loops, such as the Trans-Activation Response element (TAR) of HIV-1 and its complementary sequence (cTAR), are nucleation elements for helix destabilization by NC and also preferred recognition sites for threading intercalators. Inspired by these observations, we have recently demonstrated that 2,6-disubstituted peptidylanthraquinone-conjugates inhibit the chaperone activities of recombinant NC in vitro, and that inhibition correlates with the stabilization of TAR and cTAR stem-loop structures. We describe here enhanced NC inhibitory activity by novel conjugates that exhibit longer peptidyl chains ending with a conserved Nterminal lysine. Their efficient inhibition of TAR/cTAR annealing mediated by NC originates from the combination of at least three different mechanisms, namely, their stabilizing effects on nucleic acids dynamics by threading intercalation, their ability to target TAR RNA substrate leading to a direct competition with the protein for the same binding sites on TAR, and, finally, their effective binding to the NC protein. Our results suggest that these molecules may represent the stepping-stone for the future development of NC-inhibitors capable of targeting the protein itself and its recognition site in RNA

    Proteome analysis of human Wharton's jelly cells during in vitro expansion

    <p>Abstract</p> <p>Background</p> <p>The human umbilical cord contains mucoid connective tissue and fibroblast-like cells. These cells named Wharton's jelly cells, (WJCs) display properties similar to mesenchymal stem cells therefore representing a rich source of primitive cells to be potentially used in regenerative medicine.</p> <p>Results</p> <p>To better understand their self-renewal and potential <it>in vitro </it>expansion capacity, a reference 2D map was constructed as a proteomic data set. 158 unique proteins were identified. More than 30% of these proteins belong to cytoskeleton compartment. We also found that several proteins including Shootin1, Adenylate kinase 5 isoenzyme and Plasminogen activator-inhibitor 2 are no longer expressed after the 2<sup>nd </sup>passage of <it>in vitro </it>replication. This indicates that the proliferative potency of these cells is reduced after the initial stage of <it>in vitro </it>growing. At the end of cellular culturing, new synthesized proteins, including, ERO1-like protein alpha, Aspartyl-tRNA synthetase and Prolyl-4-hydroxylase were identified. It is suggested that these new synthesized proteins are involved in the impairment of cellular surviving during replication and differentiation time.</p> <p>Conclusions</p> <p>Our work represents an essential step towards gaining knowledge of the molecular properties of WJCs so as to better understand their possible use in the field of cell therapy and regenerative medicine.</p

    Frailty and Sarcopenia in Older Patients Receiving Kidney Transplantation

    Kidney transplantation is the treatment of choice for most of the patients with end-stage renal disease (ESRD). It improves quality of life, life expectancy, and has a lower financial burden to the healthcare system in comparison to dialysis. Every year more and more older patients are included in the kidney transplant waitlist. Within this patient population, transplanted subjects have better survival and quality of life as compared to those on dialysis. It is therefore crucial to select older patients who may benefit from renal transplantation, as well as those particularly at risk for post-transplant complications. Sarcopenia and frailty are frequently neglected in the evaluation of kidney transplant candidates. Both conditions are interrelated complex geriatric syndromes that are linked to disability, aging, comorbidities, increased mortality, and graft failure post-transplantation. Chronic kidney disease (CKD) and more importantly ESRD are characterized by multiple metabolic complications that contribute for the development of sarcopenia and frailty. In particular, anorexia, metabolic acidosis and chronic low-grade inflammation are the main contributors to the development of sarcopenia, a key component in frail transplant candidates and recipients. Both frailty and sarcopenia are considered to be reversible. Frail patients respond well to multiprofessional interventions that focus on the patients' positive frailty criteria, while physical rehabilitation and oral supplementation may improve sarcopenia. Prospective studies are still needed to evaluate the utility of formally measuring frailty and sarcopenia in the older candidates to renal transplantation as part of the transplant evaluation process

    Synthesis, characterization, crystal structures and in vitro antistaphylococcal activity of organotin(IV) derivatives with 5,7-disubstituted-1,2,4-triazolo[1,5-a]pyrimidine

    New organotin(IV) complexes of 5,7-ditertbutyl-1,2,4-triazolo[1,5-a]pyrimidine (dbtp) and 5,7-diphenyl- 1,2,4-triazolo[1,5-a]pyrimidine (dptp) with 1:1 and/or 1:2 stoichiometry were synthesized and investigated by X-ray diffraction, FT-IR and 119Sn Mössbauer in the solid state and by 1H and 13C NMR spectroscopy, in so- lution. Moreover, the crystal and molecular structures of Et2SnCl2(dbtp)2 and Ph2SnCl2(EtOH)2(dptp)2 are reported. The complexes contain hexacoordinated tin atoms: in Et2SnCl2(dbtp)2 two 5,7-ditertbutyl-1,2,4- triazolo[1,5-a]pyrimidine molecules coordinate classically the tin atom through N(3) atom and the coordina- tion around the tin atom shows a skew trapezoidal structure with axial ethyl groups. In Ph2SnCl2(EtOH)2 (dptp)2 two ethanol molecules coordinate tin through the oxygen atom and the 5,7-diphenyl-1,2,4-triazolo [1,5-a]pyrimidine molecules are not directly bound to the metal center but strictly H-bonded, through N (3), to the \OH group of the ethanol moieties; Ph2SnCl2(EtOH)2(dptp)2 has an all-trans structure and the C–Sn–C fragment is linear. On the basis of Mössbauer data, the 1:2 diorganotin(IV) complexes are advanced to have the same structure of Et2SnCl2(dbtp)2, while Me2SnCl2(dptp)2 to have a regular all-trans octahedral structure. A distorted cis-R2 trigonal bipyramidal structure is assigned to 1:1 diorganotin(IV) complexes. The in vitro antibacterial activities of the synthesized complexes have been tested against a group of reference pathogen micro-organisms and some of them resulted active with MIC values of 5 μg/mL, most of all against staphylococcal strains, which shows their inhibitory effect
