    Phase-Retrieval with Incomplete Autocorrelations Using Deep Convolutional Autoencoders

    Phase-retrieval techniques aim to recover the original signal from just the modulus of its Fourier transform, which is usually much easier to measure than its phase, but the standard iterative techniques tend to fail if only part of the modulus information is available. We show that a neural network can be trained to perform phase retrieval using only incomplete information, and we discuss advantages and limitations of this approach

    Un approccio nutrizionale potrebbe essere una strategia promettente per prevenire o rallentare la progressione di malattie neurodegenerative come il morbo di Parkinson e il morbo di Alzheimer, poiché finora non esiste l'eectiveterapia per queste malattie. Gli effetti benefici degli acidi grassi omega-3 sono ora ben consolidati da una pletora di studi attraverso il loro coinvolgimento in molteplici funzioni biochimiche, tra cui la sintesi di mediatori antinfiammatori, fluidità della membrana cellulare, segnalazione intracellulare ed espressione genica. Questa revisione sistematica prenderà in considerazione studi epidemiologici e studi clinici che hanno valutato l'impatto dell'integrazione o dell'assunzione dietetica di acidi grassi polinsaturi omega-3 su malattie neurodegenerative come il morbo di Parkinson e l'Alzheimer. In effetti, il trattamento con acidi grassi omega-3, essendo sicuro e ben tollerato, rappresenta uno strumento prezioso e biologicamente plausibile nella gestione delle malattie neurodegenerative nelle loro fasi iniziali.A nutritional approach could be a promising strategy to prevent or slow the progression of neurodegenerative diseases such as Parkinson’s and Alzheimer’s disease, since there is no eectivetherapy for these diseases so far. The beneficial effects of omega-3 fatty acids are now well established by a plethora of studies through their involvement in multiple biochemical functions, including synthesis of anti-inflammatory mediators, cell membrane fluidity, intracellular signaling, and gene expression. This systematic review will consider epidemiological studies and clinical trials that assessed the impact of supplementation or dietary intake of omega-3 polyunsaturated fatty acids on neurodegenerative diseases such as Parkinson’s and Alzheimer’s diseases. Indeed, treatment with omega-3 fatty acids, being safe and well tolerated, represents a valuable and biologically plausible tool in the management of neurodegenerative diseases in their early stages

    Evaluation of Side Effects and Long-Term Protection of a Sustained-Release Injectable Moxidectin Formulation against Dirofilaria immitis Infection in Dogs: An Observational-In Field Multicentric Study

    SIMPLE SUMMARY: The sustained-release moxidectin formulation of Afilaria SR is labelled to prevent Dirofilaria immitis infection in dogs for a period of six months. An observational—in field multicentric study was design to evaluate the tolerability and the long-term prevention of Afilaria SR in Italy. A total of 583 dogs were recruited from 2018 to 2021, receiving the drug annually and monitored by veterinary practitioners after each administration. Antigenic tests were performed 210, 365, 730, and 1095 days after the administration of the drug. None of the enrolled dogs was detected as positive, since it was possible to establish that 100% of protection was achieved. Afilaria SR was well tolerated since only the 13% of dogs demonstrated mild reaction in the injection site and only two dogs out of 583 demonstrated anaphylactoid or angioneurotic reactions. These data support the high prevention rate against Dirofilaria immitis disease in all enrolled dogs and indicate the high safety profile of the product, considering the low number and the low grade of side effects. ABSTRACT: The sustained-release moxidectin formulation Afilaria SR is a relatively new product and has been labelled to prevent Dirofilaria immitis infection in dogs for a six months-period. An observational, in field multicentric study was performed, aiming to evaluate the tolerability and the long-term prevention of Afilaria SR in Italy, a country where filariasis is endemic. The study was designed to include not less than 300 dogs, older than 6 months, of any breed. Side effects were recorded by veterinarians and antigenic tests were performed after 210, 365, 730, and 1095 days after the administration of the drug. A total of 583 dogs were recruited from 2018 to 2021 and all of them were negative with respect to antigenic tests at all time points, indicating that 100% of protection was achieved. Ranking of adverse reactions and correlation to patient features were analyzed using descriptive statistics and χ2 square test, respectively. Afilaria SR was well tolerated: 13% of dogs experienced mild reactions and only two dogs out of 583 (0.3%) demonstrated anaphylactoid/angioneurotic reactions, resolved administering corticosteroids. These data support that Afilaria SR prevented Dirofilaria immitis disease in all enrolled dogs and the low number and the low grade of side effects indicate the high safety profile of the product

    Positron emission tomography in the diagnostic work-up of screening-detected lung nodules

    Low-dose computed tomography (CT) screening for lung cancer can reduce lung cancer mortality, but overdiagnosis, false positives and invasive procedures for benign nodules are worrying. We evaluated the utility of positron emission tomography (PET)-CT in characterising indeterminate screeningdetected lung nodules. 383 nodules, examined by PET-CT over the first 6 years of the COSMOS (Continuous Observation of Smoking Subjects) study to diagnose primary lung cancer, were reviewed and compared with pathological findings (surgically-treated patients) or follow-up (negative CT for â\u89¥2 years, considered negative); 196 nodules were malignant. The sensitivity, specificity and accuracy of PET-CT for differentially diagnosing malignant nodules were, respectively, 64%, 89% and 76% overall, and 82%, 92% and 88% for baseline-detected nodules. Performance was lower for nodules found at repeat annual scans, with sensitivity ranging from 22% for nonsolid to 79% for solid nodules (p=0.0001). Sensitivity (87%) and specificity (73%) were high for nodules â\u89¥15 mm, better (sensitivity 98%) for solid nodules â\u89¥15 mm. PET-CT was highly sensitive for the differential diagnosis of indeterminate nodules detected at baseline, nodules â\u89¥15 mm and solid nodules. Sensitivity was low for sub-solid nodules and nodules discovered after baseline for which other methods, e.g. volume doubling time, should be used

    Rapid SARS-CoV-2 intra-host and within-household emergence of novel haplotypes

    In February 2020, the municipality of Vo’, a small town near Padua (Italy) was quarantined due to the first coronavirus disease 19 (COVID-19)-related death detected in Italy. To investigate the viral prevalence and clinical features, the entire population was swab tested in two sequential surveys. Here we report the analysis of 87 viral genomes, which revealed that the unique ancestor haplotype introduced in Vo’ belongs to lineage B, carrying the mutations G11083T and G26144T. The viral sequences allowed us to investigate the viral evolution while being transmitted within and across households and the effectiveness of the non-pharmaceutical interventions implemented in Vo’. We report, for the first time, evidence that novel viral haplotypes can naturally arise intra-host within an interval as short as two weeks, in approximately 30% of the infected individuals, regardless of symptom severity or immune system deficiencies. Moreover, both phylogenetic and minimum spanning network analyses converge on the hypothesis that the viral sequences evolved from a unique common ancestor haplotype that was carried by an index case. The lockdown extinguished both the viral spread and the emergence of new variant

    Highly Pathogenic Avian Influenza Virus Subtype H5N1 in Africa: A Comprehensive Phylogenetic Analysis and Molecular Characterization of Isolates

    Highly pathogenic avian influenza virus A/H5N1 was first officially reported in Africa in early 2006. Since the first outbreak in Nigeria, this virus spread rapidly to other African countries. From its emergence to early 2008, 11 African countries experienced A/H5N1 outbreaks in poultry and human cases were also reported in three of these countries. At present, little is known of the epidemiology and molecular evolution of A/H5N1 viruses in Africa. We have generated 494 full gene sequences from 67 African isolates and applied molecular analysis tools to a total of 1,152 A/H5N1 sequences obtained from viruses isolated in Africa, Europe and the Middle East between 2006 and early 2008. Detailed phylogenetic analyses of the 8 gene viral segments confirmed that 3 distinct sublineages were introduced, which have persisted and spread across the continent over this 2-year period. Additionally, our molecular epidemiological studies highlighted the association between genetic clustering and area of origin in a majority of cases. Molecular signatures unique to strains isolated in selected areas also gave us a clearer picture of the spread of A/H5N1 viruses across the continent. Mutations described as typical of human influenza viruses in the genes coding for internal proteins or associated with host adaptation and increased resistance to antiviral drugs have also been detected in the genes coding for transmembrane proteins. These findings raise concern for the possible human health risk presented by viruses with these genetic properties and highlight the need for increased efforts to monitor the evolution of A/H5N1 viruses across the African continent. They further stress how imperative it is to implement sustainable control strategies to improve animal and public health at a global level
