    Evaluation of Native Grasses for Sustainable Turfgrass in the Bioclimatic Mediterranean Region

    This study reports the results of a research project (Mi.T.E.A.Med) funded by the Italian Ministry of Agriculture. The research was organised in two phases: the first one involved the screening of the study area (Southern Italy) to find suitable turfgrass species and the second one focused on ex situ cultivation to test the ecotypes with salinity resistance. During the first step of the research, 11 sites from 6 regions of Southern and Central Italy were identified. In these sites, 24 ecotypes of Cynodon dactylon (L.) Pers. were collected and their habitus, phenology, and some biometric parameters have been determined. During the 2 years of research, both botanic and agronomic characterisation of the collected C. dactylon ecotypes was carried out. Some native accessions showed a behaviour similar to commercial cultivars, while an ecotype from the Abruzzo Region showed better results compared to the commercial cultivars for several quality indices. The results of this project showed that Mediterranean-adapted native grass species (e.g. Cynodon dactylon (L.) Pers.) are worth investigating for turfgrass, making for their performance and low resource requirement (especially water). This species can be used as promising alternatives to conventional non-native turfgrasses

    1,3,4-Oxadiazole and 1,3,4-Thiadiazole Nortopsentin Derivatives against Pancreatic Ductal Adenocarcinoma: Synthesis, Cytotoxic Activity, and Inhibition of CDK1

    A new series of nortopsentin analogs, in which the central imidazole ring of the natural lead was replaced by a 1,3,4-oxadiazole or 1,3,4-thiadiazole moiety, was efficiently synthesized. The antiproliferative activity of all synthesized derivatives was evaluated against five pancreatic ductal adenocarcinoma (PDAC) cell lines, a primary culture and a gemcitabine-resistant variant. The five more potent compounds elicited EC50 values in the submicromolar-micromolar range, associated with a significant reduction in cell migration. Moreover, flow cytometric analysis after propidium iodide staining revealed an increase in the G2-M and a decrease in G1-phase, indicating cell cycle arrest, while a specific ELISA demonstrated the inhibition of CDK1 activity, a crucial regulator of cell cycle progression and cancer cell proliferation

    Acquisition Parameters Influence Diffusion Metrics Effectiveness in Probing Prostate Tumor and Age-Related Microstructure

    : This study aimed to investigate the Diffusion-Tensor-Imaging (DTI) potential in the detection of microstructural changes in prostate cancer (PCa) in relation to the diffusion weight (b-value) and the associated diffusion length lD. Thirty-two patients (age range = 50-87 years) with biopsy-proven PCa underwent Diffusion-Weighted-Imaging (DWI) at 3T, using single non-zero b-value or groups of b-values up to b = 2500 s/mm2. The DTI maps (mean-diffusivity, MD; fractional-anisotropy, FA; axial and radial diffusivity, D// and D┴), visual quality, and the association between DTI-metrics and Gleason Score (GS) and DTI-metrics and age were discussed in relation to diffusion compartments probed by water molecules at different b-values. DTI-metrics differentiated benign from PCa tissue (p ≤ 0.0005), with the best discriminative power versus GS at b-values ≥ 1500 s/mm2, and for b-values range 0-2000 s/mm2, when the lD is comparable to the size of the epithelial compartment. The strongest linear correlations between MD, D//, D┴, and GS were found at b = 2000 s/mm2 and for the range 0-2000 s/mm2. A positive correlation between DTI parameters and age was found in benign tissue. In conclusion, the use of the b-value range 0-2000 s/mm2 and b-value = 2000 s/mm2 improves the contrast and discriminative power of DTI with respect to PCa. The sensitivity of DTI parameters to age-related microstructural changes is worth consideration

    Interferon-Tau in Maternal Peripheral Blood and Its Relationship with Progesterone and Pregnancy-Associated Glycoproteins in the Early Phases of Gestation in Water Buffalo

    The aim of this study was to investigate the interferon tau (IFNt) concentration in the peripheral maternal blood during the early phase of pregnancy in buffalo cows and improve the knowledge on the physiological importance of circulating IFNt, evaluating the possible interaction with pregnancy-associated glycoproteins (PAGs) and progesterone (P4). Blood samples were taken from buffalo cows on day 0 (day of AI), 7, 14, 18, 28, and 40 post insemination for the IFNt, PAG, and P4 analysis and to determine the IFNt mRNA expression. The animals were categorized ex post into Pregnant, Non-pregnant and Embryo mortality groups. The interferon value was influenced by group (p = 0.003), being always higher in pregnant buffalo cows than in non-pregnant ones, while the embryo mortality group showed intermediate values between those for pregnant and non-pregnant animals. The mRNA expression of IFNt was not influenced by groups or any time points. The regression analysis that included IFNt as the independent variable showed that PAGs, from day 18 (p < 0.01), and P4, from day 28 (p < 0.05), were positively associated with IFNt values. The close associations among IFNt, PAGs and P4 demonstrate that all three molecules work together for fetal–placental well-being and pregnancy support. Unfortunately, the great individual variability in circulating IFNt makes this analysis unsuitable for early pregnancy diagnosis

    MICROTREMOR MEASUREMENTS IN PALERMO, ITALY: A COMPARISON WITH MACROSEISMIC INTENSITY AND EARTHQUAKE GROUND MOTION

    The city of Palermo is an appropriate test site where the efficiency of microtremors in predicting ground motion properties during earthquakes can be checked. Palermo is a densely populated city with important historical heritage and was object of previous studies. Areas of local amplification of damage were identified in downtown Palermo using historical macroseismic data. Moreover, aftershocks of the September 6, 2002, earthquake (Mw 5.9, 40 km offshore) provided a dataset of seismograms that quantify spatial variations of ground motion. The availability of more than 2000 boreholes in the city allowed a reconstruction of the 3D structure of surface geology, indicating that all the higher damage zones correspond to sediment-filled valleys. The high variability of the surface geology is mostly due to the presence of two filled river-beds of about 150 m width. In the framework of the SESAME project (Seismic EffectS assessment using Ambient Exctations, funded by the European Union), 90 microtremor measurements were performed across several profiles crossing the soft sediment bodies. The measurement points were intensified close to the valley edges (every 20 m), according to our geological reconstruction. H/V spectral ratio on ambient noise (HVSR) show significant variations along each profile: as soon as the transition stiff to soft is crossed, a typical spectral peak exceeding a factor of 3 in amplitude appears in the HVSR. The peak falls between 1 and 2 Hz and, along each profile, the peak disappears as soon as the other edge of the valley is crossed. These results indicate that microtremors are sensitive to the presence of large impedance contrasts of deep soft soil, at least in the Palermo area, with an important implication: the HVSR method seems to be able to recognize conditions potentially favourable to the occurrence of higher damage even when local geological characters are masked by the urban growth. However, we were not able to establish a quantitative correlation between microtremor properties and ground motion (or damage) amplification

    Time trend prevalence of artificial nutrition counselling in a university hospital.

    Abstract Objectives The negative effects of malnutrition on the prognosis of hospitalized patients are well documented; however, less known is the awareness and knowledge of health care professionals about this complication. The aim of this study was to evaluate the trend of the requests for nutritional consultation in years and the prescription of artificial nutrition (AN), for adult patients at a university hospital in southern Italy in the years 2004, 2008, 2012, and 2016 to assess the progress of medical teams concerning awareness of hospital malnutrition. Methods This was a retrospective study that evaluated the time trend of nutritional consultation requests and related prescription of AN, for adult patients at a university hospital in southern Italy in the years 2004, 2008, 2012, and 2016. Of 112 233 inpatients, 2505 received a nutritional consultation with the prescription of AN. Results The number of patients on AN increased from 507 of 33 240 (1.52%) in 2004 to 730 of 29 195 (2.5%) in 2008 (P The request for AN was quite equally distributed between surgical (51.5%) and medical wards (48.5%), with a prevalence among patients with oncologic diseases (806 patients [65.6%]). As for nononcologic diseases, 20.4% involved the gastrointestinal tract and 6.3% the nervous system. Throughout the 12 y of observation, parenteral nutrition was the main prescribed support (59.8%) followed by oral nutritional supplements (26.1%) and enteral nutrition (9.3%). Mean nutritional intervention duration was 11 d (±10.8 d). Conclusions The request of AN for hospitalized patients increased over time, probably owing to improved medical consciousness of the potential risks for malnutrition and the availability of a specialized clinical nutrition team

    Imidazo[2,1-b] [1,3,4]thiadiazoles with antiproliferative activity against primary and gemcitabine-resistant pancreatic cancer cells

    A new series of eighteen imidazo [2,1-b] [1,3,4]thiadiazole derivatives was efficiently synthesized and screened for antiproliferative activity against the National Cancer Institute (NCI-60) cell lines panel. Two out of eighteen derivatives, compounds 12a and 12h, showed remarkably cytotoxic activity with the half maximal inhibitory concentration values (IC50) ranging from 0.23 to 11.4 mM, and 0.29e12.2 mM, respectively. However, two additional compounds, 12b and 13g, displayed remarkable in vitro antiproliferative activity against pancreatic ductal adenocarcinoma (PDAC) cell lines, including immortalized (SUIT-2, Capan-1, Panc-1), primary (PDAC-3) and gemcitabine-resistant (Panc-1R), eliciting IC50 values ranging from micromolar to sub-micromolar level, associated with significant reduction of cell-migration and spheroid shrinkage. These remarkable results might be explained by modulation of key regulators of epithelial-to-mesenchymal transition (EMT), including E-cadherin and vimentin, and inhibition of metalloproteinase-2/-9. High-throughput arrays revealed a significant inhibition of the phosphorylation of 45 tyrosine kinases substrates, whose visualization on Cytoscape highlighted PTK2/FAK as an important hub. Inhibition of phosphorylation of PTK2/FAK was validated as one of the possible mechanisms of action, using a specific ELISA. In conclusion, novel imidazothiadiazoles show potent antiproliferative activity, mediated by modulation of EMT and PTK2/FAK
