    An Innovative Methodological Approach for Monitoring and Chemical Characterization of Odors around Industrial Sites

    This study aims to highlight the potentialities of an innovative methodological approach for monitoring and chemical characterization of odors, especially in high concern and complex industrial areas. The proposed approach was developed in order to monitor and identify odor-active compounds responsible for odor annoyance coming from different industrial activities such as landfills, wastewater treatment plants, and petroleum plants. The methodology's strengths are as follows: (1) the tailored approach for each typology of industrial areas/sites; (2) integration of technologies able to provide real-time information about the emissive sources; (3) mapping of air pollutants on the territory aimed to identify and discriminate among different fugitive emissions responsible for odor annoyance; (4) collection of more representative air samples only during the nuisance events, thanks to the implementation of innovative sampling systems and citizens' involvement; and (5) increased analytical sensitivity in odor-active VOCs detection. This methodology reveals to be a useful tool to collect real-time information about the emission sources and their impacts on the surrounding area giving credit to citizens' complaints. Moreover, it allows to overcome the limitations of the conventional approaches related to the lack of instrumental sensitivity and to identify the chemical compounds contributing to the odor annoyance

    Genetic analysis of the role of protein kinase Ctheta in platelet function and thrombus formation.

    BACKGROUND: PKCtheta is a novel protein kinase C isozyme, predominately expressed in T cells and platelets. PKCtheta(-/-) T cells exhibit reduced activation and PKCtheta(-/-) mice are resistant to autoimmune disease, making PKCtheta an attractive therapeutic target for immune modulation. Collagen is a major agonist for platelets, operating through an immunoreceptor-like signalling pathway from its receptor GPVI. Although it has recently been shown that PKCtheta positively regulates outside-in signalling through integrin alpha(IIb)beta(3) in platelets, the role of PKCtheta in GPVI-dependent signalling and functional activation of platelets has not been assessed. METHODOLOGY/PRINCIPAL FINDINGS: In the present study we assessed static adhesion, cell spreading, granule secretion, integrin alpha(IIb)beta(3) activation and platelet aggregation in washed mouse platelets lacking PKCtheta. Thrombus formation on a collagen-coated surface was assessed in vitro under flow. PKCtheta(-/-) platelets exhibited reduced static adhesion and filopodia generation on fibrinogen, suggesting that PKCtheta positively regulates outside-in signalling, in agreement with a previous report. In contrast, PKCtheta(-/-) platelets also exhibited markedly enhanced GPVI-dependent alpha-granule secretion, although dense granule secretion was unaffected, suggesting that PKCtheta differentially regulates these two granules. Inside-out regulation of alpha(IIb)beta(3) activation was also enhanced downstream of GPVI stimulation. Although this did not result in increased aggregation, importantly thrombus formation on collagen under high shear (1000 s(-1)) was enhanced. CONCLUSIONS/SIGNIFICANCE: These data suggest that PKCtheta is an important negative regulator of thrombus formation on collagen, potentially mediated by alpha-granule secretion and alpha(IIb)beta(3) activation. PKCtheta therefore may act to restrict thrombus growth, a finding that has important implications for the development and safe clinical use of PKCtheta inhibitors

    α-Mannoside ÎČ-1,6-N-acetylglucosaminyltransferase V (MGAT5) is a mammalian glycosyltransferase involved in complex N-glycan formation, which strongly drives cancer when overexpressed. Despite intense interest, the catalytic mechanism of MGAT5 is not known in detail, precluding therapeutic exploitation. We solved structures of MGAT5 complexed to glycosyl donor and acceptor ligands, revealing an unforeseen role for donor-induced loop rearrangements in controlling acceptor substrate engagement. QM/MM metadynamics simulations of MGAT5 catalysis highlight the key assisting role of Glu297 and reveal considerable conformational distortions imposed upon the glycosyl donor during transfer. Detailed mechanistic characterization of MGAT5 will aid inhibitor development to correct cancer-associated N-glycosylation

    Breath analysis for early detection of malignant pleural mesothelioma: Volatile organic compounds (VOCs) determination and possible biochemical pathways

    Malignant pleural mesothelioma (MPM) is a rare neoplasm, mainly caused by asbestos exposure, with a high mortality rate. The management of patients with MPM is controversial due to a long latency period between exposure and diagnosis and because of non-specific symptoms generally appearing at advanced stage of the disease. Breath analysis, aimed at the identification of diagnostic Volatile Organic Compounds (VOCs) pattern in exhaled breath, is believed to improve early detection of MPM. Therefore, in this study, breath samples from 14 MPM patients and 20 healthy controls (HC) were collected and analyzed by Thermal Desorption-Gas Chromatography-Mass Spectrometry (TD-GC/MS). Nonparametric test allowed to identify the most weighting variables to discriminate between MPM and HC breath samples and multivariate statistics were applied. Considering that MPM is an aggressive neoplasm leading to a late diagnosis and thus the recruitment of patients is very difficult, a promising data mining approach was developed and validated in order to discriminate between MPM patients and healthy controls, even if no large population data are available. Three different machine learning algorithms were applied to perform the classification task with a leave-one-out cross-validation approach, leading to remarkable results (Area Under Curve AUC = 93%). Ten VOCs, such as ketones, alkanes and methylate derivates, as well as hydrocarbons, were able to discriminate between MPM patients and healthy controls and for each compound which resulted diagnostic for MPM, the metabolic pathway was studied in order to identify the link between VOC and the neoplasm. Moreover, five breath samples from asymptomatic asbestos-exposed persons (AEx) were exploratively analyzed, processed and tested by the validated statistical method as blinded samples in order to evaluate the performance for the early recognition of patients affected by MPM among asbestos-exposed persons. Good agreement was found between the information obtained by gold-standard diagnostic methods such as computed tomography CT and model output

    Review of low-cost sensors for indoor air quality: Features and applications

    Humans spend the majority of their time indoors, where they are potentially exposed to hazardous pollutants. Within this context, over the past few years, there has been an upsurge of low-cost sensors (LCS) for the measurement of indoor air pollutants, motivated both by recent technological advances and by increased awareness of indoor air quality (IAQ) and its potential negative health impacts. Although not meeting the performance requirements for reference regulatory-equivalent monitoring indoors, LCS can provide informative measurements, offering an opportunity for high-resolution monitoring, emission source identification, exposure mitigation and managing IAQ and energy efficiency, among others. This article discusses the strengths and limitations that LCS offer for applications in the field of IAQ monitoring; it provides an overview of existing sensor technologies and gives recommendations for different indoor applications, considering their performance in the complex indoor environment and discussing future trends
