Rapid Prototyping Using 3-D Welding
Rapid prototyping systems are based, almost exclusively on polymer, or paper materials.
The dimensions of the parts produced are limited by the volume of the processing area within
the machine, and parts tend to warp or distort due to shrinkage and lack of support. Also the
mechanical properties of the part are restricted to those of the processable materials and thus,

Environments for sonic ecologies
This paper outlines a current lack of consideration for the environmental context of Evolutionary Algorithms used for the generation of music. We attempt to readdress this balance by outlining the benefits of developing strong coupling strategies between agent and en- vironment. It goes on to discuss the relationship between artistic process and the viewer and suggests a placement of the viewer and agent in a shared environmental context to facilitate understanding of the artistic process and a feeling of participation in the work. The paper then goes on to outline the installation ‘Excuse Me and how it attempts to achieve a level of Sonic Ecology through the use of a shared environmental context
Application of PCR to a clinical and environmental investigation of a case of equine botulism
PCR for the detection of botulinum neurotoxin gene types A to E was used in the investigation of a case of equine botulism. Samples from a foal diagnosed with toxicoinfectious botulism in 1985 were reanalyzed by PCR and the mouse bioassay in conjunction with an environmental survey. Neurotoxin B was detected by mouse bioassay in culture enrichments of serum, spleen, feces, and intestinal contents. PCR results compared well with mouse bioassay results, detecting type B neurotoxin genes in these samples and also in a liver sample. Other neurotoxin types were not detected by either test. Clostridium botulinum type B was shown to be prevalent in soils collected from the area in which the foal was raised. Four methods were used to test for the presence of botulinum neurotoxin-producing organisms in 66 soil samples taken within a 5-km radius: PCR and agarose gel electrophoresis (types A to E), PCR and an enzyme-linked assay (type B), hybridization of crude alkaline cell lysates with a type B-specific probe, and the mouse bioassay (all types). Fewer soil samples were positive for C. botulinum type B by the mouse bioassay (15%) than by any of the DNA-based detection systems. Hybridization of a type B-specific probe to DNA dot blots (26% of the samples were positive) and PCR-enzyme-linked assay (77% of the samples were positive) were used for the rapid analysis of large numbers of samples, with sensitivity limits of 3 x 10(6) and 3,000 cells, respectively. Conventional detection of PCR products by gel electrophoresis was the most sensitive method (300-cell limit), and in the present environmental survey, neurotoxin B genes only were detected in 94% of the samples
A comparative high-resolution study of predissociation linewidths in the Schumann-Runge bands of O2
Results are presented of a comparative study in which three distinct high-resolution experimental techniques (vacuum-ultraviolet laser spectroscopy, laser-induced fluorescence spectroscopy and vacuum-ultraviolet Fourier-transform spectroscopy) were used to study predissociation in the Schumann-Runge bands of O2 B 3Σ⎯u(v′)←X 3Σ⎯g(v″) with v′=13 and 14. Our measurements are the first to be performed at high resolution for these levels and represent a significant advance on previous knowledge, characterizing completely the fine-structure and rotation dependencies of the B 3Σ⎯u(v=13 and 14)-state predissociation for the first time. The measured fine-structurespecific linewidths will result in significant improvements in the parameterization of models describing predissociation of the B-state and will have an impact on the development of realistic photochemical models of the terrestrial atmosphere. Good agreement was found between linewidths measured using vacuum-ultraviolet laser spectroscopy and laser-induced fluorescence spectroscopy, but unexpected difficulties arose in determining quantitative linewidths using vacuum-ultraviolet Fourier-transform spectroscopy. For each experimental technique, the instrumental resolution had to be carefully controlled and monitored in order to ensure reliable interpretation of the measured spectra.The collaborative aspects of this research have been partially supported by reciprocal U.S.-Australian Cooperative

Validation of the DECAF score to predict hospital mortality in acute exacerbations of COPD
Background
Hospitalisation due to acute
exacerbations of COPD (AECOPD) is common, and
subsequent mortality high. The DECAF score was derived
for accurate prediction of mortality and risk strati
fi
cation
to inform patient care. We aimed to validate the DECAF
score, internally and externally, and to compare its
performance to other predictive tools.
Methods
The study took place in the two hospitals
within the derivation study (internal validation) and in
four additional hospitals (external validation) between
January 2012 and May 2014. Consecutive admissions
were identi
fi
ed by screening admissions and searching
coding records. Admission clinical data, including DECAF
indices, and mortality were recorded. The prognostic
value of DECAF and other scores were assessed by the
area under the receiver operator characteristic (AUROC)
curve.
Results
In the internal and external validation cohorts,
880 and 845 patients were recruited. Mean age was
73.1 (SD 10.3) years, 54.3% were female, and mean
(SD) FEV
1
45.5 (18.3) per cent predicted. Overall
mortality was 7.7%. The DECAF AUROC curve for
inhospital mortality was 0.83 (95% CI 0.78 to 0.87) in
the internal cohort and 0.82 (95% CI 0.77 to 0.87) in
the external cohort, and was superior to other
prognostic scores for inhospital or 30-day mortality.
Conclusions
DECAF is a robust predictor of mortality,
using indices routinely available on admission. Its
generalisability is supported by consistent strong
performance; it can identify low-risk patients (DECAF
0
–
1) potentially suitable for Hospital at Home or early
supported discharge services, and high-risk patients
(DECAF 3
–
6) for escalation planning or appropriate early
palliation.

B-cell dysregulation in Crohn's disease is partially restored with infliximab therapy
Background: B-cell depletion can improve a variety of chronic inflammatory diseases, but does not appear beneficial for patients with Crohn's disease. Objective: To elucidate the involvement of B cells in Crohn's disease, we here performed an 'in depth' analysis of intestinal and blood B-cells in this chronic inflammatory disease. Methods: Patients with Crohn's disease were recruited to study B-cell infiltrates in intestinal biopsies (n = 5), serum immunoglobulin levels and the phenotype and molecular characteristics of blood B-cell subsets (n = 21). The effects of infliximab treatment were studied in 9 patients. Results: Granulomatous tissue showed infiltrates of B lymphocytes rather than Ig-secreting plasma cells. Circulating transitional B cells and CD21low B cells were elevated. IgM memory B cells were reduced and natural effector cells showed decreased replication histories and somatic hypermutation (SHM) levels. In contrast, IgG and IgA memory B cells were normally present and their Ig gene transcripts carried increased SHM levels. The numbers of transitional and natural effector cells were normal in patients who responded clinically well to infliximab. Conclusions: B cells in patients with Crohn's disease showed signs of chronic stimulation with localization to granulomatous tissue and increased molecular maturation of IgA and IgG. Therapy with TNFα-blockers restored the defect in IgM memory B-cell generation and normalized transitional B-cell levels, making these subsets candidate markers for treatment monitoring. Together, these results suggest a chronic, aberrant B-cell response in patients with Crohn's disease, which could be targeted with new therapeutics that specifically regulate B-cell function
Observations of Coronal Mass Ejections with the Coronal Multichannel Polarimeter
The Coronal Multichannel Polarimeter (CoMP) measures not only the
polarization of coronal emission, but also the full radiance profiles of
coronal emission lines. For the first time, CoMP observations provide
high-cadence image sequences of the coronal line intensity, Doppler shift and
line width simultaneously in a large field of view. By studying the Doppler
shift and line width we may explore more of the physical processes of CME
initiation and propagation. Here we identify a list of CMEs observed by CoMP
and present the first results of these observations. Our preliminary analysis
shows that CMEs are usually associated with greatly increased Doppler shift and
enhanced line width. These new observations provide not only valuable
information to constrain CME models and probe various processes during the
initial propagation of CMEs in the low corona, but also offer a possible
cost-effective and low-risk means of space weather monitoring.Comment: 6 figures. Will appear in the special issue of Coronal Magnetism,

Genotype, B-vitamin status, and androgens affect spaceflight-induced ophthalmic changes
Ophthalmic changes have occurred in a subset of astronauts on International Space Station missions. Visual deterioration is considered the greatest human health risk of spaceflight. Affected astronauts exhibit higher concentrations of 1-carbon metabolites (e.g., homocysteine) before flight. We hypothesized that genetic variations in 1-carbon metabolism genes contribute to susceptibility to ophthalmic changes in astronauts. We investigated 5 polymorphisms in the methionine synthase reductase (MTRR), methylenetetrahydrofolate reductase (MTHFR), serine hydroxymethyltransferase (SHMT), and cystathionine β-synthase (CBS) genes and their association with ophthalmic changes after flight in 49 astronauts. The number of G alleles of MTRR 66 and C alleles of SHMT1 1420 both contributed to the odds of visual disturbances. Preflight dehydroepiandrosterone was positively associated with cotton wool spots, and serum testosterone response during flight was associated with refractive change. Block regression showed that B-vitamin status and genetics were significant predictors of many of the ophthalmic outcomes that we observed. In one example, genetics trended toward improving (P = 0.10) and B-vitamin status significantly improved (P < 0.001) the predictive model for refractive change after flight. We document an association between MTRR 66 and SHMT1 1420 polymorphisms and spaceflight-induced vision changes. This line of research could lead to therapeutic options for both space travelers and terrestrial patients
Fertility, Living Arrangements, Care and Mobility
There are four main interconnecting themes around which the contributions in this book are based. This introductory chapter aims to establish the broad context for the chapters that follow by discussing each of the themes. It does so by setting these themes within the overarching demographic challenge of the twenty-first century – demographic ageing. Each chapter is introduced in the context of the specific theme to which it primarily relates and there is a summary of the data sets used by the contributors to illustrate the wide range of cross-sectional and longitudinal data analysed
Charged pion form factor between Q^2=0.60 and 2.45 GeV^2. II. Determination of, and results for, the pion form factor
The charged pion form factor, Fpi(Q^2), is an important quantity which can be
used to advance our knowledge of hadronic structure. However, the extraction of
Fpi from data requires a model of the 1H(e,e'pi+)n reaction, and thus is
inherently model dependent. Therefore, a detailed description of the extraction
of the charged pion form factor from electroproduction data obtained recently
at Jefferson Lab is presented, with particular focus given to the dominant
uncertainties in this procedure. Results for Fpi are presented for
Q^2=0.60-2.45 GeV^2. Above Q^2=1.5 GeV^2, the Fpi values are systematically
below the monopole parameterization that describes the low Q^2 data used to
determine the pion charge radius. The pion form factor can be calculated in a
wide variety of theoretical approaches, and the experimental results are
compared to a number of calculations. This comparison is helpful in
understanding the role of soft versus hard contributions to hadronic structure
