    Effects of Glycol on Leachability and Efficacy of Boron Wood Preservatives

    Although boron has many advantages as a wood preservative, this chemical performs poorly in leaching exposures. In this study, we investigated the potential for decreasing the leachability of boron preservatives with polyethylene glycol (PEG). Southern pine (Pinus spp.) test samples impregnated with combinations of sodium borate or boric acid and PEG were subjected to both leaching and decay tests. Samples treated sequentially with sodium borate or boric acid and then with PEG-400 or PEG-600 showed a significantly increased resistance to boron leaching. However, decay tests indicated that blocks treated with sodium borate or boric acid and PEG experienced slightly higher weight losses at nearly all retention levels, possibly as a result of PEG depletion. Thus, although the results suggest that bulking agents may enhance the resistance of boron to leaching, the enhanced leach resistance may be a temporary effect

    Antioxidant and antimicrobial activities of Morchella conica Pers.

    Antioxidant capacity and antimicrobial activities of Morchella conica Pers. extracts obtained with ethanol were investigated in this study. Four complementary test systems; namely DPPH free radical scavenging, -carotene/linoleic acid systems, total phenolic compounds and total flavonoid concentration were used. Inhibition values of M. conica ethanol extracts, buthylated hydroxyanisol (BHA) and -tocopherol standards were found to be 96.9, 98.9 and 99.2%, respectively, at aconcentration of 160 ìg/ml. When compared the inhibition levels of methanol extract of M. conica and standards in linoleic acid system, it was observed that the higher the concentration of both M. conicaethanol extract and the standards the higher the inhibition effect. Total flavonoid amount was 9.17±0.56ìg mg-1 quercetin equivalent while the phenolic compound amount was 41.93±0.29 ìg mg-1 pyrocatecholequivalent in the ethanolic extract. The antimicrobial effect of M. conica ethanol extract was tested against six species of Gram-positive bacteria, seven species of Gram-negative bacteria and one speciesof yeast. The M. conica ethanol extract had a narrow antibacterial spectrum against tested microorganisms. The most susceptible bacterium was M. flavus. The crude extract was found active on S. aureus ATCC 25923 and S. aureus Cowan I. The M. conica ethanol extract did not exhibit anticandidal activity against C. albican

Four complementary test systems; namely DPPH freeradical scavenging, -carotene/linoleic acid systems, total phenolic compounds and total flavonoid concentration have been used. Inhibition values of R. flava extracts, BHA and -tocopherol standardswere found to be 94.7, 98.9 and 99.2%, respectively, at 160ƒÊg/ml. When compared the inhibition levels of ethanol extract of R. flava and standards in linoleic acid system, it was observed that the higher theconcentration of both RF ethanol extract and the standards the higher the inhibition effect. Total flavonoid amount was 8.27}0.28 ƒÊg mg-1 quercetin equivalent while the total phenolic compound amountwas 39.83}0.32 ƒÊg mg-1 pyrocatechol equivalent in the ethanolic extract. The ethanol extract of R. flava inhibited the growth of Gram-positive bacteria better than Gram-negative bacteria and yeast. The crude extract showed no antibacterial activity against Pseudomonas aeruginosa, Escherichia coli, Morganella morganii and Proteus vulgaris. The antimicrobial activity profile of R. flava against tested strains indicated that Micrococcus flavus, Micrococcus luteus and Yersinia enterocolitica was the most susceptible bacteria of all the test strains. R. flava was found to be inactive against Candida albicans

    BAZI AHLAT (Pyrus L.) TÜRLERİNİN TOHUM ÖZELLİKLERİ VE ÇİMLENDİRME OLANAKLARI ÜZERİNE ARAŞTIRMALAR

    Bu çalışmada, ahlat (Pyrus elaeagnifolia Pall.), yabani armut (P. communis L.) ve badem yapraklı ahlat (P. amygdaliformis Vill.)’ta bazı tohum özellikleri ve farklı soğuk-ıslak katlama işlem kombinasyonları ile değişik ekim zamanlarının bu türlerin tohumlarının çimlenme yüzdelerine olan etkileri incelenmiştir. Tohumlara uygulanan soğuk-ıslak katlama işlem kombinasyonları ile ekim zamanı işlemleri sonucunda elde edilen çimlenme verilerin analizi “SPSS İstatistik Paket Programı”nda değerlendirilmiştir. Yapılan Varyans Analizi ve Duncan Testi sonuçlarına göre; her 3 türde de ekimden önce uygulanan değişik soğuk-ıslak katlama işlem kombinasyonları ile ekim zamanı işlemlerinin çimlenme yüzdeleri bakımından aralarında 0.001 olasılık düzeyinde anlamlı farklılıklar ortaya çıkmıştır. Buna göre; her 3 türde de 9 (2 Kasım 2004 tarihinde doğrudan ekim), 11 (2 Ocak 2005 tarihinde doğrudan ekim) ve 2 (6-7°C sıcaklıkta 2 ay soğuk-katlama 2 Mart tarihinde ekim) nolu ön işlemler çimlenme yüzdesini olumlu yönde en fazla etkileyen ön işlemler olmuştur. Buna karşılık, her 3 türde de tohumun çimlenme yüzdesini en az etkileyen ön işlemler 15 (2 Mayıs 2005 tarihinde doğrudan ekim) ve 14 (2 Nisan 2005 tarihinde doğrudan ekim) nolu ön işlemler olmuştur.     Histopathological placental lesions in mild gestational hyperglycemic and diabetic women

    Objective: To investigate and compare the incidence of histopathological placental lesions in mild gestational hyperglycemia, gestational diabetes and overt diabetes at term and preterm gestation.Research design and methods: One-hundred-and-thirty-one placental samples were collected from Diabetes mellitus (DM) positive screened patients. Two diagnostic tests, glycemic profile and 100 g oral glucose tolerance test (OGTT) in parallel identified 4 groups normoglycemic, mild gestational hyperglycemia (MGH), gestational DM (GDM) or overt DM (DM). Placental tissue specimens and sections from 4 groups were obtained by uniform random sampling and stained with hematoxylin-eosin.Results: Placentas from MGH group presented 17 types of histopathological change and higher rates of syncytial nodes and endarteritis. GDM placentas presented only nine types of histopathological change, high rates of dysmaturity, low rates of calcification and no syncytial nodes. Overt DM placentas showed 22 types of histopathological change, 21 of which were present in the preterm period. There were histopathological similarities between MGH and DM placentas, but the former exhibited a higher incidence of endarteritis, which has been described as a post-mortem phenomenon.Conclusion: Our results confirmed that the distinct placental changes associated with DM and MGH depend on gestational period during which the diabetic insult occurs. It may reasonably be inferred that subclinical maternal hyperglycemia during pregnancy, as showed in MGH group, is responsible for increased placental endarteritis, a postmortem lesion in the live fetus

    Urinary Exosomal microRNA-451-5p Is a Potential Early Biomarker of Diabetic Nephropathy in Rats

    Non-invasive renal signatures can help in serial monitoring of diabetic patients. We tested whether urinary exosomal (UE) microRNA (miR) analysis could non-invasively predict renal pathology in diabetic rats during the course of diabetes. Diabetes mellitus (DM) was induced in male Wistar rats by a single intraperitoneal injection of streptozotocin (STZ, 50 mg/kg body weight). Non-diabetic control (CTRL) rats were injected with vehicle. Insulin (INS) treatment (5U/d, s.c.) was provided to 50% of the DM rats. Urine samples were collected at weeks 3, 6, and 9 following injections and UE prepared. An increase in miR-451-5p and miR-16, observed by pilot small RNA sequencing of UE RNA, was confirmed by quantitative real-time polymerase chain reaction (qPCR) and selected for further study. Subsets of rats were euthanized after 3, 6, and 9 weeks of diabetes for renal pathology analysis, including determination of the tubulointerstitial fibrotic index (TFI) and glomerulosclerotic index (GI) scores. qPCR showed a substantial rise in miR-451-5p in UE from DM rats during thecourse of diabetes, with a significant rise (median fold change >1000) between 3 and 6 weeks. Moreover, UE miR-451-5p at 6 weeks predicted urine albumin at 9 weeks (r = 0.76). A delayed but significant rise was also observed for miR-16. In contrast, mean urine albumin only increased 21% between 3 and 6 weeks (non-significant rise), and renal TFI and GI were unchanged till 9 weeks. Renal expression of miR-451-5p and miR-16 (at 10 weeks) did not correlate with urine levels, and moreover, was negatively associated with indices of renal pathology (r�-0.70, p = 0.005 for TFI and r�-0.6, p�0.02 for GI). Overall, a relative elevation in renal miR-451-5p and miR-16 in diabetes appeared protective against diabetes- induced kidney fibrosis; while UE miR-451-5p may hold prognostic value as an earlyand sensitive non-invasive indicator of renal diseas
