    User-Item Reciprocity in Recommender Systems: Incentivizing the Crowd

    Data consumption has changed significantly in the last 10 years. The digital revolution and the Internet has brought an abundance of information to users. Recommender systems are a popular means of finding content that is both relevant and personalized. However, today’s users require better recommender systems, able of producing continuous data feeds keeping up with their instantaneous and mobile needs. The CrowdRec project addresses this demand by providing context-aware, resource-combining, socially-informed, interactive and scalable recommendations. The key insight of CrowdRec is that, in order to achieve the dense, high-quality, timely information required for such systems, it is necessary to move from passive user data collection, to more active techniques fostering user engagement.     Innovations in Sub-National Government in Europe

    Improvement in government is often rooted in decentralised layers of government. In this article the authors discuss the recent history of innovations in sub-national government in Europe. They focus on two general trends and developments. Cases of interesting practices and developments illustrate the general trends. For each general trend a distinction is made between two kinds of innovations: structural reforms and new ways of working within existing institutional settings. The first trend concerns changing relations between government, civil society and citizens, in response to the increased contestation of the nature of traditional representative democracy. Examples of structural reforms are: increased citizen participation, the expanded use of referenda and the introduction of the directly elected mayor. New ways of working are: forms of coproduction between the public sector and the third sector. The second trend described in this article is change within government itself, mainly the rise of ‘governance’ and changing administrative scales. Structural reforms take several forms: scale enlargement to enhance system capacity, scale reduction to enhance system capacity, scale enlargement to enhance citizen effectiveness, and scale reduction to enhance citizen effectiveness. Next to them, new ways of working can be observed within governments: cross-boarder co-operations, inter-municipal co-operations, and new forms of performance assessment and fiscal management. The authors conclude the article with a few challenges to both academics and governments.La mejora del gobierno se encuentra arraigada con frecuencia en los niveles descentralizados. En este artículo es el cambio dentro del gobierno mismo, principalmente la emergencia de la “gobernanza” y el cambio en el tamaño administrativo. Las reformas estructurales adoptan formas diversas: el aumento del tamaño para mejorar la capacidad del sistema, el aumento del tamaño para favorecer la participación ciudadana, y la reducción del tamaño para favorecer asimismo la participación ciudadana. Además, se advierten nuevas formas de acción dentro de los mismos gobiernos: cooperación transfronteriza, cooperación intermunicipal y nuevas formas de evaluación del rendimiento y de la gestión financiera.     Examining How Middle School Science Teachers Implement a Multimedia-enriched Problem-based Learning Environment

    This study examined how a group of ten middle school teachers implemented a technology enriched problem-based learning (PBL) environment. The goal was to understand their motivation, document their implementation techniques, and identify factors that teachers considered important in using technology-based PBL tools in their teaching. The analysis identified four factors that provided the impetus for teachers to consider the adoption of technology-based PBL instruction. These factors are (1) the PBL program addresses the teachers’ curricular needs and implementing it has campus administrative and technical support, (2) the method is aligned with teachers’ pedagogical beliefs, (3) the PBL program offers a new way of teaching and promotes the development of higher-order thinking skills, and (4) the PBL program challenges students in a captivating manner and supports the learning needs of all students. Teachers’ implementation techniques with over 1,000 sixth graders were documented in detail with regard to: 1) the teacher’s roles, 2) the student’s role, and 3) the classroom interactions during the implementation of the PBL program. In addition, a detailed description of contrasting narratives of two pairs of teachers is provided, illustrating the range of implementation techniques that can occur using the same PBL program to allow for individualized instruction to meet different students’ needs. The goal of providing detailed implementation practices is to address the lack of “how to” in PBL implementation in K-12 classrooms as indicated in the literature and offer insights and ideas to those interested in adopting and implementing PBL.     Peripheral decarboxylase inhibitors paradoxically induce aromatic L-amino acid decarboxylase

    Peripheral decarboxylase inhibitors (PDIs) prevent the conversion of levodopa to dopamine in the blood by the enzyme aromatic L-amino acid decarboxylase (AADC). Alterations in enzyme activity may contribute to the required higher dosages of levodopa observed in many patients with Parkinson’s disease. We evaluated the effect of levodopa/PDI use on serum AADC enzyme activity. Serum AADC enzyme activity was evaluated in three independent cohorts of patients with Parkinson’s disease or parkinsonism (n = 301) and compared between patients on levodopa/PDI vs. patients not on this medication. AADC enzyme activity was elevated in 62% of patients on levodopa/PDI treatment, compared to 19% of patients not on levodopa/PDI (median 90 mU/L vs. 50 mU/L, p < 0.001). Patients with elevated AADC activity had longer disease duration and higher doses of levodopa/PDI. These findings may implicate that peripheral AADC induction could underlie a waning effect of levodopa, necessitating dose increases to maintain a sustained therapeutic effect

    Tyrosine hydroxylase deficiency: a treatable disorder of brain catecholamine biosynthesis

    Tyrosine hydroxylase deficiency is an autosomal recessive disorder resulting from cerebral catecholamine deficiency. Tyrosine hydroxylase deficiency has been reported in fewer than 40 patients worldwide. To recapitulate all available evidence on clinical phenotypes and rational diagnostic and therapeutic approaches for this devastating, but treatable, neurometabolic disorder, we studied 36 patients with tyrosine hydroxylase deficiency and reviewed the literature. Based on the presenting neurological features, tyrosine hydroxylase deficiency can be divided in two phenotypes: an infantile onset, progressive, hypokinetic-rigid syndrome with dystonia (type A), and a complex encephalopathy with neonatal onset (type B). Decreased cerebrospinal fluid concentrations of homovanillic acid and 3-methoxy-4-hydroxyphenylethylene glycol, with normal 5-hydroxyindoleacetic acid cerebrospinal fluid concentrations, are the biochemical hallmark of tyrosine hydroxylase deficiency. The homovanillic acid concentrations and homovanillic acid/5-hydroxyindoleacetic acid ratio in cerebrospinal fluid correlate with the severity of the phenotype. Tyrosine hydroxylase deficiency is almost exclusively caused by missense mutations in the TH gene and its promoter region, suggesting that mutations with more deleterious effects on the protein are incompatible with life. Genotype-phenotype correlations do not exist for the common c.698G>A and c.707T>C mutations. Carriership of at least one promotor mutation, however, apparently predicts type A tyrosine hydroxylase deficiency.
