    Seabed morphology and bottom water masses related to benthic habitats at the Cristóbal Colón diapir (NW of the Guadalquivir ridge, Gulf of Cádiz)

    The seabed morphology and the sub-superficial characteristics of the Cristóbal Colón diapir located on the continental slope of the Gulf of Cadiz have been analysed from data obtained in the framework of the LIFE+INDEMARES/CHICA project. The aim of this study is to recognize the morphological features and the geological processes generated by the bottom water masses and their influence on the habitats and associated benthic communities. The NACW affects the generation of morphological features on the summit, revealing that different oceanographic conditions favoured the carbonate mound growth in the past. The interface between the NACW and the MOW sweeps the bottom from the SE to the NW and the presence of benthic communities dominated by filter feeders on the contouritic drift indicates that this current is strong enough to favour the availability of nutrients and organic particles and to develop both the contouritic deposits on the SE flank and the moats on the N and W flanks.Versión del edito

    Biodiversity Assessment and Geographical Affinities of Discards in Clam Fisheries in the Atlantic–Mediterranean Transition (Northern Alboran Sea)

    This study focused on the assessment and quantification of discards generated by clam fisheries along the northern Alboran Sea (western Mediterranean). Discard samples (n = 278) were collected throughout one year on board nine commercial vessels. A total of 129 species were identified, mostly represented by molluscs (72 spp.), arthropods (20 spp.) and echinoderms (12 spp.). Molluscs dominated in terms of abundance (67.5%) and biomass (94.2%). The superfamily Paguroidea (i.e. hermit crabs), together with undersized target individuals, were the most abundant taxa. The abundance and biomass of discards displayed significant maximum values in winter, which could be partly related to biotic factors including population dynamics of some dominant species. Multivariate analyses indicated the presence of different assemblages related to the targeted bivalve species, reflecting the transition between a fine surface-sands biocoenosis exposed to wave action and a well-sorted fine sands biocoenosis below 5 m depth. Analysis of biogeographical affinities showed that most discarded species (73.2%) have an extensive Atlantic range, whereas 7.1% have a restricted distribution within the Mediterranean. The presence of subtropical species highlights the uniqueness of this area (the Atlantic–Mediterranean transition) in European seas.     A surface plasmon resonance based approach for measuring response to pneumococcal vaccine

    Incidence of pneumococcal disease has increased worldwide in recent years. Response to pneumococcal vaccine is usually measured using the multiserotype enzyme-linked immunosorbent assay (ELISA) pneumococcal test. However, this approach presents several limitations. Therefore, the introduction of new and more robust analytical approaches able to provide information on the efficacy of the pneumococcal vaccine would be very beneficial for the clinical management of patients. Surface plasmon resonance (SPR) has been shown to offer a valuable understanding of vaccines' properties over the last years. The aim of this study is to evaluate the reliability of SPR for the anti-pneumococcal capsular polysaccharides (anti-PnPs) IgGs quantification in vaccinated. Fast protein liquid chromatography (FPLC) was used for the isolation of total IgGs from serum samples of vaccinated patients. Binding-SPR assays were performed to study the interaction between anti-PnPs IgGs and PCV13. A robust correlation was found between serum levels of anti-PnPs IgGs, measured by ELISA, and the SPR signal. Moreover, it was possible to correctly classify patients into "non-responder", "responder" and "high-responder" groups according to their specific SPR PCV13 response profiles. SPR technology provides a valuable tool for reliably characterize the interaction between anti-PnPs IgGs and PCV13 in a very short experimental time

    Assessment of a New ROS1 Immunohistochemistry Clone (SP384) for the Identification of ROS1 Rearrangements in Patients with Non–Small Cell Lung Carcinoma: the ROSING Study

    Introduction: The ROS1 gene rearrangement has become an important biomarker in NSCLC. The College of American Pathologists/International Association for the Study of Lung Cancer/Association for Molecular Pathology testing guidelines support the use of ROS1 immunohistochemistry (IHC) as a screening test, followed by confirmation with fluorescence in situ hybridization (FISH) or a molecular test in all positive results. We have evaluated a novel anti-ROS1 IHC antibody (SP384) in a large multicenter series to obtain real-world data. Methods: A total of 43 ROS1 FISH-positive and 193 ROS1 FISH-negative NSCLC samples were studied. All specimens were screened by using two antibodies (clone D4D6 from Cell Signaling Technology and clone SP384 from Ventana Medical Systems), and the different interpretation criteria were compared with break-apart FISH (Vysis). FISH-positive samples were also analyzed with next-generation sequencing (Oncomine Dx Target Test Panel, Thermo Fisher Scientific). Results: An H-score of 150 or higher or the presence of at least 70% of tumor cells with an intensity of staining of 2+ or higher by the SP384 clone was the optimal cutoff value (both with 93% sensitivity and 100% specificity). The D4D6 clone showed similar results, with an H-score of at least 100 (91% sensitivity and 100% specificity). ROS1 expression in normal lung was more frequent with use of the SP384 clone (p < 0.0001). The ezrin gene (EZR)-ROS1 variant was associated with membranous staining and an isolated green signal FISH pattern (p = 0.001 and p = 0.017, respectively). Conclusions: The new SP384 ROS1 IHC clone showed excellent sensitivity without compromising specificity, so it is another excellent analytical option for the proposed testing algorithm
