    Computer literacy in UK education: an evolving strategy

    For the past three decades, the United Kingdom (UK) has been searching for the best methods of providing computer literacy and competence for elementary and secondary pupils. Initially, schools developed computer studies as a separate area of study instead of promoting the use of computers across the curriculum, an approach supported by the Micro-electronics Education Programme (MEP). The debate between these two approaches involved parents, pupils, and employers as well as MEP experts and inservice teacher educators. In 1987, upon realizing that the bulk of information technology resources in schools had been used only for students 16 years or older who were taking General Certificate of Secondary Education (GCSE) classes, the National Curriculum demanded an information technology dimension in each of the core subjects and a cross-curricular approach to information technology competence. Despite the good intentions of such educational changes, it remains to be seen if this new strategy can succeed in its aim to make information technology competence a natural part of every pupil's repertoire

    Simulate_PCR for amplicon prediction and annotation from multiplex, degenerate primers and probes

    BACKGROUND: Pairing up primers to amplify desired targets and avoid undesired cross reactions can be a combinatorial challenge. Effective prediction of specificity and inclusivity from multiplexed primers and TaqMan®/Luminex® probes is a critical step in PCR design. RESULTS: Code is described to identify all primer and probe combinations from a list of unpaired, unordered candidates that should produce a product. It predicts and extracts all amplicon sequences in a large sequence database from a list of primers and probes, allowing degenerate bases and user-specified levels of primer-target mismatch tolerance. Amplicons hit by TaqMan®/Luminex® probes are indicated, and products may be annotated with gene information from NCBI. Fragment length distributions are calculated to predict electrophoretic gel banding patterns. CONCLUSIONS: Simulate_PCR is the only freely available software that can be run from the command line for high throughput applications which can calculate all products from large lists of primers and probes compared to a large sequence database such as nt. It requires no prior knowledge of how primers should be paired. Degenerate bases are allowed and entire amplicon sequences are extracted and annotated with gene information. Examples are provided for sets of TaqMan®/Luminex® PCR signatures predicted to amplify all HIV-1 genomes, all Coronaviridae genomes, and a group of antibiotic resistance genes.     Using guitar learning to probe the Action Observation Network's response to visuomotor familiarity

    Watching other people move elicits engagement of a collection of sensorimotor brain regions collectively termed the Action Observation Network (AON). An extensive literature documents more robust AON responses when observing or executing familiar compared to unfamiliar actions, as well as a positive correlation between amplitude of AON response and an observer's familiarity with an observed or executed movement. On the other hand, emerging evidence shows patterns of AON activity counter to these findings, whereby in some circumstances, unfamiliar actions lead to greater AON engagement than familiar actions. In an attempt to reconcile these conflicting findings, some have proposed that the relationship between AON response amplitude and action familiarity is nonlinear in nature. In the present study, we used an elaborate guitar training intervention to probe the relationship between movement familiarity and AON engagement during action execution and action observation tasks. Participants underwent fMRI scanning while executing one set of guitar sequences with a scanner-compatible bass guitar and observing a second set of sequences. Participants then acquired further physical practice or observational experience with half of these stimuli outside the scanner across 3 days. Participants then returned for an identical scanning session, wherein they executed and observed equal numbers of familiar (trained) and unfamiliar (untrained) guitar sequences. Via region of interest analyses, we extracted activity within AON regions engaged during both scanning sessions, and then fit linear, quadratic and cubic regression models to these data. The data best support the cubic regression models, suggesting that the response profile within key sensorimotor brain regions associated with the AON respond to action familiarity in a nonlinear manner. Moreover, by probing the subjective nature of the prediction error signal, we show results consistent with a predictive coding account of AON engagement during action observation and execution that also takes into account effects of changes in neural efficiency

    “Getting Turned On”: Using ICT Training To Promote Active Ageing In New York City

    The purpose of this mixed-method study was to examine how participation in a senior’s technology training program influences the social environments and active ageing of older New Yorkers. Findings demonstrate increased and sustained use, improved ability and confidence with computer and Internet technology, and a substantial and positive effect on social connectedness, access to information, and social and civic participation among participants. Authors conclude with a discussion of how comprehensive, community-based ICT training programs (such as OATS) can support the ongoing engagement and re-engagement of older adults within society by building, maintaining and restoring their place within their various communities

    In May 2014, the first planting of the Royal Botanic Garden Edinburgh (RBGE) conservation hedge took place, when the Reverend Anne Brennan planted a tree which had originated as a cutting from the ancient and historic European yew, Taxus baccata, in the churchyard of her church at Fortingall, Perthshire. This is one of almost 2,000 plants that will eventually form a conservation hedge of significant scientific and conservation value. The International Conifer Conservation Programme (ICCP), based at RBGE, has actively sought other opportunities to establish conservation hedges via its network of ‘safe sites’, using a range of different conifer species. This initiative is being driven by the potential for relatively large numbers of genotypes from a single threatened species to be stored in a linear space. It is well established that seed banks have a great capacity to store large amounts of genetic diversity, so we should simply consider conservation hedges in a similar manner. These super-hedges cram relatively large amounts of genetic material into a small space, capturing a great range of wild traits and potentially contributing to the restoration of wild populations. To date, conservation hedges have been planted at five separate locations at RBGE’s Edinburgh Garden as well as at four ICCP external ‘safe sites’. Although this article focuses on the establishment of conservation hedges using conifers, we have also highlighted some conservation hedges that comprise non-coniferous species

    A study of the antecedents and consequences of psychological ownership in organizational settings

    Psychological ownership is a feeling of possession in the absence of any formal or legal claims of ownership. In this study, the authors aimed to extend previous empirical testing of psychological ownership in work settings to encompass both job-based and organization-based psychological ownership as well as related work attitudes and behavioral outcomes. Questionnaire data from 68 employees and their managers revealed that job-based psychological ownership and organization-based psychological ownership are distinct work attitudes that are distinguishable from job satisfaction and organizational commitment. Psychological ownership predicted job satisfaction and organizational commitment and mediated the relationship between autonomy and these work attitudes. There was no support for a relationship between psychological ownership and behavioral outcomes. The authors discuss the limitations of the study and the implications of psychological ownership

    Blue cone monochromacy: causative mutations and associated phenotypes.

    PurposeTo perform a phenotypic assessment of members of three British families with blue cone monochromatism (BCM), and to determine the underlying molecular genetic basis of disease.MethodsAffected members of three British families with BCM were examined clinically and underwent detailed electrophysiological and psychophysical testing. Blood samples were taken for DNA extraction. Molecular analysis involved the amplification of the coding regions of the long (L) and medium (M) wave cone opsin genes and the upstream locus control region (LCR) by polymerase chain reaction (PCR). Gene products were directly sequenced and analyzed.ResultsIn all three families, genetic analysis identified that the underlying cause of BCM involved an unequal crossover within the opsin gene array, with an inactivating mutation. Family 1 had a single 5'-L-M-3' hybrid gene, with an inactivating Cys203Arg (C203R) mutation. Family 3 had an array composed of a C203R inactivated 5'-L-M-3' hybrid gene followed by a second inactive gene. Families 1 and 3 had typical clinical, electrophysiological, and psychophysical findings consistent with stationary BCM. A novel mutation was detected in Family 2 that had a single hybrid gene lacking exon 2. This family presented clinical and psychophysical evidence of a slowly progressive phenotype.ConclusionsTwo of the BCM-causing family genotypes identified in this study comprised different hybrid genes, each of which contained the commonly described C203R inactivating mutation. The genotype in the family with evidence of a slowly progressive phenotype represents a novel BCM mutation. The deleted exon 2 in this family is not predicted to result in a shift in the reading frame, therefore we hypothesize that an abnormal opsin protein product may accumulate and lead to cone cell loss over time. This is the first report of slow progression associated with this class of mutation in the L or M opsin genes in BCM
