    Evaluation of the immunomodulatory activities of the probiotic strain lactobacillus fermentum UCO-979C

    Lactobacillus fermentum UCO-979C, a strain isolated from a human stomach, was previously characterized by its potential probiotic properties. The UCO-979C strain displayed the ability to beneficially regulate the innate immune response triggered by Helicobacter pylori infection in human gastric epithelial cells. In this work, we conducted further in vitro studies in intestinal epithelial cells (IECs) and in vivo experiments in mice in order to characterize the potential immunomodulatory effects of L. fermentum UCO-979C on the intestinal mucosa. Results demonstrated that the UCO-979C strain is capable to differentially modulate the immune response of IECs triggered by Toll-like receptor 4 (TLR4) activation through the modulation of TLR negative regulators' expression. In addition, we demonstrated for the first time that L. fermentum UCO-979C is able to exert its immunomodulatory effect in the intestinal mucosa in vivo. The feeding of mice with L. fermentum UCO-979C significantly increased the production of intestinal IFN-γ, stimulated intestinal and peritoneal macrophages and increased the number of Peyer's patches CD4+ T cells. In addition, L. fermentum UCO-979C augmented intestinal IL-6, reduced the number of immature B220+CD24high B cells from Peyer's patches, enhanced the number of mature B B220+CD24low cells, and significantly increased intestinal IgA content. The results of this work revealed that L. fermentum UCO-979C has several characteristics making it an excellent candidate for the development of immunobiotic functional foods aimed to differentially regulate immune responses against gastric and intestinal pathogens.Fil: Garcia Castillo, Valeria.     An eBPF-XDP hardware-based network slicing architecture for future 6G front- to back-haul networks

    The heterogeneous requirements imposed by different vertical businesses have motivated a networking paradigm shift in the next generation of mobile networks (beyond 5G and 6G), leading to critical operation competitiveness of improved productivity, performance and efficiency. Furthermore, with the global digital revolution, such as Industry 4.0, and a connected world, network virtualisation together with high reliability and high performance communications have become crucial elements for mobile network operators. To minimise the negative effects that could affect critical services, network slicing is widely recognised as a key technology with the objective of meeting the Service-Level Agreements (SLAs) and Key Performance Indicators (KPIs) in future 6G networks. In this context, it is essential to introduce a programmable data plane able to enforce flexible Quality of Service (QoS) commitments, while providing high-performance packet processing and real-time monitoring capabilities. To this end, this paper is focused on designing, prototyping and evaluating a novel framework that leverages a set of hardware-based technologies including eXpress Data Path (XDP), extended Barkeley Packet Filter (eBPF) and Smart Network Interface Cards (SmartNICs) to offload network functionality with the objective of providing high-performance pre-6G front-, mid-and back-haul network communications and thus, decreasing the overhead incurs by the Linux Kernel. The proposed solution is implemented based on bypassing the Linux Kernel and accelerating the communication, while providing network slice control and real-time monitoring capabilities. The main aim of this framework is to ensure network communications in forthcoming 6G infrastructures by guaranteeing 6G KPIs and avoiding system overload.     5G-QoE:QoE Modelling for Ultra-HD Video Streaming in 5G Networks

    Traffic on future fifth-generation (5G) mobile networks is predicted to be dominated by challenging video applications such as mobile broadcasting, remote surgery and augmented reality, demanding real-time, and ultra-high quality delivery. Two of the main expectations of 5G networks are that they will be able to handle ultra-high-definition (UHD) video streaming and that they will deliver services that meet the requirements of the end user's perceived quality by adopting quality of experience (QoE) aware network management approaches. This paper proposes a 5G-QoE framework to address the QoE modeling for UHD video flows in 5G networks. Particularly, it focuses on providing a QoE prediction model that is both sufficiently accurate and of low enough complexity to be employed as a continuous real-time indicator of the 'health' of video application flows at the scale required in future 5G networks. The model has been developed and implemented as part of the EU 5G PPP SELFNET autonomic management framework, where it provides a primary indicator of the likely perceptual quality of UHD video application flows traversing a realistic multi-tenanted 5G mobile edge network testbed. The proposed 5G-QoE framework has been implemented in the 5G testbed, and the high accuracy of QoE prediction has been validated through comparing the predicted QoE values with not only subjective testing results but also empirical measurements in the testbed. As such, 5G-QoE would enable a holistic video flow self-optimisation system employing the cutting-edge Scalable H.265 video encoding to transmit UHD video applications in a QoE-aware manner

    Effectiveness of a healthy lifestyle promotion program as adjunctive teletherapy for treatment-resistant major depression during COVID 19 pandemic: A randomized clinical trial protocol

    INTRODUCTION: Treatment-resistant depression (TRD) has a high prevalence and can be exacerbated by poor physical health and economic hardships, which have become common stressors during the current COVID-19 pandemic. The therapeutic approaches used to treat these patients are not always available, may be not be accepted by some patients, and often require face-to-face interactions. OBJECTIVE: The main aim of this study will be to evaluate the effectiveness of an Internet-based adjuvant lifestyle-based intervention for patients with TRD. METHODS: This will be a parallel, randomized, and controlled clinical trial. A total of 180 patients with TRD will be randomly allocated (1:1:1) to 1 of 3 groups: treatment prescribed by the mental health team and written suggestions for lifestyle changes (placebo control group); treatment prescribed by the mental health team, written suggestions for lifestyle changes, and an 8-week mindfulness-based cognitive therapy program (active control group); or treatment prescribed by the mental health team, written suggestions for lifestyle changes, and an 8-week lifestyle change promotion program (intervention group). We will perform this study during the COVID-19 pandemic, and will administer interventions by teletherapy, and contact participants by telephone calls, text messages, and/or teleconferences. We will collect patient data using questionnaires administered at baseline, immediately after the intervention, and after 6 and 12 months. The primary outcome will be score on the Beck Depression Inventory-II. The secondary outcomes will be score on the Clinical Global Impressions Scale (used to quantify and track patient progress and treatment response over time) and health-related quality of life measured using the European Quality of Life-5 Dimensions Questionnaire. DISCUSSION: Patients with TRD are especially vulnerable when face-to-face psychotherapy is unavailable. The main strength of the proposed study is the novelty of the intervention to be used as an adjuvant therapy. Our results may provide guidance for treatment of patients with TRD in future situations that require lockdown measures. CLINICALTRIALS REGISTRATION NUMBER: NCT04428099

    Sex and gender perspectives in colorectal cancer

As a consequence, the impact of sex and gender on both tumour biology and clinical outcomes has been largely underestimated. Although interrelated and often used interchangeably, sex and gender are not equivalent concepts. Sex is a biological attribute that defines species according to their chromosomal makeup and reproductive organ, while gender refers to a chosen sexual identity. Sex dimorphisms are rarely taken into account, in either preclinical or clinical research, with inadequate analysis of differences in outcomes according to sex or gender still widespread, reflecting a gap in our knowledge for a large proportion of the target population. Underestimation of sex-based differences in study design and analyses has invariably led to 'one-drug' treatment regimens for both males and females. For patients with colorectal cancer (CRC), sex also has an impact on the disease incidence, clinicopathological features, therapeutic outcomes, and tolerability to anticancer treatments. Although the global incidence of CRC is higher in male subjects, the proportion of patients presenting right-sided tumours and BRAF mutations is higher among females. Concerning sex-related differences in treatment efficacy and toxicity, drug dosage does not take into account sex-specific differences in pharmacokinetics. Toxicity associated with fluoropyrimidines, targeted therapies, and immunotherapies has been reported to be more extensive for females with CRC than for males, although evidence about differences in efficacy is more controversial. This article aims to provide an overview of the research achieved so far into sex and gender differences in cancer and summarize the growing body of literature illustrating the sex and gender perspective in CRC and their impact in relation to tumour biology and treatment efficacy and toxicity. We propose endorsing research on how biological sex and gender influence CRC as an added value for precision oncology
