Exploring Young Womenâs Interest in Fluid Power with Workshop Experiences
Fluid power is not an engineering topic usually presented to students in grades K-12. However undergraduate students in various programs are required to learn this concept. Evidence exists that indicates college students are more easily engaged when fluid power has been introduced earlier in their schooling. Thus, a variety of organizations are beginning work toward providing programs to creatively present the topic to younger students. After an examination of existing literature, we found minimal evidence of this type of work having been carried out and reported on. This study examines data from workshops designed to introduce fluid power to middle and high school students. These workshops introduced the use of fluid power components along with relevant theory. Students were queried after the workshop and after to assess interest/enjoyment, motivation, and interest in the workshop materials. The results presented in this paper are useful for future workshop designers and presenters. We share specific information regarding student interest, timing, and type of materials that result in higher levels of interest along with the quality of presentation and demonstration materials. Also providing guidance on how gender makeup of groups during the workshop influenced these results. Further research opportunities are presented as well as suggestions on how to continue to increase interest, enjoyment, and value in fluid power as this may be applicable other areas of STEM
A Single Tri-Epitopic Antibody Virtually Recapitulates the Potency of a Combination of Three Monoclonal Antibodies in Neutralization of Botulinum Neurotoxin Serotype A.
The standard of treatment for botulism, equine antitoxin, is a foreign protein with associated safety issues and a short serum half-life which excludes its use as a prophylactic antitoxin and makes it a less-than-optimal therapeutic. Due to these limitations, a recombinant monoclonal antibody (mAb) product is preferable. It has been shown that combining three mAbs that bind non-overlapping epitopes leads to highly potent botulinum neurotoxin (BoNT) neutralization. Recently, a triple human antibody combination for BoNT/A has demonstrated potent toxin neutralization in mouse models with no serious adverse events when tested in a Phase I clinical trial. However, a triple antibody therapeutic poses unique development and manufacturing challenges. Thus, potentially to streamline development of BoNT antitoxins, we sought to achieve the potency of multiple mAb combinations in a single IgG-based molecule that has a long serum half-life. The design, production, and testing of a single tri-epitopic IgG1-based mAb (TeAb) containing the binding sites of each of the three parental BoNT/A mAbs yielded an antibody of nearly equal potency to the combination. The approach taken here could be applied to the design and creation of other multivalent antibodies that could be used for a variety of applications, including toxin elimination
A Three Monoclonal Antibody Combination Potently Neutralizes Multiple Botulinum Neurotoxin Serotype E Subtypes.
Human botulism is most commonly caused by botulinum neurotoxin (BoNT) serotypes A, B, and E. For this work, we sought to develop a human monoclonal antibody (mAb)-based antitoxin capable of binding and neutralizing multiple subtypes of BoNT/E. Libraries of yeast-displayed single chain Fv (scFv) antibodies were created from the heavy and light chain variable region genes of humans immunized with pentavalent-toxoid- and BoNT/E-binding scFv isolated by Fluorescence-Activated Cell Sorting (FACS). A total of 10 scFv were isolated that bound one or more BoNT/E subtypes with nanomolar-level equilibrium dissociation constants (KD). By diversifying the V-regions of the lead mAbs and selecting for cross-reactivity, we generated three scFv that bound all four BoNT/E subtypes tested at three non-overlapping epitopes. The scFvs were converted to IgG that had KD values for the different BoNT/E subtypes ranging from 9.7 nM to 2.28 pM. An equimolar combination of the three mAbs was able to potently neutralize BoNT/E1, BoNT/E3, and BoNT/E4 in a mouse neutralization assay. The mAbs have potential utility as therapeutics and as diagnostics capable of recognizing multiple BoNT/E subtypes. A derivative of the three-antibody combination (NTM-1633) is in pre-clinical development with an investigational new drug (IND) application filing expected in 2018
The effects of moderate prenatal alcohol exposure on the organization of exploratory behavior by adult female rats
A large body of research has indicated that moderate Prenatal Alcohol Exposure (PAE - 60 mg/dl peak BAC) can produce impairments on cognitive processes, such as spatial learning and memory. The neurobiological basis of these impairments is poorly understood but is frequently linked to alterations in hippocampal functioning. Although the hippocampus has a central role in learning and memory, damage to the hippocampus can also produce impairments in locomotor and exploratory behaviors by rodents (reviewed in Thompson et al., 2018). Rodent exploratory behavior is organized around home bases, which serves as central points of attraction from which rats and mice organize their exploratory trips into the remaining environment. We are unaware of studies investigating the relationship between moderate prenatal alcohol exposure and exploratory behavior. Thus, in the present study, we tested adult female rats in two experiments where they were allowed to freely explore a circular open field for ~30min. Here, we report that, with respect to home base behavior, exploratory movements were largely intact in rats exposed to alcohol prenatally. Specifically, animals rapidly established home bases within 5min of the beginning of testing and the bases were established in one or two locations in the environment. They made exploratory trips away and back to the home base and made several stops at this location along with bouts of grooming behavior. The results are discussed with respect to theories of hippocampal function and the impact of moderate prenatal alcohol on spatial behavior
Health Vulnerability Model for Latinx Sexual and Gender Minorities: Typologies with Socioeconomic Stability, Health Care Access, and Social Characteristics Indicators
Vulnerability can undermine positive health outcomes and challenge healthcare services access. However, to date, vulnerable populations research has been limited by overly broad definitions, lack of clear indicators, and failure to explore subtypes of vulnerability. Informed by literature and theory, this analysis used a specific operationalization of health vulnerability to identify typologies among a sample of Latinx sexual and gender minorities. We analyzed baseline data from Latinx sexual and gender minorities (N = 186) recruited for a community-based HIV intervention. We performed latent class analysis to operationalize vulnerability using eight socioeconomic stability, health care access, and social characteristics indicators. We identified three typologies of vulnerability from our sample: Low Education and High Social Support (63.4% of sample), High Education and Year-round Employment (18.8%), and High Education and High Discrimination (17.7%). Using specific indicators produced more nuanced vulnerability typologies which, after further testing, can assist in informing tailored health promotion interventions
Evaluation with in vivo optical coherence tomography and histology of the vascular effects of the everolimus-eluting bioresorbable vascular scaffold at two years following implantation in a healthy porcine coronary artery model: implications of pilot results for future pre-clinical studies
To quantify with in vivo OCT and histology, the device/vessel interaction after implantation of the bioresorbable vascular scaffold (BVS). We evaluated the area and thickness of the strut voids previously occupied by the polymeric struts, and the neointimal hyperplasia (NIH) area covering the endoluminal surface of the strut voids (NIHEV), as well as the NIH area occupying the space between the strut voids (NIHBV), in healthy porcine coronary arteries at 2, 3 and 4 years after implantation of the device. Twenty-two polymeric BVS were implanted in the coronary arteries of 11 healthy Yucatan minipigs that underwent OCT at 2, 3 and 4 years after implantation, immediately followed by euthanasia. The areas and thicknesses of 60 corresponding strut voids previously occupied by the polymeric struts and the size of 60 corresponding NIHEV and 49 NIHBV were evaluated with both OCT and histology by 2 independent observers, using a single quantitative analysis software for both techniques. At 3 and 4 years after implantation, the strut voids were no longer detectable by OCT or histology due to complete polymer resorption. However, analysis performed at 2 years still provided clear delineation of these structures, by both techniques. The median [ranges] areas of these strut voids were 0.04 [0.03â0.16] and 0.02 [0.01â0.07] mm2 by histology and OCT, respectively. The mean (±SD) thickness by histology and OCT was 220 ± 40 and 120 ± 20 Όm, respectively. The median [ranges] NIHEV by histology and OCT was 0.07 [0.04â0.20] and 0.03 [0.01â0.08] mm2, while the mean (±SD) NIHBV by histology and OCT was 0.13 ± 0.07 and 0.10 ± 0.06 mm2. Our study indicates that in vivo OCT of the BVS provides correlated measurements of the same order of magnitude as histomorphometry, and is reproducible for the evaluation of certain vascular and device-related characteristics. However, histology systematically gives larger values for all the measured structures compared to OCT, at 2 years post implantation

Cancer Informatics for Cancer Centers (CI4CC): Building a Community Focused on Sharing Ideas and Best Practices to Improve Cancer Care and Patient Outcomes.
Cancer Informatics for Cancer Centers (CI4CC) is a grassroots, nonprofit 501c3 organization intended to provide a focused national forum for engagement of senior cancer informatics leaders, primarily aimed at academic cancer centers anywhere in the world but with a special emphasis on the 70 National Cancer Institute-funded cancer centers. Although each of the participating cancer centers is structured differently, and leaders' titles vary, we know firsthand there are similarities in both the issues we face and the solutions we achieve. As a consortium, we have initiated a dedicated listserv, an open-initiatives program, and targeted biannual face-to-face meetings. These meetings are a place to review our priorities and initiatives, providing a forum for discussion of the strategic and pragmatic issues we, as informatics leaders, individually face at our respective institutions and cancer centers. Here we provide a brief history of the CI4CC organization and meeting highlights from the latest CI4CC meeting that took place in Napa, California from October 14-16, 2019. The focus of this meeting was "intersections between informatics, data science, and population science." We conclude with a discussion on "hot topics" on the horizon for cancer informatics
Sodium and Oxygen Abundances in the Open Cluster NGC 6791 from APOGEE H-Band Spectroscopy
The open cluster NGC 6791 is among the oldest, most massive and metal-rich
open clusters in the Galaxy. High-resolution -band spectra from the Apache
Point Observatory Galactic Evolution Experiment (APOGEE) of 11 red giants in
NGC 6791 are analyzed for their chemical abundances of iron, oxygen, and
sodium. The abundances of these three elements are found to be homogeneous
(with abundance dispersions at the level of 0.05 - 0.07 dex) in these
cluster red giants, which span much of the red-giant branch (T
3500K - 4600K), and include two red-clump giants. From the infrared
spectra, this cluster is confirmed to be among the most metal-rich clusters in
the Galaxy ( = 0.34 0.06), and is found to have a roughly
solar value of [O/Fe] and slightly enhanced [Na/Fe]. Non-LTE calculations for
the studied Na I lines in the APOGEE spectral region (16373.86\AA\ and
16388.85\AA) indicate only small departures from LTE ( 0.04 dex)
for the parameter range and metallicity of the studied stars. The previously
reported double population of cluster members with different Na abundances is
