Analysis of Synaptic Proteins in the Cerebrospinal Fluid as a New Tool in the Study of Inborn Errors of Neurotransmission
Abstract In a few rare diseases, specialised studies in cerebrospinal fluid (CSF) are required to identify the underlying metabolic disorder. We aimed to explore the
possibility of detecting key synaptic proteins in the CSF, in particular dopaminergic and gabaergic, as new procedures
that could be useful for both pathophysiological and diagnostic purposes in investigation of inherited disorders
of neurotransmission. Dopamine receptor type 2 (D2R), dopamine transporter (DAT) and vesicular monoamine transporter type 2 (VMAT2) were analysed in CSF samplesfrom 30 healthy controls (11 days to 17 years) by western blot analysis. Because VMAT2 was the only protein with intracellular localisation, and in order to compare results, GABA vesicular transporter, which is another intracellular protein, was also studied. Spearmanâs correlation and
Studentâs t tests were applied to compare optical density signals between different proteins. All these synaptic proteins could be easily detected and quantified in the
CSF. DAT, D2R and GABA VT expression decrease with age, particularly in the first months of life, reflecting the expected intense synaptic activity and neuronal circuitry formation. A statistically significant relationship was found
between D2R and DAT expression, reinforcing the previous evidence of DAT regulation by D2R. To our knowledge, there are no previous studies on human CSF reporting a reliable
analysis of these proteins. These kinds of studies could help elucidate new causes of disturbed dopaminergic and gabaergic
transmission as well as understanding different responses to L-dopa in inherited disorders affecting dopamine metabolism.
Moreover, this approach to synaptic activity in vivo can be extended to different groups of proteins and diseases
In Silico Detection and Typing of Plasmids using PlasmidFinder and Plasmid Multilocus Sequence Typing
In the work presented here, we designed and developed two easy-to-use Web tools for in silico detection and characterization of whole-genome sequence (WGS) and whole-plasmid sequence data from members of the family Enterobacteriaceae. These tools will facilitate bacterial typing based on draft genomes of multidrug-resistant Enterobacteriaceae species by the rapid detection of known plasmid types. Replicon sequences from 559 fully sequenced plasmids associated with the family Enterobacteriaceae in the NCBI nucleotide database were collected to build a consensus database for integration into a Web tool called PlasmidFinder that can be used for replicon sequence analysis of raw, contig group, or completely assembled and closed plasmid sequencing data. The PlasmidFinder database currently consists of 116 replicon sequences that match with at least at 80% nucleotide identity all replicon sequences identified in the 559 fully sequenced plasmids. For plasmid multilocus sequence typing (pMLST) analysis, a database that is updated weekly was generated from www.pubmlst.org and integrated into a Web tool called pMLST. Both databases were evaluated using draft genomes from a collection of Salmonella enterica serovar Typhimurium isolates. PlasmidFinder identified a total of 103 replicons and between zero and five different plasmid replicons within each of 49 S. Typhimurium draft genomes tested. The pMLST Web tool was able to subtype genomic sequencing data of plasmids, revealing both known plasmid sequence types (STs) and new alleles and ST variants. In conclusion, testing of the two Web tools using both fully assembled plasmid sequences and WGS-generated draft genomes showed them to be able to detect a broad variety of plasmids that are often associated with antimicrobial resistance in clinically relevant bacterial pathogens. Stability of Spatial Optical Solitons
We present a brief overview of the basic concepts of the soliton stability
theory and discuss some characteristic examples of the instability-induced
soliton dynamics, in application to spatial optical solitons described by the
NLS-type nonlinear models and their generalizations. In particular, we
demonstrate that the soliton internal modes are responsible for the appearance
of the soliton instability, and outline an analytical approach based on a
multi-scale asymptotic technique that allows to analyze the soliton dynamics
near the marginal stability point. We also discuss some results of the rigorous
linear stability analysis of fundamental solitary waves and nonlinear impurity
modes. Finally, we demonstrate that multi-hump vector solitary waves may become
stable in some nonlinear models, and discuss the examples of stable
(1+1)-dimensional composite solitons and (2+1)-dimensional dipole-mode solitons

Formation and Propagation of Matter Wave Soliton Trains
Attraction between atoms in a Bose-Einstein-Condensate renders the condensate
unstable to collapse. Confinement in an atom trap, however, can stabilize the
condensate for a limited number of atoms, as was observed with 7Li, but beyond
this number, the condensate collapses. Attractive condensates constrained to
one-dimensional motion are predicted to form stable solitons for which the
attractive interactions exactly compensate for the wave packet dispersion. Here
we report the formation or bright solitons of 7Li atoms created in a quasi-1D
optical trap. The solitons are created from a stable Bose-Einstein condensate
by magnetically tuning the interactions from repulsive to attractive. We
observe a soliton train, containing many solitons. The solitons are set in
motion by offsetting the optical potential and are observed to propagate in the
potential for many oscillatory cycles without spreading. Repulsive interactions
between neighboring solitons are inferred from their motion
Atom--Molecule Coherence in a Bose-Einstein Condensate
Coherent coupling between atoms and molecules in a Bose-Einstein condensate
(BEC) has been observed. Oscillations between atomic and molecular states were
excited by sudden changes in the magnetic field near a Feshbach resonance and
persisted for many periods of the oscillation. The oscillation frequency was
measured over a large range of magnetic fields and is in excellent quantitative
agreement with the energy difference between the colliding atom threshold
energy and the energy of the bound molecular state. This agreement indicates
that we have created a quantum superposition of atoms and diatomic molecules,

Maximal Spontaneous Photon Emission and Energy Loss from Free Electrons
Free electron radiation such as Cerenkov, Smith--Purcell, and transition
radiation can be greatly affected by structured optical environments, as has
been demonstrated in a variety of polaritonic, photonic-crystal, and
metamaterial systems. However, the amount of radiation that can ultimately be
extracted from free electrons near an arbitrary material structure has remained
elusive. Here we derive a fundamental upper limit to the spontaneous photon
emission and energy loss of free electrons, regardless of geometry, which
illuminates the effects of material properties and electron velocities. We
obtain experimental evidence for our theory with quantitative measurements of
Smith--Purcell radiation. Our framework allows us to make two predictions. One
is a new regime of radiation operation---at subwavelength separations, slower
(nonrelativistic) electrons can achieve stronger radiation than fast
(relativistic) electrons. The second is a divergence of the emission
probability in the limit of lossless materials. We further reveal that such
divergences can be approached by coupling free electrons to photonic bound
states in the continuum (BICs). Our findings suggest that compact and efficient
free-electron radiation sources from microwaves to the soft X-ray regime may be
achievable without requiring ultrahigh accelerating voltages.Comment: 7 pages, 4 figure
Urban Movement and Alcohol Intake Strongly Predict Defaulting from Tuberculosis Treatment: An Operational Study
BACKGROUND: High levels of defaulting from treatment challenge tuberculosis control in many African cities. We assessed defaulting from tuberculosis treatment in an African urban setting. METHODS: An observational study among adult patients with smear-positive pulmonary tuberculosis receiving treatment at urban primary care clinics in Kampala, Uganda. Defaulting was defined as having missed two consecutive monthly clinic visits while not being reported to have died or continued treatment elsewhere. Defaulting patients were actively followed-up and interviewed. We assessed proportions of patients abandoning treatment with and without the information obtained through active follow-up and we examined associated factors through multivariable logistic regression. RESULTS: Between April 2007 and April 2008, 270 adults aged â„15 years were included; 54 patients (20%) were recorded as treatment defaulters. On active follow-up vital status was established of 28/54 (52%) patients. Of these, 19 (68%) had completely stopped treatment, one (4%) had died and eight (29%) had continued treatment elsewhere. Extrapolating this to all defaulters meant that 14% rather than 20% of all patients had truly abandoned treatment. Daily consumption of alcohol, recorded at the start of treatment, predicted defaulting (adjusted odds ratio [OR(adj)] 4.4, 95%CI 1.8-13.5), as did change of residence during treatment (OR(adj) 8.7, 95%CI 1.8-41.5); 32% of patients abandoning treatment had changed residence. CONCLUSIONS: A high proportion of tuberculosis patients in primary care clinics in Kampala abandon treatment. Assessing change of residence during scheduled clinic appointments may serve as an early warning signal that the patient may default and needs adherence counseling
Living biointerfaces based on non-pathogenic bacteria to direct cell differentiation
Genetically modified Lactococcus lactis, non-pathogenic bacteria expressing the FNIII7-10 fibronectin fragment as a protein membrane have been used to create a living biointerface between synthetic materials and mammalian cells. This FNIII7-10 fragment comprises the RGD and PHSRN sequences of fibronectin to bind α5ÎČ1 integrins and triggers signalling for cell adhesion, spreading and differentiation. We used L. lactis strain to colonize material surfaces and produce stable biofilms presenting the FNIII7-10 fragment readily available to cells. Biofilm density is easily tunable and remains stable for several days. Murine C2C12 myoblasts seeded over mature biofilms undergo bipolar alignment and form differentiated myotubes, a process triggered by the FNIII7-10 fragment. This biointerface based on living bacteria can be further modified to express any desired biochemical signal, establishing a new paradigm in biomaterial surface functionalisation for biomedical applications
Mexican World Heritage information on the web: Institutional presence and visibility
This study offers a global overview of the presence and visibility of web information on UNESCO World Heritage located in Mexico, via the analysis of official websites and Web 2.0 information. Cultural heritage is a
determining factor in linking people to their history, and contributes to increasing cultural tourism and economic development. The study starts from the hypothesis that the design of these has an influence on the dissemination
and popularity of the aforementioned heritage. The relationships between the administrative organization of the country and Internet protocols are compared. A webometric study of the official Mexican websites was carried
out. An evaluation sheet was designed to allow the assessment of aspects relating to identification, presence, accessibility and content. The multilingual nature of this information and its presence on social networks
and Wikipedia was analysed. The analysis of URLs confirms that the domain .mx is used in 84% of cases. The results indicate a noticeable use of Web 2.0 dissemination of the heritage assets on YouTube (51%) and Facebook (40%),
followed by 23% on Twitter. The Web Accessibility Initiative (WAI) guidelines are not yet frequently applied.
