Microbiota and body weight control: weight watchers within?
Despite several decades of research, managing body weight remains an unsolved clinical problem. Health problems associated with dysregulated body weight, such as obesity and cachexia, exhibit several gut microbiota alterations. There is an increased interest in utilising the gut microbiota for body weight control, as it responds to intervention and plays an important role in energy extraction from food, as well as biotransformation of nutrients.
This review provides an overview of the role of the gut microbiota in the physiological and metabolic alterations observed in two body weight dysregulation-related disorders, namely obesity and cachexia. Second, we assess the available evidence for different strategies, including caloric restriction, intermittent fasting, ketogenic diet, bariatric surgery, probiotics, prebiotics, synbiotics, high-fibre diet, and fermented foods - effects on body weight and gut microbiota composition. This approach was used to give insights into the possible link between body weight control and gut microbiota configuration.
Despite extensive associations between body weight and gut microbiota composition, limited success could be achieved in the translation of microbiota-related interventions for body weight control in humans. Manipulation of the gut microbiota alone is insufficient to alter body weight and future research is needed with a combination of strategies to enhance the effects of lifestyle interventions
Efectos conductuales inducidos por drogas de abuso psicoestimulantes en ratas a distintas edades durante el desarrollo: Papel de la neurogénesis hipocampal
[spa] La literatura científica sugiere que cuanto más temprano es el inicio en el
consumo de drogas más probable es la progresión hacia el abuso y la adicción. Los
psicoestimulantes, como la cocaína o los derivados anfetamínicos (MDMA,
metanfetamina y d-anfetamina), son unas de las drogas más consumidas a nivel
mundial, iniciándose su consumo durante la adolescencia. Numerosos estudios han
demostrado diferencias en los efectos inducidos por las drogas de abuso dependiendo de
la edad en la que se inicia el consumo. La adolescencia está considerada como una etapa
de cambios tanto a nivel comportamental como molecular, haciendo que sea un periodo
de especial susceptibilidad a los efectos neuroquímicos producidos por las drogas de
abuso. Durante la adolescencia se ha observado un incremento de los efectos
placenteros inducidos por las drogas y una disminución de los efectos negativos durante
la abstinencia, pudiendo ser una de las principales causas que potencian su consumo.
Además, datos epidemiológicos han puesto de manifiesto que la adolescencia supone un
factor de riesgo en el desarrollo de futuras comorbilidades psiquiátricas (adiccióndepresión) asociadas a desregulaciones en marcadores de palasticidad (p. ej.,
neurogénesis hipocampal).
En este contexto, en la presente Tesis Doctoral estudió (1) las neuroadaptaciones
inducidas durante la adolescencia en ratas después de la administración de diferentes
psicoestimulantes (cocaína, MDMA, metanfetamina y d-anfetamina) para identificar los
periodos de mayor vulnerabilidad a los efectos neurotóxicos (p. ej. neurogénesis
hipocampal y marcadores de neuroplasticidad), (2) los efectos negativos inmediatos y a
largo plazo (p. ej., durante la abstinencia forzada) inducidos por la cocaína o
metanfetamina admsinitrada en el periodo de mayor vulnerabilidad adolescente; y (3)
los posibles efectos beneficiosos del tratamiento con el shock electroconvulsivo para
mejorar las consecuencias negativas a largo plazo a nivel comportamental y molecular
inducidas por la administración de metanfetamina durante el periodo de vulnerabilidad
adolescente.
Para llevar a cabo estos objetivos, se utilizaron ratas macho adolescentes de la
cepa Sprague-Dawley. Además, se realizaron pruebas comportamentales para evaluar el
fenotipo prodepresivo (test de natación forzada, campo abierto y test de consumo
voluntario de sacarosa) y proadictivo (test de consumo voluntario de droga) inducidos por la cocaína o metanfetamina. Finalmente, se utilizó una unidad de terapia
electroconvulsiva para evaluar las mejoras a nivel comportamental y molecular
causadas por el shock electroconvulsivo tras la administración de metanfetamina.
Los resultados obtenidos en el primer objetivo demostraron un efecto
neurotóxico diferencial inducido por las drogas de estudio (cocaína vs. derivados
anfetamínicos) dependiendo de la edad de administración. Por un lado, se observó una
mayor vulnerabilidad durante la adolescencia temprana-media (PND 33-39) tras la
administración de cocaína en dos regiones cerebrales. La cocaína desreguló los niveles
del marcador multifuncional FADD (Fas-Associated Death Domain) a nivel
hipocampal (véase Artículo I) y de los receptores cannabinoides en la corteza prefrontal
(véase Artículo II). Por otro lado, se observó una mayor vulnerabilidad durante la
adolescencia tardía (PND 54-57) tras la administración de los derivados anfetamínicos
(véase Artículos III y IV). Durante la adolescencia tardía estas drogas disminuyeron la
proliferación y supervivencia celular en el giro dentado del hipocampo (véase Artículo
IV), mostrando cierto grado de insensibilidad o una mejor adaptación a los efectos
negativos inducidos por los derivados anfetamínicos durante los primeros estadios de la
adolescencia. El segundo objetivo demostró un efecto duradero de las consecuencias
negativas inducidas por la administración de cocaína y metanfetamina durante sus
respectivos periodos de vulnerabilidad adolecente. En particular, la administración de
cocaína durante la adolescencia temprana-media (PND 33-39) indujo un incremento a
largo plazo del desespero conductual (tras una reexposición aguda a la cocaína, PND
71) y de la anhedonia (PND 76-77), asociado a una disminución de la expresión de cFos en el estriado dorsal (véase Artículo V). Por otro lado, la administración de
metanfetamina durante la adolescencia tardía (PND 54-57) y la etapa adulta incrementó
los efectos negativos tras un largo periodo de abstinencia. Los principales resultados
demostraron un incremento del fenotipo prodepresivo (anhedonia) y proadictivos
(incremento del consumo voluntario de metanfetamina) (véase Artículo VI) y una
disminución de los marcadores para la neurogénesis (véase Artículo VII). Estos
resultados sugieren que la administración de ambas drogas (cocaína o metanfetamina)
administrada durante los periodos de vulnerabilidad adolescente aumenta la
susceptibilidad a sufrir comorbilidades psiquiátricas y adaptaciones neuronales que
persisten hasta la etapa adulta, pudiendo ser en parte responsables de las recaídas al
consumo (véase Artículo VI, incremento en el consumo voluntario de metanfetamina). El tercer objetivo demostró que la aplicación del shock electroconvulsivo como
tratamiento (véase Anexo Experimental I) mejoró las consecuencias negativas inducidas
tras la administración de metanfetamina. En concreto, los resultados indicaron que ratas
tratadas con metanfetamina durante la adolescencia tardía (PND 54-57) y que fueron
reexpuestas a la droga durante la etapa adulta (PND 91-95) no mostraron el esperado
efecto antidepresivo inducido por el shock electroconvulsivo. En cambio, el tratamiento
con el shock electroconvulsivo previno los efectos neurotóxicos inducidos por la
metanfetamina sobre la neurogénesis hipocampal, así como sobre el sistema
dopaminérgico en el estriado. Estos resultados sugieren un potencial terapéutico de este
tratamiento sobre el posible desarrollo de comorbilidades psiquiátricas y en la recaída al
consumo durante la abstinencia prolongada a esta droga psicoestimulante.
En conclusión, los resultados mostrados en la presente Tesis Doctoral ponen de
manifiesto que la adolescencia es un periodo de gran vulnerabilidad al consumo de los
psicoestimulantes y que la administración de dichas sustancias durante esta etapa
vulnerable conlleva al desarrollo de futuras comorbilidades psiquiátricas en la etapa
adulta (incremento del fenotipo prodepresivo y proadictivo) asociado a desregulaciones
en marcadores de neuroplasticidad (p. ej., neurogénesis hipocampal) que pueden
mejorar tras la aplicación de tratamientos no farmacológicos como el shock
electroconvulsivo.[cat] La literatura científica suggereix que quan abans s’inicia el consum de les
drogues més probable és la progressió cap a l’abús i l’addicció. Els psicoestimulants,
com la cocaïna o els derivats amfetamínics (MDMA, metamfetamina o d-amfetamina),
són uns dels tipus de drogues més consumides a nivell mundial, iniciant-se el seu
consum durant l’etapa adolescent. Nombrosos estudis han demostrat una diferència en
els efectes induïts per les drogues d’abús depenent de l’edat en la que s’inicia el
consum. L’adolescència està considerada com un període de canvis a nivell
comportamental i molecular, fent que sigui especialment susceptible als efectes
neuroquímics produïts per les drogues d’abús. Durant l’adolescència hi ha un increment
dels efectes plaents induïts per les drogues i una disminució dels efectes negatius
observats durant l’abstinència, podent ser una de les principals causes potenciadores del
consum. A més, dades epidemiològiques han posat de manifest que l’adolescència
suposa un factor de risc en el desenvolupament de futures comorbiditats psiquiàtriques
(adicció-depressió) associades a desregulacions en marcadors de plasticitat (p. ex.,
neurogènesi hipocampal).
En aquest context, la present Tesi Doctoral va estudiar (1) les neuroadaptacions
induïdes durant l’adolescència a rates després de l’administració de diferents
psicoestimulants (cocaïna, MDMA, metamfetamina i d-amfetamina) per identificar el
períodes de major vulnerabilitat als efectes neurotòxics (p. ex., neurogènesi hipocampal
i marcadors de neuroplasticitat), (2) els efectes negatius immediats i a llarg termini (p.
ex., durant l’abstinència forçada) induïts per la cocaïna o la metamfetamina
administrada durant el període de major vulnerabilitat adolescent; i (3) el possible efecte
beneficiós del tractament amb el xoc electroconvulsiu per millorar les conseqüències
negatives a llarg termini a nivell comportamental i molecular induïdes per
l’administració de metamfetamina durant el període de vulnerabilitat adolescent.
Per aconseguir aquests objectius, es va utilitzar un model de rata mascle
adolescent del cep Sprague-Dawley. A més, es van realitzar proves comportamental per
evaluar el fenotip prodepressiu (test de natació forçada, camp obert i test de consum
voluntari de sacarosa) i proadictiu (test de consum voluntari de droga) induïts per la
cocaïna o metamfetamina. Finalment, es va utilitzar una unitat de teràpia
electroconvulsiva per avaluar les millores a nivell comportamental i molecular causades
per el xoc electroconvulsiu després de l’administració de metamfetamina Els resultats al primer objectiu van demostrar un efecte neurotòxic diferencial
induït per les drogues d’estudi (cocaïna vs. derivats amfetamínics) depenent de l’edat
d’administració. Per una banda, es va observar una major vulnerabilitat durant
l’adolescència primerenca-mitjana (dia post natal, PND 33-39) després de
l’administració de cocaïna a dues regions cerebrals diferents. La cocaïna va desregular
el marcador multifuncional FADD (Fas-Associated Death Domain) a nivell hipocampal
(vegeu Article I) i els receptors cannabinoides a l’escorça prefrontal (vegeu Article II).
Per altra banda, es va observar una major vulnerabilitat durant l’adolescència tardana
(PND 54-57) després de l’administració del derivats amfetamínics (vegeu Articles III i
IV). Durant l’adolescència tardana aquestes drogues van disminuir la proliferació i
supervivència cel·lular en el gir dentat de l’hipocamp (vegeu Article IV). Aquests
resultats van demostrar cert grau de d’insensibilitat o una millor adaptació als efectes
negatius induïts pels derivats amfetamínics durant el primer estadis de l’adolescència.
El segon objectiu va mostrar un efecte persistent de les conseqüències negatives
induides per l’administració de cocaïna i metamfetamina durant els seus respectius
períodes de vulnerabilitat adolescent. En particular, l’administració de cocaïna durant
l’adolescència primerenca-mitjana (PND 33-39) va induir un increment del desesper
conductual (després de la reexposició aguda a la cocaïna, PND 71) i de l’anhedonia
(PND 76-77), associat a una disminució de l’expressió de c-Fos a l’estriat dorsal (vegeu
Article V). D’altre banda, l’administració de metamfetamina durant l’adolescència
tardana (PND 54-57) i l’etapa adulta va incrementar els efectes negatius després d’un
llarg període d’abstinència. Els principals resultat van mostrar un increment del fenotip
prodepressiu (anhedonia) i proadictiu (increment del consum voluntari de
metanfetamina) (vegeu Article VI) i una disminució de marcadors per a la neurogènesi
(vegeu Article VII). Aquests resultats suggereixen que ambdues drogues (cocaïna i
metamfetamina) administrades durant els períodes de vulnerabilitat adolescent
augmenten la susceptibilitat a sofrir comorbiditats psiquiàtriques i adaptacions
neuronals que persisteixen fins l’etapa adulta, podent ser en part responsables de les
recaigudes al consum (vegeu Article VI, increment del consum voluntari de
metamfetamina). El tercer objectiu va demostrar que l’aplicació del xoc
electroconvulsiu com a tractament (vegeu Annex Experimental I) millora les
conseqüències negatives induïdes per l’administració de metamfetamina. En concret, els
resultats van indicar que rates tractades amb metamfetamina durant l’adolescència
tardana (PND 54-57) que varen ser reexposades a la droga durant l’etapa adulta (PND 91-95) no mostraren l’esperat efecte antidepressiu produït pel xoc electroconvulsiu. En
canvi, el tractament amb el xoc electroconvulsiu va prevenir els efectes neurotòxics
induïts per la metamfetamina sobre la neurogènesi hipocampal, així com sobre el
sistema dopaminèrgic en l’estriat. Aquests resultats suggereixen un potencial terapèutic
d’aquest tractament sobre el possible desenvolupament de comorbiditats psiquiàtriques i
en la recaiguda al consum durant l’abstinència prolongada a aquesta droga
psicoestimulant.
En conclusió, els resultats mostrats a la present Tesi Doctoral posen de manifest
que l’adolescència és un període de vulnerabilitat al consum dels psicoestimulants i que
l’administració d’aquestes substàncies durant l’etapa vulnerable comporta el
desenvolupament de futures comorbiditats psiquiàtriques en l’etapa adulta (increment
del fenotip prodepressiu i proadictiu) associat a desregulacions en marcadors de
neuroplasticitat (p. ex., neurogènesis hipocampal) que poden millorar després de
l’aplicació de tractaments no farmacològics com el xoc electroconvulsiu.[eng] The literature suggests that early drug use is a key factor for later progression to
abuse and addiction. Psychostimulants, such as cocaine or amphetamine-like
compounds (MDMA, methamphetamine or d-amphetamine), are one of the most widely
drugs consumed worldwide, beginning their consumption during adolescence.
Numerous studies have shown a difference in the effects induced by drugs of abuse
depending on the age of drug initiation. Adolescence is a period of great change at the
behavioral and molecular levels, which is more susceptible to the neurochemical effects
of specifics drugs of abuse. A potential cause of early drug consumption during
adolescence could be explained by the observed increase in the pleasant effects and the
decrease in the negative effects during abstinence induced by these drugs. Moreover,
epidemiological data show that adolescence is a risk factor in the development of future
psychiatric comorbidities (addiction-depression) associated with the dysregulation of
neuroplasticity markers (i. e., hippocampal neurogenesis).
In this context, the present Doctoral Thesis studied in rats: (1) whether there was
a vulnerable age period during adolescence in which different psychostimulants
(cocaine, MDMA, methamphetamine and d-amphetamine) induced neurotoxic effects
(i. e., hippocampal neurogenesis and neuroplasticity markers); (2) the immediate and
prolonged negative effects (i. e., emerging during forced abstinence) induced by cocaine
or methamphetamine when administered during the vulnerable period; and (3) the
possible beneficial effects of an electroconvulsive shock treatment on improving the
long-term negative behavioral and molecular consequences caused by
methamphetamine administration during the vulnerable period.
To achieve these objectives, adolescent male Sprague-Dawley rats were used.
Different behavioral tests were performed to determine the pro-depressive (forced-swim
test, open field, voluntary sucrose consumption) and pro-addictive (voluntary drug
consumption) phenotypes induced by cocaine or methamphetamine. Finally, an
electroconvulsive therapy unit was used to improve the negative behavioral and
molecular consequences induced by methamphetamine administration.
The results from the first objective demonstrated a differential neurotoxic effect
induced by cocaine vs. amphetamine-like compounds depending on the age period of
adolescence administration. On the one hand, a greater vulnerability period was observed during early-mid adolescence (post-natal day, PND 33-39) after cocaine
administration in two cerebral regions. Cocaine dysregulated the multimodal protein
FADD (Fas-Associated Death Domain) in the hippocampus (see Article I) and the
cannabinoid receptors (CB1 and CB2) in the prefrontal cortex (see Article II). On the
other hand, a greater vulnerability period was observed during late adolescence (PND
54-57) by amphetamine-like drugs (MDMA, methamphetamine and d-amphetamine)
(see Article III and IV). During late adolescence, these drugs decreased cell
proliferation and cell survival in the dentate gyrus of the hippocampus (see Article IV).
These data demonstrate certain degree of insensitivity or a better adaptation to the
negative effects induced by these amphetamine-like compounds during the early stages
of adolescence. The second objective showed persistent negative consequences after
cocaine and methamphetamine administration during their respective windows of
adolescent vulnerability. In particular, cocaine administration (PND 33-39) induced an
increase in behavioral despair (after an acute cocaine re-exposure on PND 71) and
anhedonia (PND 76-77), associated with a decrease in the expression of c-Fos in the
dorsal striatum (see Article V). On the other hand, methamphetamine administration
during late adolescence (PND 54-57) or adulthood exerted negative effects during
abstinence such as the observed increase in pro-depressive (anhedonia) and proaddictive (increase voluntary methamphetamine consumption) phenotypes (see Article
VI) and the decrease in neurogenic markers (see Article VII). Taken together, both
drugs (cocaine and methamphetamine) when administered during the vulnerable period
increased the susceptibility to develop psychiatric comorbidities and neuronal
adaptations that persist into adulthood, which could be responsible for relapse (see
Article VI, increase of the voluntary methamphetamine consumption). The third
objective demonstrated that an electroconvulsive shock treatment (see Experimental
Annex I for technical characterization) improved the long-term consequences induced
by methamphetamine. In particular, the results indicated that rats treated with a history
of methamphetamine administration during late adolescence (PND 54-57) and later reexposed to the drug in adulthood (PND 91-95) did not show the expected
antidepressant-like effects induced by the electroconvulsive shock treatment (see
Experimental Annex II). Moreover, the neurotoxic effects induced by
methamphetamine on hippocampal neurogenesis, as well as on the dopaminergic system
in the striatum, were prevented by the electroconvulsive shock. These results suggest a potential therapeutic effect for this treatment on preventing the development of
psychiatric comorbidities and on relapse during prolonged abstinence.
In conclusion, the data presented in this Doctoral Thesis demonstrate that
adolescence is a period of great vulnerability to the negative effects induced by early
psychostimulant consumption. Moreover, the administration of cocaine and/or
methamphetamine during adolescence leads to the development of psychiatric
comorbidities in adulthood (increased pro-depressive and pro-addictive phenotypes)
associated with dysregulations of neuroplasticity markers (i. e., hippocampal
neurogenesis). The negative effects induced by adolescent methamphetamine later on in
adulthood are partly improved by a therapeutic treatment such as electroconvulsive
shock
Associations between mental health, alcohol consumption and drinking motives during COVID-19 second lockdown in Ireland.
AIMS
The novel coronavirus pandemic (COVID-19) has impacted the lives of people worldwide since March 2020. Social restrictions aimed at flattening the curve may be associated with an increase in mental health problems and have raised concerns regarding their effect on alcohol consumption. The objective of this study was to characterize changes in alcohol use during lockdown in Ireland and associations with drinking motives and psychopathological symptoms.
METHODS
We collected data from 713 adults (aged 18-60) during the second lockdown period (October/December 2020). By means of an online survey, participants self-reported their alcohol use before COVID and during lockdown. Motives to drink and psychopathological symptoms were also recorded.
RESULTS
Our findings showed that 66% decreased their alcohol consumption, while 15% increased their alcohol consumption. An older age and coping motives were the strongest predictors of increased alcohol use during lockdown. Depression and hostility were the specific psychopathological dimensions associated with drinking to cope.
CONCLUSIONS
Older adults who drink to cope-mainly with depression symptomatology-are an important at-risk population, in line with predictions from alcohol self-medication frameworks. Future research is needed to incorporate strategies into the public mental health ecosystem
Microbiota-gut-brain axis as a regulator of reward processes.
Our gut harbours trillions of microorganisms essential for the maintenance of homeostasis and host physiology in health and disease. In the last decade, there has been a growing interest in understanding the bidirectional pathway of communication between our microbiota and the central nervous system. With regard to reward processes there is accumulating evidence from both animal and human studies that this axis may be a key factor in gating reward valence. Focusing on the mesocorticolimbic pathway, we will discuss how the intestinal microbiota is involved in regulating brain reward functions, both in natural (i.e. eating, social or sexual behaviours) and non-natural reinforcers (drug addiction behaviours including those relevant to alcohol, psychostimulants, opioids and cannabinoids). We will integrate preclinical and clinical evidence suggesting that the microbiota-gut-brain axis could be implicated in the development of disorders associated with alterations in the reward system and how it may be targeted as a promising therapeutic strategy
Evaluating the daily modulation of <scp>FADD</scp> and related molecular markers in different brain regions in male rats
Fas-Associated protein with Death Domain (FADD), a key molecule controlling cell fate by balancing apoptotic versus non-apoptotic functions, is dysregulated in post-mortem brains of subjects with psychopathologies, in animal models capturing certain aspects of these disorders, and by several pharmacological agents. Since persistent disruptions in normal functioning of daily rhythms are linked with these conditions, oscillations over time of key biomarkers, such as FADD, could play a crucial role in balancing the clinical outcome. Therefore, we characterized the 24-h regulation of FADD (and linked molecular partners: p-ERK/t-ERK ratio, Cdk-5, p35/p25, cell proliferation) in key brain regions for FADD regulation (prefrontal cortex, striatum, hippocampus). Samples were collected during Zeitgeber time (ZT) 2, ZT5, ZT8, ZT11, ZT14, ZT17, ZT20, and ZT23 (ZT0, lights-on or inactive period; ZT12, lights-off or active period). FADD showed similar daily fluctuations in all regions analyzed, with higher values during lights off, and opposite to p-ERK/t-ERK ratios regulation. Both Cdk-5 and p35 remained stable and did not change across ZT. However, p25 increased during lights off, but exclusively in striatum. Finally, no 24-h modulation was observed for hippocampal cell proliferation, although higher values were present during lights off. These results demonstrated a clear daily modulation of FADD in several key brain regions, with a more prominent regulation during the active time of rats, and suggested a key role for FADD, and molecular partners, in the normal physiological functioning of the brain's daily rhythmicity, which if disrupted might participate in the development of certain pathologies
