    Quantification of Total Phenolic and Carotenoid Content in Blackberries (Rubus Fructicosus L.) Using Near Infrared Spectroscopy (NIRS) and Multivariate Analysis

    A rapid method to quantify the total phenolic content (TPC) and total carotenoid content (TCC) in blackberries using near infrared spectroscopy (NIRS) was carried out aiming to provide reductions in analysis time and cost for the food industry. A total of 106 samples were analysed using the Folin-Ciocalteu method for TPC and a method based on Ultraviolet-Visible Spectrometer for TCC. The average contents found for TPC and TCC were 24.27 mg·g−1 dw and 8.30 µg·g−1 dw, respectively. Modified partial least squares (MPLS) regression was used for obtaining the calibration models of these compounds. The RPD (ratio of the standard deviation of the reference data to the standard error of prediction (SEP)) values from external validation for both TPC and TCC were between 1.5 < RPDp < 2.5 and RER values (ratio of the range in the reference data to SEP) were 5.92 for TPC and 8.63 for TCC. These values showed that both equations were suitable for screening purposes. MPLS loading plots showed a high contribution of sugars, chlorophyll, lipids and cellulose in the modelling of prediction equations

    Trend analysis in tobacco use in Andalusia (2011-2014) and factors related to use

    Nowadays, smoking is the single greatest preventable cause of death in the world and tobacco is the most prevalent drug ever used. Considering that many people begin smoking in early adolescence, it is essential to further research this matter in order to better understand this behavior as well as enact adequate prevention and health-promotion programs. Despite data confirming a decrease in tobacco consumption in the last few years, it is still necessary to pay more attention to factors that may be related to adolescent smoking. Regarding these factors, a relationship between age and gender in relation to tobacco use has been fully established, showing higher use among older adolescents and females. Furthermore, parental smoking has shown to be a common catalyst for smoking initiation during adolescence. However, results are not completely consistent regarding this association. Some studies suggest that close friends’ smoking provides a substantial source of modeling of experimentation with tobacco, even more powerful than parents’ smoking. The current study seeks to identify trends in smoking behaviour (2011 – 2014) and investigates the relationship between age, gender, parental smoking and best friend’s smoking, and tobacco use among Andalusian adolescent students. The sample for this study is composed of 17,662 students aged 11 to 18 years old: 15,684 in 2011 (7,822 girls) and 1,978 in 2014 (962 girls). In both cases, the data are representative of Andalusian adolescent students. We used some items of the tobacco consumption questionnaire designed by the international team of Health Behaviour in School-aged Children study (HBSC): adolescent current tobacco consumption, tobacco use of best friend, mother and father. Data confirm a decrease in adolescent tobacco use between 2011 and 2014. Females smoke more than males in 2011 and 2014, although gender differences diminish in 2014. Moreover, age, gender and close people’s smoking explain a higher percentage of variance in 2011 than in 2014. Results are discussed in terms of the importance of monitoring tobacco use in adolescence in order to adjust intervention policies. In this respect, even though this study confirms a decline in consumption, this may be attributable to the application of anti-smoking laws or the implementation of prevention programs, as well as a change in recent years in factors related to us

    Occurrence of melatonin and indolic compounds derived from L-tryptophan yeast metabolism in fermented wort and commercial beers

    Melatonin and serotonin are bioactive compounds present in foods and beverages and related to neuroprotection and anti-angiogenesis, among other activities. They have been described in wines and the role of yeast in their formation is clear. Thus, this study evaluates the content of these bioactives and other related indolic compounds in beer. For this purpose, commercial beers were analyzed by a validated UHPLC-HRMS method and sample treatment optimized due to the low concentrations expected. Moreover, a wort was fermented with different commercial beer yeast (Abbaye, Diamond, SafAle, SafLager) in order to monitor the formation of these bioactives during the elaboration process. Results show that indolic compounds such as N-acetylserotonin and 3-indoleacetic acid are produced during the alcoholic fermentation of wort. Moreover, the occurrence of four indolic compounds (5-hydroxytryptophan, N-acetylserotonin, 3-indoleacetic acid, L-tryptophan ethyl ester) in commercial beers is reported for the first time.Ministerio de Economía y Competitividad AGL2013-47300-C3-2-R, AGL2016-77505-C3-2-

    “Cómo hacer...en arte”: colección de piezas audiovisuales de acceso abierto sobre técnicas y procedimientos del arte contemporáneo

    La serie de videos “CÓMO HACER....EN ARTE”, toma como punto de partida obras artísticas contemporáneas de diversos géneros y estilos para contextualizarlas y explicar las técnicas que están detrás de ellas utilizando para ello pequeños vídeos de entre 5 y 10 minutos de duración. Se conciben por tanto como pequeñas píldoras audiovisuales que nos acercan al cómo hacer, sin por ello menospreciar el qué hacer, pero con un acercamiento eminentemente práctico. De esta manera, en vez de la enseñanza de técnicas diversas aisladas de su contenido, se potencia desde el primer momento poner las técnicas al servicio de las ideas y su expresión. “COMO HACER” centra su trabajo en mostrar cómo se resuelven de forma práctica problemas teóricos y conceptuales planteados, identificando diversas alternativas reales para solucionar un problema, caso o proyecto. Se concibe como un proyecto transversal, planteado desde el contexto de varias asignaturas impartidas desde el Departamento de Dibujo y Grabado de la Facultad de Bellas Artes: animación, videojuegos, tecnologías digitales…, asincrónico, pues a partir de este material generado, se plantean ejercicios para que cada alumno trabaje a su propio ritmo y según sus propias motivaciones

    Development of a rapid lateral flow immunoassay test for detection of exosomes previously enriched from cell culture medium and body fluids

    Exosomes are cell-secreted nanovesicles (40–200 nm) that represent a rich source of novel biomarkers in the diagnosis and prognosis of certain diseases. Despite the increasingly recognized relevance of these vesicles as biomarkers, their detection has been limited due in part to current technical challenges in the rapid isolation and analysis of exosomes. The complexity of the development of analytical platforms relies on the heterogeneous composition of the exosome membrane. One of the most attractive tests is the inmunochromatographic strips, which allow rapid detection by unskilled operators. We have successfully developed a novel lateral flow immunoassay (LFIA) for the detection of exosomes based on the use of tetraspanins as targets. We have applied this platform for the detection of exosomes purified from different sources: cell culture supernatants, human plasma and urine. As proof of concept, we explored the analytical potential of this LFIA platform to accurately quantify exosomes purified from a human metastatic melanoma cell line. The one-step assay can be completed in 15 min, with a limit of detection of 8.54×105 exosomes/µL when a blend of anti-CD9 and anti-CD81 were selected as capture antibodies and anti-CD63 labelled with gold nanoparticles as detection antibody. Based on our results, this platform could be well suited to be used as a rapid exosome quantification tool, with promising diagnostic applications, bearing in mind that the detection of exosomes from different sources may require adaptation of the analytical settings to their specific composition.FICYT; Ministerio de Educación; Ministerio de Economía y Competitividad; Gobierno Regional de Asturias; Gobierno Regional de Madri
