    Social contact networks and mixing among students in K-12 schools in Pittsburgh, PA

    Students attending schools play an important role in the transmission of influenza. In this study, we present a social network analysis of contacts among 1,828 students in eight different schools in urban and suburban areas in and near Pittsburgh, Pennsylvania, United States of America, including elementary, elementary-middle, middle, and high schools. We collected social contact information of students who wore wireless sensor devices that regularly recorded other devices if they are within a distance of 3 meters. We analyzed these networks to identify patterns of proximal student interactions in different classes and grades, to describe community structure within the schools, and to assess the impact of the physical environment of schools on proximal contacts. In the elementary and middle schools, we observed a high number of intra-grade and intra-classroom contacts and a relatively low number of inter-grade contacts. However, in high schools, contact networks were well connected and mixed across grades. High modularity of lower grades suggests that assumptions of homogeneous mixing in epidemic models may be inappropriate; whereas lower modularity in high schools suggests that homogenous mixing assumptions may be more acceptable in these settings. The results suggest that interventions targeting subsets of classrooms may work better in elementary schools than high schools. Our work presents quantitative measures of age-specific, school-based contacts that can be used as the basis for constructing models of the transmission of infections in schools

More recently, technological advances have permitted the use of wireless sensor devices, which can be worn by individuals interacting in a particular social context to record high resolution mixing patterns.     Transcriptome analysis of Bupleurum chinense focusing on genes involved in the biosynthesis of saikosaponins

    <p>Abstract</p> <p>Background</p> <p><it>Bupleurum chinense </it>DC. is a widely used traditional Chinese medicinal plant. Saikosaponins are the major bioactive constituents of <it>B. chinense</it>, but relatively little is known about saikosaponin biosynthesis. The 454 pyrosequencing technology provides a promising opportunity for finding novel genes that participate in plant metabolism. Consequently, this technology may help to identify the candidate genes involved in the saikosaponin biosynthetic pathway.</p> <p>Results</p> <p>One-quarter of the 454 pyrosequencing runs produced a total of 195, 088 high-quality reads, with an average read length of 356 bases (NCBI SRA accession SRA039388). A <it>de novo </it>assembly generated 24, 037 unique sequences (22, 748 contigs and 1, 289 singletons), 12, 649 (52.6%) of which were annotated against three public protein databases using a basic local alignment search tool (E-value ≤1e-10). All unique sequences were compared with NCBI expressed sequence tags (ESTs) (237) and encoding sequences (44) from the <it>Bupleurum </it>genus, and with a Sanger-sequenced EST dataset (3, 111). The 23, 173 (96.4%) unique sequences obtained in the present study represent novel <it>Bupleurum </it>genes. The ESTs of genes related to saikosaponin biosynthesis were found to encode known enzymes that catalyze the formation of the saikosaponin backbone; 246 cytochrome P450 (<it>P450</it>s) and 102 glycosyltransferases (<it>GT</it>s) unique sequences were also found in the 454 dataset. Full length cDNAs of 7 <it>P450</it>s and 7 uridine diphosphate <it>GT</it>s (<it>UGT</it>s) were verified by reverse transcriptase polymerase chain reaction or by cloning using 5' and/or 3' rapid amplification of cDNA ends. Two <it>P450</it>s and three <it>UGT</it>s were identified as the most likely candidates involved in saikosaponin biosynthesis. This finding was based on the coordinate up-regulation of their expression with <it>β-AS </it>in methyl jasmonate-treated adventitious roots and on their similar expression patterns with <it>β-AS </it>in various <it>B. chinense </it>tissues.</p> <p>Conclusions</p> <p>A collection of high-quality ESTs for <it>B. chinense </it>obtained by 454 pyrosequencing is provided here for the first time. These data should aid further research on the functional genomics of <it>B. chinense </it>and other <it>Bupleurum </it>species. The candidate genes for enzymes involved in saikosaponin biosynthesis, especially the <it>P450</it>s and <it>UGT</it>s, that were revealed provide a substantial foundation for follow-up research on the metabolism and regulation of the saikosaponins.</p

    Exploring Usability of School Closure Data for Influenza-Like Illness Surveillance

    We compared tempOral patterns of respiratory illness-related unplanned school closures (USC) with influenza-like illness (ILI) data from outpatient provider visits to determine usability of these data for additional insight regarding ILI activity. We found significant correlation between USC and ILINet data (R= 0.54 with p-value &lt;0.0001). The occurrence pattern of respiratory illness-related USCs similarly corresponded with that of ILI activity regardless of the severity of influenza season. This suggests that monitoring USCs can be a useful addition to existing influenza surveillance systems, particularly during severe influenza seasons when respiratory illness-related USCs may occur more frequently

    Application of BRB to Seismic Mitigation of Steel Truss Arch Bridge Subjected to Near-Fault Ground Motions

    In this paper, the seismic response of a steel truss arch bridge subjected to near-fault ground motions is studied. Then, the idea of applying buckling restrained braces (BRBs) to a steel truss arch bridge in near-fault areas is proposed and validated. Firstly, the basic characteristics of near-fault ground motions are identified and distinguished. Furthermore, the seismic response of a long span steel truss arch bridge in the near fault area is analyzed by elastic-plastic time analysis. Finally, the braces prone to buckling failure are replaced by BRBs to reduce the seismic response of the arch rib through their energy dissipation properties. Four BRB schemes were proposed with different yield strengths, but the same initial stiffness. The basic period of the structure remains the same. The results show that near-fault ground motion will not only obviously increase the displacement and internal force response of the bridge, but also cause more braces to buckle. By replacing a portion of the normal bars with BRBs, the internal forces and displacements of the arch ribs can be reduced to some extent, which is more prominent under the action of pulsed ground motion. There is a clear correlation between the damping effect and the parameters of BRB, so an optimized solution should be obtained by comparison and calculation

    An Enhanced Artificial Electric Field Algorithm with Sine Cosine Mechanism for Logistics Distribution Vehicle Routing

    Aiming at the scheduling problem of logistics distribution vehicles, an enhanced artificial electric field algorithm (SC-AEFA) based on the sine cosine mechanism is proposed. The development of the SC-AEFA was as follows. First, a map grid model for enterprise logistics distribution vehicle path planning was established. Then, an enhanced artificial electric field algorithm with the sine cosine mechanism was developed to simulate the logistics distribution vehicle scheduling, establish the logistics distribution vehicle movement law model, and plan the logistics distribution vehicle scheduling path. Finally, a distribution business named fresh enterprise A in the Fuzhou Strait Agricultural and Sideline Products Trading Market was selected to test the effectiveness of the method proposed. The theoretical proof and simulation test results show that the SC-AEFA has a good optimization ability and a strong path planning ability for enterprise logistics vehicle scheduling, which can improve the scheduling ability and efficiency of logistics distribution vehicles and save transportation costs

    Mycn ameliorates cardiac hypertrophy-induced heart failure in mice by mediating the USP2/JUP/Akt/β-catenin cascade

    Abstract Background Pathological cardiac hypertrophy is associated with cardiac dysfunction and is a key risk factor for heart failure and even sudden death. This study investigates the function of Mycn in cardiac hypertrophy and explores the interacting molecules. Methods A mouse model of cardiac hypertrophy was induced by isoproterenol (ISO). The cardiac dysfunction was assessed by the heart weight-to-body weight ratio (HW/BW), echocardiography assessment, pathological staining, biomarker detection, and cell apoptosis. Transcriptome alteration in cardiac hypertrophy was analyzed by bioinformatics analysis. Gain- or loss-of-function studies of MYCN proto-oncogene (Mycn), ubiquitin specific peptidase 2 (USP2), and junction plakoglobin (JUP) were performed. The biological functions of Mycn were further examined in ISO-treated cardiomyocytes. The molecular interactions were verified by luciferase assay or immunoprecipitation assays. Results Mycn was poorly expressed in ISO-treated mice, and its upregulation reduced HW/BW, cell surface area, oxidative stress, and inflammation while improving cardiac function of mice. It also reduced apoptosis of cardiomyocytes in mice and those in vitro induced by ISO. Mycn bound to the USP2 promoter to activate its transcription. USP2 overexpression exerted similar myocardial protective functions. It stabilized JUP protein by deubiquitination modification, which blocked the Akt/β-catenin pathway. Knockdown of JUP restored phosphorylation of Akt and β-catenin protein level, which negated the protective effects of USP2. Conclusion This study demonstrates that Mycn activates USP2 transcription, which mediates ubiquitination and protein stabilization of JUP, thus inactivating the Akt/β-catenin axis and alleviating cardiac hypertrophy-induced heart failure
