    Mechanical Behavior Analysis and Testing of Marine Riser in Deepwater Drilling

    In this chapter, the mechanical model and control equation have been established to analyse the mechanical behaviour of marine riser in working condition. The control equation has been solved by weighted residual method, and the analysis model has been verified by finite element method (FEM) in ABAQUS framework. Based on this, the deformation and stress distribution of the marine riser have been acquired. Then, a simulation experimental system has been introduced, and the system composition, functions and operational approach of the experimental setup have been stated in detail. After that, a tubular sample has been manufactured to simulate the marine riser, and the simulation experiments have been carried out based on this setup, where the experimental procedures, key aspects, difficult points of the experiment and its corresponding solutions have been elaborated. At last, the strain value of the specimen has been measured successfully after the experiment, and the stress state of the specimen has been obtained based on the analysis

    Drilling Performance Optimization Based on Mechanical Specific Energy Technologies

    Mechanical specific energy (MSE) has been widely used to quantify drilling efficiency and maximize rate of penetration (ROP) in oil and gas wells drilling. In this chapter, MSE models respectively for directional or horizontal drilling and rotating drilling with positive displacement motor (PDM) are established based on the evaluation of virtues and defects of available MSE models. Meanwhile methods for drilling performance prediction and optimization based on MSE technologies are presented. Field data presented in this chapter indicates that the developed MSE models estimate MSE values with a reasonable approximation in the absence of reliable torque measurements, the method for optimizing drilling parameters can estimate optimum WOB values with different RPM to drill a specific formation interval with PDM. It also show that the optimum WOB is low for rotating drilling with PDM compared with the conventional drilling without PDM, increasing WOB does not always increase ROP but is more likely to decrease ROP. The drilling performance prediction and optimization methods based on MSE technologies could be effectively used to maximize ROP and allow operators to drill longer and avoid unnecessary trips, and is worthy to be applied and promoted with highly diagnostic accuracy, effective optimizing and simple operation

    Disentangling Spatial and Temporal Learning for Efficient Image-to-Video Transfer Learning

    Recently, large-scale pre-trained language-image models like CLIP have shown extraordinary capabilities for understanding spatial contents, but naively transferring such models to video recognition still suffers from unsatisfactory temporal modeling capabilities. Existing methods insert tunable structures into or in parallel with the pre-trained model, which either requires back-propagation through the whole pre-trained model and is thus resource-demanding, or is limited by the temporal reasoning capability of the pre-trained structure. In this work, we present DiST, which disentangles the learning of spatial and temporal aspects of videos. Specifically, DiST uses a dual-encoder structure, where a pre-trained foundation model acts as the spatial encoder, and a lightweight network is introduced as the temporal encoder. An integration branch is inserted between the encoders to fuse spatio-temporal information. The disentangled spatial and temporal learning in DiST is highly efficient because it avoids the back-propagation of massive pre-trained parameters. Meanwhile, we empirically show that disentangled learning with an extra network for integration benefits both spatial and temporal understanding. Extensive experiments on five benchmarks show that DiST delivers better performance than existing state-of-the-art methods by convincing gaps. When pre-training on the large-scale Kinetics-710, we achieve 89.7% on Kinetics-400 with a frozen ViT-L model, which verifies the scalability of DiST.     Study on horizontal completion with composite tubular string in coal reservoir with complex coal structure

    Aiming at the problems such as large deformation difference of coal structure, strong heterogeneity of physical properties, complex coal structure during horizontal drilling, single completion & stimulation technology and so on, a completion technology with composite tubular string for coalbed methane (CBM) horizontal well is proposed. Completion tools, inner operation tubing and outer completion string are designed. The outer completion string is composed of casing and screen pipe. The mechanics and hydraulics calculation model of dual tubular string are optimized. A field test of this completion technology had been accomplished in No. 15 coal seam in Yangquan, Shanxi Province, China. A dual tubular string of 659.5 m was run into the depth of 1591 m, and the horizonal well section in coal seam is divided into four sections by external casing packers (ECP). The tests of dual tubular string running, hydraulic circulation removing dual tubular string blocking and sticking, well washing, ECP expansion sealing and segmental completion in coal seam were accomplished. Based on the engineering data of this well, the force of dual tubular string was calculated by finite difference method, and the related hydraulic calculation was carried out. The results show that the inner tubing increases the lateral force of completion string, and the friction resistance on downhole tubular string increases by 5642.75 N. The pressure of completion tubular string is mainly affected by hydraulic loss of inner tubing and nozzles. The recommended displacement of hydraulic circulation is 16~20 L/s to remove rock and cuttings and maintain wellbore stability during completion string running. The displacement is increased to 20~24 L/s to eliminate the damage of drilling fluid to shaft wall during well washing operation. The research and field test verified the feasibility and innovation of the completion technology with composite tubular string for horizontal well, which provides a reliable basis for horizontal well diversified and adaptive stimulation in coal reservoir with complicated structure

    Probiotics MEP+ can increase fowl weight and weft efficiency, therefore it is important to know probiotics MEP+ effect at different dosage toward reproduction aspect. This research aimed to examine duck reproduction organ size suplemented with probiotics MEP+ with different dosage within 30 days. This research used Completely Randomized Design (CRD) with 4 treatments with different dosages within 30 days which was without probiotic\u27s application or control (K), 0,75 ml/kg wefts (P1) dose, 1,5 ml/kg wefts (P2) dose, a n d 3 ml/kg wefts (P3) dose. Each treatment repeated 8 times. Total 40 ducks raised in floor dry cage system. At 31st day of treatment duck reproduction system organ was measured. Whole results show increase average data (Β±SD) for weight of both right and left testis, and liver weight with highly probiotics dosage it, however the analysis result statistic not significant (P>0,05) except weight of right left testis with duck weight or gonadosomatic indeks (GSI) were very significant (P<0,01) among all treatment at different dosages was compared control. The results is confirmed that probiotic\u27s MEP+ treatment with different dosages within 30 days gave no effect towards duck reproduction system organ size except to gonadosomatic indeks (GSI) male duck

    The objective of this research is to stipulate the profile and learning outcome of the graduates of the Study Program of Accounting Education, the Faculty of Teacher Training and Education, Sebelas Maret University. It is intended to help the Indonesian government to prepare the qualified teacher of Accounting subject matter according to the perceptions of alumni, lecturers, stakeholders, profession associations, and decision makers.The data sources of research were 96 students, 248 alumni, 15 lecturers, 15 stakeholders, Association of Accounting Educator Profession of Indonesia, and Chief of the Study Program of Accounting Education, Sebelas Maret University. The data of research were collected through observation, documentation, and FGD. They were analyzed by using the qualitative approach.The results of research show that (1) the profile of the graduates of the Study Program of Accounting Education includes the prospective teachers of Accounting subject matter for Vocational High Schools and Senior Secondary Schools who major in Introduction to Accounting and Finance, Number Processing/Spreadsheet, Banking, Accounting of Service and Trading Company, Financial Accounting, Accounting Computer, Accounting of Manufacturing Company, and Tax Administration; prospective Accounting instructors at non-formal education programs;; edupreneurs in the field of accounting and finance; junior researchers in the field of accounting and finance; and assistant accountants, and (2) the learning outcome expected includes attitude, knowledge, general and special skills, characters, and transferable soft skills which are relevant with the demands of the general public

    DeepSeekMoE: Towards Ultimate Expert Specialization in Mixture-of-Experts Language Models

    In the era of large language models, Mixture-of-Experts (MoE) is a promising architecture for managing computational costs when scaling up model parameters. However, conventional MoE architectures like GShard, which activate the top-KK out of NN experts, face challenges in ensuring expert specialization, i.e. each expert acquires non-overlapping and focused knowledge. In response, we propose the DeepSeekMoE architecture towards ultimate expert specialization. It involves two principal strategies: (1) finely segmenting the experts into mNmN ones and activating mKmK from them, allowing for a more flexible combination of activated experts; (2) isolating KsK_s experts as shared ones, aiming at capturing common knowledge and mitigating redundancy in routed experts. Starting from a modest scale with 2B parameters, we demonstrate that DeepSeekMoE 2B achieves comparable performance with GShard 2.9B, which has 1.5 times the expert parameters and computation. In addition, DeepSeekMoE 2B nearly approaches the performance of its dense counterpart with the same number of total parameters, which set the upper bound of MoE models. Subsequently, we scale up DeepSeekMoE to 16B parameters and show that it achieves comparable performance with LLaMA2 7B, with only about 40% of computations. Further, our preliminary efforts to scale up DeepSeekMoE to 145B parameters consistently validate its substantial advantages over the GShard architecture, and show its performance comparable with DeepSeek 67B, using only 28.5% (maybe even 18.2%) of computations

    ACVR1, a Therapeutic Target of Fibrodysplasia Ossificans Progressiva, Is Negatively Regulated by miR-148a

    Fibrodysplasia ossificans progressiva (FOP) is a rare congenital disorder of skeletal malformations and progressive extraskeletal ossification. There is still no effective treatment for FOP. All FOP individuals harbor conserved point mutations in ACVR1 gene that are thought to cause ACVR1 constitutive activation and activate BMP signal pathway. The constitutively active ACVR1 is also found to be able to cause endothelial-to-mesenchymal transition (EndMT) in endothelial cells, which may cause the formation of FOP lesions. MicroRNAs (miRNAs) play an essential role in regulating cell differentiation. Here, we verified that miR-148a directly targeted the 3β€² UTR of ACVR1 mRNA by reporter gene assays and mutational analysis at the miRNA binding sites, and inhibited ACVR1 both at the protein level and mRNA level. Further, we verified that miR-148a could inhibit the mRNA expression of the Inhibitor of DNA binding (Id) gene family thereby suppressing the BMP signaling pathway. This study suggests miR-148a is an important mediator of ACVR1, thus offering a new potential target for the development of therapeutic agents against FOP

    Enhancement of Naringenin Bioavailability by Complexation with Hydroxypropoyl-Ξ²-Cyclodextrin

    The abundant flavonoid aglycone, naringenin, which is responsible for the bitter taste in grapefruits, has been shown to possess hypolipidemic and anti-inflammatory effects both in vitro and in vivo. Recently, our group demonstrated that naringenin inhibits hepatitis C virus (HCV) production, while others demonstrated its potential in the treatment of hyperlipidemia and diabetes. However, naringenin suffers from low oral bioavailability critically limiting its clinical potential. In this study, we demonstrate that the solubility of naringenin is enhanced by complexation with Ξ²-cyclodextrin, an FDA approved excipient. Hydroxypropoyl-Ξ²-cyclodextrin (HPΞ²CD), specifically, increased the solubility of naringenin by over 400-fold, and its transport across a Caco-2 model of the gut epithelium by 11-fold. Complexation of naringenin with HPΞ²CD increased its plasma concentrations when fed to rats, with AUC values increasing by 7.4-fold and Cmax increasing 14.6-fold. Moreover, when the complex was administered just prior to a meal it decreased VLDL levels by 42% and increased the rate of glucose clearance by 64% compared to naringenin alone. These effects correlated with increased expression of the PPAR co-activator, PGC1Ξ± in both liver and skeletal muscle. Histology and blood chemistry analysis indicated this route of administration was not associated with damage to the intestine, kidney, or liver.
