    The diet of the Tasmanian Devil, Sarcophilus harrisii, as determined from analysis of scat and stomach contents

    Knowledge of the diets of carnivores is an essential precursor to understanding their role as predators in ecosystems. To date, understanding of the diet of Tasmanian Devils, Sarcophilus harrisii, is limited and based upon largely qualitative descriptions. We examined the diets of Tasmanian Devils at six sites by identifying undigested hair, bone and feathers found in their scats. These sites range across different habitat types in coastal and inland Tasmania, and encompass devil populations that are known as both free of the Devil Facial Tumour Disease (DFTD) and populations that are infected by the disease. Tasmanian Devil scats at coastal sites (n=27) contained ten species of mammal, as well as birds, fish and insects. Scats collected from inland sites (n= 17) were comprised of six mammalian species, birds and invertebrates. The most common food items were birds, Common Brushtail and Ringtail possums (Trichosurus vulpecula and Pseudocheirus peregrinus respectively), Tasmanian Pademelons (Thylogale billardierii) and Bennett's Wallabies (Macropus ruftgriseus). O fall the scats, 61% contained only one food group, 32% contained two groups, 4% contained three food items and only one scat (2%) contained four food groups. We supplement this information with stomach contents from road-killed devils, and compare our results with those of previous studies, with a view to furthering our understanding ofthe ecology ofthe threatened Tasmanian Devil. Such information will be important for the management of wild and captive devil populations, particularly in light of DFTD

    Blind Normalization of Speech From Different Channels

    We show how to construct a channel-independent representation of speech that has propagated through a noisy reverberant channel. This is done by blindly rescaling the cepstral time series by a non-linear function, with the form of this scale function being determined by previously encountered cepstra from that channel. The rescaled form of the time series is an invariant property of it in the following sense: it is unaffected if the time series is transformed by any time-independent invertible distortion. Because a linear channel with stationary noise and impulse response transforms cepstra in this way, the new technique can be used to remove the channel dependence of a cepstral time series. In experiments, the method achieved greater channel-independence than cepstral mean normalization, and it was comparable to the combination of cepstral mean normalization and spectral subtraction, despite the fact that no measurements of channel noise or reverberations were required (unlike spectral subtraction).Comment: 25 pages, 7 figure

    Studying seabird diet through genetic analysis of faeces: a case study on Macaroni Penguins (Eudyptes chrysolophus)

    Determination of seabird diet usually relies on the analysis of stomach-content remains obtained through stomach flushing; this technique is both invasive and logistically difficult. We evaluate the usefulness of DNA-based faecal analysis in a dietary study on chick-rearing macaroni penguins (Eudyptes chrysolophus) at Heard Island. Conventional stomach-content data was also collected, allowing comparison of the approaches. Methodology/Principal Findings. Preyspecific PCR tests were used to detect dietary DNA in faecal samples and amplified prey DNA was cloned and sequenced. Of the 88 faecal samples collected, 39 contained detectable DNA from one or more of the prey groups targeted with PCR tests. Euphausiid DNA was most commonly detected in the early (guard) stage of chick-rearing, and detection of DNA from the myctophid fish Krefftichthys anderssoni and amphipods became more common in samples collected in the later (cre`che) stage. These trends followed those observed in the penguins’ stomach contents. In euphausiid-specific clone libraries the proportion of sequences from the two dominant euphausiid prey species (Euphausia vallentini and Thysanoessa macrura) changed over the sampling period; again, this reflected the trend in the stomach content data. Analysis of prey sequences in universal clone libraries revealed a higher diversity of fish prey than identified in the stomachs, but non-fish prey were not well represented. Conclusions/Significance. The present study is one of the first to examine the full breadth of a predator’s diet using DNA based faecal analysis. We discuss methodological difficulties encountered and suggest possible refinements. Overall, the ability of the DNA-based approach to detect temporal variation in the diet of macaroni penguins indicates this non-invasive method will be generally useful for monitoring population-level dietary trends in seabirds

    Terrestrial mammals of a sheep-grazing property on Bruny Island, Tasmania

    Land-based mammals were surveyed in a mosaic of dry sclerophyll forests and pasture on a sheep-grazing property on Bruny Island, Tasmania, using a range of methods in August 2010. 'This is the first mammal survey of a sheep-grazing property in Tasmania and the first large-scale survey of mammals on Bruny Island. Ten species were recorded comprising seven native and three introduced species. The Little Forest Bat, Vespadelus vulturnus, and the Black Rat, Rattus rattus, were recorded for the first time on Bruny Island, although both are probably long-term residents. No mammal species listed as rare or threatened under Tasmanian or Australian legislation were found on the property. Large numbers of Eastern Quolls, Dasyurus viverrinus, Brushtail Possums, Trichosurus vulpecula, Tasmanian Pademelons, Thylogale billardierii, and Bennetts Wallabies, Macropus rufogriseus, were recorded in a range of dry sclerophyll forests and in pasture. Longnosed Potoroos, Potorous tridactylus, were recorded widely on the property in native vegetation with relatively thick ground cover. Eastern Quoll capture rates were highest in pasture areas and in Eucalyptus ovata forest. Brushtail Possums, Long-nosed Potoroos, Tasmanian Pademelons and Bennetts Wallabies were virtually unrecorded from E. tenuiramis forest and woodlands. Given the level of survey effort and their potential to occur on the property it was remarkable that no Tasmanian Bettong, Bettongia gaimardi, Eastern Barred Bandicoot, Perameles gunnii, Southern Brown Bandicoot, lsoodon obesulus, or introduced House Mouse, Mus musculus, were recorded. We found that camera trapping was more cost-efficient than cage trapping for detecting the presence of mammals on "Murrayfield".     The RCK2 domain of the human BKCa channel is a calcium sensor

    Large conductance voltage and Ca2+-dependent K+ channels (BKCa) are activated by both membrane depolarization and intracellular Ca2+. Recent studies on bacterial channels have proposed that a Ca2+-induced conformational change within specialized regulators of K+ conductance (RCK) domains is responsible for channel gating. Each pore-forming α subunit of the homotetrameric BKCa channel is expected to contain two intracellular RCK domains. The first RCK domain in BKCa channels (RCK1) has been shown to contain residues critical for Ca2+ sensitivity, possibly participating in the formation of a Ca2+-binding site. The location and structure of the second RCK domain in the BKCa channel (RCK2) is still being examined, and the presence of a high-affinity Ca2+-binding site within this region is not yet established. Here, we present a structure-based alignment of the C terminus of BKCa and prokaryotic RCK domains that reveal the location of a second RCK domain in human BKCa channels (hSloRCK2). hSloRCK2 includes a high-affinity Ca2+-binding site (Ca bowl) and contains similar secondary structural elements as the bacterial RCK domains. Using CD spectroscopy, we provide evidence that hSloRCK2 undergoes a Ca2+-induced change in conformation, associated with an α-to-β structural transition. We also show that the Ca bowl is an essential element for the Ca2+-induced rearrangement of hSloRCK2. We speculate that the molecular rearrangements of RCK2 likely underlie the Ca2+-dependent gating mechanism of BKCa channels.     Processes influencing differences in Arctic and Antarctic Trough Mouth Fan sedimentology

    Trough Mouth Fans (TMFs) are sediment depocentres that form along high-latitude continental margins at the mouths of some cross-shelf troughs. They reflect the dynamics of past ice sheets over multiple glacial cycles and processes operating on (formerly) glaciated continental shelves and slopes, such as erosion, reworking, transport and deposition. The similarities and differences in TMF morphology and formation processes of the Arctic and Antarctic regions remain poorly constrained. Here, we analyse the dimensions and geometries of 15 TMFs from Arctic and Antarctic margins and the grain-size distribution of 82 sediment cores centred on them. We compare the grain-size composition of sub- and proglacial diamictons deposited on the shelves and glacigenic-debris flows (GDFs) deposited on the adjacent TMFs and find a significant difference between Arctic and Antarctic margins. Antarctic margins show a coarser grain-size composition for both GDFs and shelf diamictons. This significant difference provides insight into high-latitude sediment input, transportation and glacial/interglacial regimes. We suggest that surface run-off and river discharge are responsible for enhanced fine-grained sediment input in the Arctic compared to in the Antarctic

    On the extraction of weak transition strengths via the (3He,t) reaction at 420 MeV

    Differential cross sections for transitions of known weak strength were measured with the (3He,t) reaction at 420 MeV on targets of 12C, 13C, 18O, 26Mg, 58Ni, 60Ni, 90Zr, 118Sn, 120Sn and 208Pb. Using this data, it is shown the proportionalities between strengths and cross sections for this probe follow simple trends as a function of mass number. These trends can be used to confidently determine Gamow-Teller strength distributions in nuclei for which the proportionality cannot be calibrated via beta-decay strengths. Although theoretical calculations in distorted-wave Born approximation overestimate the data, they allow one to understand the main experimental features and to predict deviations from the simple trends observed in some of the transitions.Comment: 4 pages, 2 figure
