    Background: Dysbiosis in lung cancer has been underexplored. The aim of this study was to define the bacterial and fungal microbiota of the bronchi in central lung cancer and to compare it with that of the oral and intestinal compartments. Methods: Twenty-five patients with central lung cancer and sixteen controls without antimicrobial intake during the previous month were recruited. Bacterial and fungal distribution was determined by massive sequencing of bronchial biopsies and saliva and faecal samples. Complex computational analysis was performed to define the core lung microbiota. Results: Affected and contralateral bronchi of patients have almost identical microbiota dominated by Streptococcus, whereas Pseudomonas was the dominant genera in controls. Oral and pulmonary ecosystems were significantly more similar in patients, probably due to microaspirations. Streptococcal abundance in the bronchi differentiated patients from controls according to a ROC curve analysis (90.9% sensitivity, 83.3% specificity, AUC = 0.897). The saliva of patients characteristically showed a greater abundance of Streptococcus, Rothia, Gemella and Lactobacillus. The mycobiome of controls (Candida) was significantly different from that of patients (Malassezia). Cancer patients’ bronchial mycobiome was similar to their saliva, but different from their contralateral bronchi. Conclusions: The central lung cancer microbiome shows high levels of Streptococcus, and differs significantly in its composition from that of control subjects. Changes are not restricted to tumour tissue, and seem to be the consequence of microaspirations from the oral cavity. These findings could be useful in the screening and even diagnosis of this disease

    New insights into the genetic etiology of Alzheimer's disease and related dementias

    Characterization of the genetic landscape of Alzheimer's disease (AD) and related dementias (ADD) provides a unique opportunity for a better understanding of the associated pathophysiological processes. We performed a two-stage genome-wide association study totaling 111,326 clinically diagnosed/'proxy' AD cases and 677,663 controls. We found 75 risk loci, of which 42 were new at the time of analysis. Pathway enrichment analyses confirmed the involvement of amyloid/tau pathways and highlighted microglia implication. Gene prioritization in the new loci identified 31 genes that were suggestive of new genetically associated processes, including the tumor necrosis factor alpha pathway through the linear ubiquitin chain assembly complex. We also built a new genetic risk score associated with the risk of future AD/dementia or progression from mild cognitive impairment to AD/dementia. The improvement in prediction led to a 1.6- to 1.9-fold increase in AD risk from the lowest to the highest decile, in addition to effects of age and the APOE ε4 allele

    Background & Aims: The mechanisms by which Foxp3+ T regulatory cells (Treg) accumulate in HCV infected livers are not known. Here, we studied the role of chemokines CCL17 and CCL22 in this process. Methods: Chemokine mRNA levels were determined by qPCR in liver biopsies from 26 HCV chronically infected patients (CHC), 11 patients with treatment-induced sustained virological response (SVR), 16 patients with other liver diseases unrelated to HCV, and 24 normal livers. Double-immunofluorescence Foxp3/CD3 or CD11c/CCL22 was performed in liver sections. Chemokine production by monocyte-derived dendritic cells (MDDC) co-cultured with uninfected or HCV-JFH1 infected Huh7 cells was measured by qPCR and ELISA. Chemotactic activity of culture supernatants was also tested. Results: Foxp3+ Treg were increased in CHC livers as compared to controls. Patients with CHC showed elevated intrahepatic levels of CCL17 mRNA compared to normal livers or livers from subjects with SVR or other forms of liver disease. Intrahepatic CCL22 expression was also higher in CHC than in healthy subjects or SVR patients but similar to that observed in other liver diseases. Dendritic cells producing CCL22 could be found inside the hepatic lobule in CHC patients. Contact between MDDC and HCV-JFH1- infected Huh7 cells induced the expression of CCL17 and CCL22 in a process partially dependent on ICAM-1.     A Middle to Late Miocene Trans-Andean Portal: Geologic Record in the Tatacoa Desert

    Integration of several geologic lines of evidence reveals the prevalence of a lowland trans-Andean portal communicating western Amazonia and the westernmost Andes from at least middle Miocene until Pliocene times. Volcanism and crustal shortening built up relief in the southernmost Central and Eastern Cordilleras of Colombia, closing this lowland gap. Independent lines of evidence consist first, of field mapping in the Tatacoa Desert with a coverage area of ∼381 km2, 1,165 km of geological contact traces, 164 structural data points, and 3D aerial digital mapping models. This map documents the beginning of southward propagation of the southernmost tip of the Eastern Cordillera’s west-verging, fold-and-thrust belt between ∼12.2 and 13.7 Ma. Second, a compilation of new and published detrital zircon geochronology in middle Miocene strata of the Tatacoa Desert shows three distinctive age populations: middle Miocene, middle Eocene, and Jurassic; the first two sourced west of the Central Cordillera, the latter in the Magdalena Valley. Similar populations with the three distinctive peaks have now been recovered in western Amazonian middle Miocene strata. These observations, along with published molecular and fossil fish data, suggest that by Serravallian times (∼13 Ma), the Northern Andes were separated from the Central Andes at ∼3°N by a fluvial system that flowed into the Amazon Basin through the Tatacoa Desert. This paleogeographic configuration would be similar to a Western Andean, or Marañon Portal. Late Miocene flattening of the subducting Nazca slab caused the eastward migration of the Miocene volcanic arc, so that starting at ∼4 Ma, large composite volcanoes were built up along the axis of today's Central Cordillera, closing this lowland Andean portal and altering the drainage patterns to resemble a modern configuration.ISSN:2296-646

    Maradona Inc: Performance politics off the pitch

    This article addresses the blurring boundaries between celebrity, media sports, cultural identity and politics. I explore the case of media sport star Diego Maradona, whose strategies of public representation highlight the way in which politics and popular culture can overlap. First, the article offers a general overview of the theorization of football in Latin America. Central to my discussion is the problematic use of concepts like `identity' and `belonging' to define football politics. I insist on a theoretical reformulation of political culture in global contexts, and point to an understanding of media personalities such as Maradona as currency within markets of cultural and political production. This analysis suggests that `Maradona Inc.' can be understood as a corporate and performative semiosphere that is continuously shifting, so that fans can project onto the fetish-like and vacant image of the celebrity whatever they wish
