    Role of enzymic antioxidants in mediating oxidative stress and contrasting wound healing capabilities in oral mucosal/skin fibroblasts and tissues

    Unlike skin, oral mucosal wounds are characterized by rapid healing and minimal scarring, attributable to the “enhanced” healing properties of oral mucosal fibroblasts (OMFs). As oxidative stress is increasingly implicated in regulating wound healing outcomes, this study compared oxidative stress biomarker and enzymic antioxidant profiles between patient-matched oral mucosal/skin tissues and OMFs/skin fibroblasts (SFs) to determine whether superior oral mucosal antioxidant capabilities and reduced oxidative stress contributed to these preferential healing properties. Oral mucosa and skin exhibited similar patterns of oxidative protein damage and lipid peroxidation, localized within the lamina propria/dermis and oral/skin epithelia, respectively. SOD1, SOD2, SOD3 and catalase were primarily localized within epithelial tissues overall. However, SOD3 was also widespread within the lamina propria localized to OMFs, vasculature and the extracellular matrix. OMFs were further identified as being more resistant to reactive oxygen species (ROS) generation and oxidative DNA/protein damage than SFs. Despite histological evaluation suggesting that oral mucosa possessed higher SOD3 expression, this was not fully substantiated for all OMFs examined due to inter-patient donor variability. Such findings suggest that enzymic antioxidants have limited roles in mediating privileged wound healing responses in OMFs, implying that other non-enzymic antioxidants could be involved in protecting OMFs from oxidative stress overall

    In recent years, UV-free blue light has been proposed to supplement current treatment modalities for periodontal disease, owing to its antimicrobial properties which are specific to periopathogenic bacterial species. However, little work has sought to investigate the influence of blue light upon periodontal support tissues. The aim of this study was to investigate discrete molecular targets of 457 nm blue light in primary human gingival fibroblasts (pHGFs) and primary human gingival keratinocytes (pHGKs), with a view to identify cellular responses and proteinaceous and genetic targets which characterise photobiomodulation (PBM) in these cell types. pHGFs or pHGKs received doses of 457 nm blue light ranging from 3 J/cm2 to 90 J/cm2 , prior to experimental assay. pHGFs displayed reductions in cell health, while pHGK proliferation and metabolic activity was enhanced following irradiation (3 – 90 J/cm2 ). Treatment of pHGFs with blue light (3 – 60 J/cm2 ) induced significant generation of reactive oxygen species (ROS), which was inhibited by the presence of an ROS scavenger, N-acetyl cysteine (NAC). Comparable trends in ROS generation and scavenging were also evident in pHGKs irradiated with blue light, however these responses were not significant. pHGFs displayed subtle stimulation of antioxidant responses, while pHGKsshowed no change relative to untreated controls. Analyses of extracellular matrix genes and growth factor secretion from pHGFs revealed significant increases in hepatocyte growth factor (HGF), vascular endothelial growth factor (VEGF) and matrix metalloproteinase 1 (MMP-1), in response to blue light. Presence of NAC during irradiations resulted in inhibition of MMP-1 gene upregulation, thereby correlating generation of ROS with subsequent gene expression and protein secretion. Preliminary insights into use of ex vivo rodent gingival tissue to model PBM responses showed promise, with no detrimental effects observable after a single high fluence dose (60 J/cm2 ), compared to untreated samples. In conclusion, pHGFs and pHGKs displayed differential responses to 457 nm blue light. These findings corroborate current literature, which point toward cell type specific PBM responses. Importantly ROS was established as a PBM mechanism through which secondary cellular responses are mediated. Additionally, growth factors associated with gingival wound healing and tissue remodelling were also influenced by blue light treatments. These findings provide ii novel evidence that UV-free wavelengths of blue light have the potential to confer preferential effects upon gingival tissues and, as such, be used as adjunctive or prophylactic treatments in the management of periodontal disease

    In vitro cytocompatibility of a Zr-based metallic glass modified by laser surface texturing for potential implant applications

    Existing studies have shown the benefit of laser surface texturing (LST) in promoting the cytocompatibility of traditional metallic biomaterials. Researchers have also reported the potential of bulk metallic glasses (BMGs) as an alternative class of material for biomedical applications. However, investigations specifically focussed on studying the cytocompatibility of BMG surfaces processed with LST are still lacking. The present work demonstrated the feasibility of nanosecond LST as a method to modify the cytocompatibility of a Zr-based BMG material known as Vitreloy 105. Two different types of laser-induced surface patterns, namely grooves and dimples, were considered. Their respective influence on the resulting cell viability, attachment and morphology was studied and compared against the cytocompatibility of the original BMG surface. It was found that MG63 osteoblast-like cells on the groove-textured surface exhibited higher viability and better adhesion compared to those on the original and dimple-textured surfaces. Possible underlying mechanisms associated with LST, which can affect the in vitro cytocompatibility of Vitreloy 105 were discussed based on the induced changes to surface chemistry, wettability and roughness. It is suggested that the higher surface roughness, increased presence of metallic oxides and enhanced hydrophilicity of the groove-textured sample were the main contributors to its improved cytocompatibility
