    Ovalbumin-Based Porous Scaffolds for Bone Tissue Regeneration

    Cell differentiation on glutaraldehyde cross-linked ovalbumin scaffolds was the main focus of this research. Salt leaching and freeze drying were used to create a three-dimensional porous structure. Average pore size was 147.84 ± 40.36 μm and 111.79 ± 30.71 μm for surface and cross sectional area, respectively. Wet compressive strength and elastic modulus were 6.8 ± 3.6 kPa. Average glass transition temperature was 320.1 ± 1.4°C. Scaffolds were sterilized with ethylene oxide prior to seeding MC3T3-E1 cells. Cells were stained with DAPI and Texas red to determine morphology and proliferation. Average cell numbers increased between 4-hour- and 96-hour-cultured scaffolds. Alkaline phosphatase and osteocalcin levels were measured at 3, 7, 14, and 21 days. Differentiation studies showed an increase in osteocalcin at 21 days and alkaline phosphatase levels at 14 days, both indicating differentiation occurred. This work demonstrated the use of ovalbumin scaffolds for a bone tissue engineering application

    The impact of COVID-19 hygienic measures on food choice and eating behaviour

    The COVID-19 pandemic has led to several changes that have affected not only emotional and psychological well-being but also eating behavior. Herein, we sought to evaluate if depicting pandemic-related sanitary measures in a food consumption video would impact food appraisal, decreasing food attractiveness and the desire to eat. Participants performed, on two different days, an online protocol in which foods presented in a video, and following, in pictures, were evaluated regarding visual aspect, expected smell and taste, and desire to eat. The videos presented each day differed only regarding the presence/absence of sanitary elements adopted during the pandemic, composing COVID/non-COVID conditions. For both conditions, sweet and high-calorie foods were better evaluated when compared to salty and low-calorie foods, with the sweet food evaluation higher for the non-COVID condition when compared to the COVID-condition. Only for the non-COVID condition was the reported desire to eat higher immediately after the video, and for both conditions, it was higher at the end of the task when compared to the baseline. Correlation analysis suggests that depression and anxiety symptoms may reduce the smell appreciation of foods presented in the pandemic scenario.     Impacts of COVID-19 sanitary cues on hedonic appreciation of foods

    The COVID-19 pandemic led to several lifestyle changes, including eating behavior. Herein, we aimed to evaluate how pandemic-related sanitary cues presented in food videos impact food appraisal and desire to eat, and their priming after-effects on subsequent food pictures presented without such cues. In two online sessions, separated by 4 to 7 days, participants watched either a Non-Pandemic or a Pandemic video of a woman eating, the latter containing sanitary elements adopted during the pandemic. The order of the videos was counterbalanced across participants over the two experimental sessions. Videos were followed by images of food from different categories. After observing both videos and each picture, participants were instructed to evaluate the visual aspect, expected smell and flavor, and rate their desire to eat. Our study demonstrated (1) higher hedonic responses to the Non-Pandemic compared to the Pandemic video, (2) a priming effect showing higher appreciation for sweet foods after the Non-Pandemic compared to the Pandemic video, (3) that food exposure gradually increases one’s desire to eat, but such effects are impacted by pandemic sanitary cues, and (4) greater hedonic responses are given for sweet and high-calorie foods compared to salty and low-calorie ones, irrespective of pandemic priming.     Main geochemical characteristics of the deep geothermal brine at Vendenheim (Alsace, France) with constraints on temperature and fluid circulation

    International audienceThe first analytical results relative to the native geothermal brine discharged from the two deep wells drilled at Vendenheim, in the Rhine Graben, in Alsace (France), obtained within the framework of the Vendenheim FONROCHE geothermal project and the H2020 European DEEP-EGS project, suggest the geochemical composition of this brine is very similar to that of the fluid which was discharged from the neighboring Cronenbourg deep well, in the past. It is also close to that of the brines discharged from the other deep wells located in more northern areas of the Rhine Graben, such as Soultz-sous-Forêts, Rittershoffen, in France, and Landau, Insheim, in Germany. This Na-Cl brine has a TDS value around 100 g/l and a pH value close to 5, before cooling and degassing. Except for the calcium and strontium concentration values, which are much lower than those in the Soultz-sous-Forêts, Rittershoffen, Landau and Insheim brines, the other concentration values of major and trace species are comparable. Given the similarity of the geochemical composition of all these waters, their origin is probably similar but also multiple, because it results from processes of mixing between primary brines formed by advanced evaporation of seawater (probably until the stage of halite precipitation) and meteoric freshwaters, plus contributions from halite dissolution following successive marine transgression-regression cycles from the Triassic to Oligocene. As for the other deep brines, the main solute cation geothermometers give estimations of reservoir temperature close to 225 ± 25°C for the Vendenheim native brine. This estimation probably corresponds to the temperature of equilibrium at which this brine acquires its chemical composition by interaction with the reservoir rocks. The mineralogical assemblage in equilibrium with the brine at this temperature was described in previous studies. The concordant estimations of reservoir temperature, using thermometric relationships such Na-Li and Mg/Li, especially developed for oilfield and sedimentary basin brines, and existing thermal gradients from 40 to 60°C/km, suggest that the deep brines discharged from the granite basement could probably originate from Triassic sedimentary formations (Buntsandstein, for example) located at great depth (≥ 4 km), in the centre of the Rhine Graben, in which they acquire their high salinity and chemical composition at temperatures close to 225 ± 25°C. This assumption seems to be also supported by their Li, B and Sr isotopic signatures. These hot brines would then migrate through a complex, but still poorly defined system of deep faults (probably NE-SW but also NW-SE faults), from the sedimentary centre of the Rhine Graben to the granite-fractured basement and the Graben's NW borders

    Transcriptomes of peripheral blood mononuclear cells from juvenile dermatomyositis patients show elevated inflammation even when clinically inactive

    In juvenile dermatomyositis (JDM), the most common pediatric inflammatory myopathy, weakness is accompanied by a characteristic rash that often becomes chronic and is associated with vascular damage. We hoped to understand the molecular underpinnings of JDM, particularly when untreated, which would facilitate the identification of novel mechanisms and clinical targets that might disrupt disease progression. We studied the RNA-Seq data from untreated JDM peripheral blood mononuclear cells (PBMCs; n = 11), PBMCs from a subset of the same patients when clinically inactive (n = 8/11), and separate samples of untreated JDM skin and muscle (n = 4 each). All JDM samples were compared to non-inflammatory control tissues. The untreated JDM PBMCs showed a strong signature for type1 interferon response, along with IL-1, IL-10, and NF-κB. Surprisingly, PBMCs from clinically inactive JDM individuals had persistent immune activation that was enriched for IL-1 signaling. JDM skin and muscle both showed evidence for type 1 interferon activation and genes related to antigen presentation and decreased expression of cellular respiration genes. Additionally, we found that PBMC gene expression correlates with disease activity scores (DAS; skin, muscle, and total domains) and with nailfold capillary end row loop number (an indicator of microvascular damage). This included otoferlin, which was significantly increased in untreated JDM PBMCs and correlated with all 3 DAS domains. Overall, these data demonstrate that PBMC transcriptomes are informative of molecular disruptions in JDM and provide transcriptional evidence of chronic inflammation despite clinical quiescence

    mHealth Video Gaming For Human Papillomavirus Vaccination Among College Men-Qualitative Inquiry For Development

    Background: Human papillomavirus (HPV) is the most common sexually transmitted infection in the United States. Persistent infection with HPV can cause various cancers; however, HPV vaccination can prevent infections associated with high risk, cancerous strains of the virus. As it relates to HPV, college age men have been identified as one of the catch-up vaccination groups. Among college age men, gaming is an extremely popular extracurricular activity. Further, video games have emerged as a popular public health intervention tool. Therefore, this study aims to collect qualitative data on how to develop, implement and evaluate the effectiveness of a gaming intervention to increase HPV risk perceptions, improve self-efficacy and increase intention to receive the HPV vaccine among male college students (18–26 years old). Methods: Four focus group sessions ranging from eight to ten individuals were conducted among male college students from one large research-intensive university in the South. Using grounded theory, data from focus group interviews were coded using NVivo software to identify emergent themes. Results: Participants emphasized that although customization was not viewed as important by college aged males, the ability to tailor in game experiences or experience different things each time they played (creative freedom) was more important. They encouraged that the digital game be created on a mobile platform, incorporate health messages, and be informative to reach their population. Furthermore, they suggested innovative way to disseminate the game, which included having health department/health care providers prescribe the game to patients as an end of clinical interaction strategy. Conclusions: College age men, are natural avid gamers, enjoy game play, and can engage in learning online or offline. While platform preference varies among gamer type, college age men in our study emphasized that mobile based gaming is the most advantageous way to increase knowledge/awareness and encourage positive in game behavior which can impact out of game behaviors such as vaccination. Because of the level of access and natural disposition of mHealth technology seen as an “extension of the self”, games for health developers should consider the mobile platform as the ideal for the target demographic

    Optimising Antimicrobial Selection and Duration in the Treatment of Febrile Neutropenia in Children

    Febrile neutropenia (FN) is a frequent complication of cancer treatment in children. Owing to the potential for overwhelming bacterial sepsis, the recognition and management of FN requires rapid implementation of evidenced-based management protocols. Treatment paradigms have progressed from hospitalisation with broad spectrum antibiotics for all patients, through to risk adapted approaches to management. Such risk adapted approaches aim to provide safe care through incorporating antimicrobial stewardship (AMS) principles such as implementation of comprehensive clinical pathways incorporating de-escalation strategies with the imperative to reduce hospital stay and antibiotic exposure where possible in order to improve patient experience, reduce costs and diminish the risk of nosocomial infection. This review summarises the principles of risk stratification in FN, the current key considerations for optimising empiric antimicrobial selection including knowledge of antimicrobial resistance patterns and emerging technologies for rapid diagnosis of specific infections and summarises existing evidence on time to treatment, investigations required and duration of treatment. To aid treating physicians we suggest the key features based on current evidence that should be part of any FN management guideline and highlight areas for future research. The focus is on treatment of bacterial infections although fungal and viral infections are also important in this patient group

    Creation and confidence: BME students as academic partners…but where were the staff?

    This Case Study documents the REACT project 'Creation and Confidence' based at Sheffield Hallam University, which has a larger-than-expected Black Minority and Ethnic (BME) attainment gap; hence, this student group has been constructed as 'hard to reach'. The project team consisted of a range of academic and professional services staff alongside three dedicated student researchers. The project set out to achieve: gaining evidence-based insights into the use of co-design and peer-learning as conduits of confidence-building for and belonging of BME students; developing a scalable approach to building confidence for and fostering belonging of all students; raising awareness of the need to think differently about explanations for BME underachievement. In reality, the team found that staff engagement constituted the biggest barrier, as - no matter how much incontrovertible evidence was presented - other facets of institutional provision were always identified as having priority, which resulted in inertia. This study documents the emotional labour of trying to effect change within a resistant culture. Whilst some of the aims remain unachieved – and, arguably, were always going to be unachievable - there have been some very positive developments and enlightening lessons

    Variants Within Genes \u3ci\u3eEDIL3\u3c/i\u3e and \u3ci\u3eADGRB3\u3c/i\u3e are Associated With Divergent Fecal Egg Counts in Katahdin Sheep at Weaning

    Gastrointestinal nematodes (GIN) pose a severe threat to sheep production worldwide. Anthelmintic drug resistance coupled with growing concern regarding potential environmental effects of drug use have demonstrated the necessity of implementing other methods of GIN control. The aim of this study was to test for genetic variants associated with resistance or susceptibility to GIN in Katahdin sheep to improve the current understanding of the genetic mechanisms responsible for host response to GIN. Linear regression and casecontrol genome-wide association studies were conducted with high-density genotype data and cube-root transformed weaning fecal egg counts (tFEC) of 583 Katahdin sheep. The casecontrol GWAS identified two significant SNPs (P-values 1.49e-08 to 1.01e-08) within introns of the gene adhesion G protein-coupled receptor B3 (ADGRB3) associated with lower fecal egg counts. With linear regression, four significant SNPs (P-values 7.82e-08 to 3.34e-08) were identified within the first intron of the gene EGF-like repeats and discoidin domains 3 (EDIL3). These identified SNPs were in very high linkage disequilibrium (r2 of 0.996–1), and animals with alternate homozygous genotypes had significantly higher median weaning tFEC phenotypes compared to all other genotypes. Significant SNPs were queried through public databases to identify putative transcription factor binding site (TFBS) and potential lncRNA differences between reference and alternate alleles. Changes in TFBS were predicted at two SNPs, and one significant SNPwas found to bewithin a predicted lncRNA sequencewith greater than 90% similarity to a known lncRNA in the bovine genome. The gene EDIL3 has been described in other species for its roles in the inhibition and resolution of inflammation. Potential changes of EDIL3 expression mediated through lncRNA expression and/or transcription factor binding may impact the overall immune response and reduce the ability of Katahdin sheep to control GIN infection. This study lays the foundation for further research of EDIL3 and ADGRB3 towards understanding genetic mechanisms of susceptibility to GIN, and suggests these SNPs may contribute to genetic strategies for improving parasite resistance traits in sheep
