    A Transposon in Comt Generates mRNA Variants and Causes Widespread Expression and Behavioral Differences among Mice

    Background: Catechol-O-methyltransferase (COMT) is a key enzyme responsible for the degradation of dopamine and norepinephrine. COMT activity influences cognitive and emotional states in humans and aggression and drug responses in mice. This study identifies the key sequence variant that leads to differences in Comt mRNA and protein levels among mice, and that modulates synaptic function and pharmacological and behavioral traits. Methodology/Principal Findings: We examined Comt expression in multiple tissues in over 100 diverse strains and several genetic crosses. Differences in expression map back to Comt and are generated by a 230 nt insertion of a B2 short interspersed element (B2 SINE) in the proximal 39 UTR of Comt in C57BL/6J. This transposon introduces a premature polyadenylation signal and creates a short 39 UTR isoform. The B2 SINE is shared by a subset of strains, including C57BL/6J

    Unexpected changes in the oxic/anoxic interface in the Black Sea

    THE Black Sea is the largest anoxic marine basin in the world today1. Below the layer of oxygenated surface water, hydrogen sulphide builds up to concentrations as high as 425 μM in the deep water down to a maximum depth of 2,200 m (ref. 2). The hydrographic regime is characterized by low-salinity surface water of river origin overlying high-salinity deep water of Mediterranean origin1,3. A steep pycnocline, centred at about 50 m is the primary physical barrier to mixing and is the origin of the stability of the anoxic (oxygen/hydrogen sulphide) interface. Here we report new observations, however, that indicate dramatic changes in the oceanographic characteristics of the anoxic interface of the Black Sea over decadal or shorter timescales. The anoxic, sulphide-containing interface has moved up in the water column since the last US cruises in 1969 and 1975. In addition, a suboxic zone overlays the sulphide-containing deep water. The expected overlap of oxygen and sulphide was not present. We believe that these observations result from horizontal mixing or flushing events that inject denser, saltier water into the relevant part of the water column. It is possible that man-made reduction in freshwater inflow into the Black Sea could cause these changes, although natural variability cannot be discounted. © 1989 Nature Publishing Group

    Foi avaliada a eficácia da administração prolongada de azitromicina e pirimetamina em camundongos infectados com cepa cistogênica de Toxoplasma gondii. Os animais foram inoculados intraperitonealmente com um cisto de T. gondii e, após 20 dias, divididos em quatro grupos: GI infectados não tratados, GII infectados e tratados concomitantemente com pirimetamina (12,5mg/kg/dia) e azitromicina (100mg/kg/dia), GIII infectados e tratados com a mesma dose de pirimetamina e GIV infectados e tratados da mesma forma com azitromicina. O tratamento, via oral, estendeu-se por 120 dias; após este período os animais foram sacrificados e foi feita a contagem dos cistos no cérebro. A associação de ambos os medicamentos proporcionou melhores resultados, diminuindo a contagem de cistos no cérebro dos animais tratados de forma concomitante.<br>The efficacy of prolonged administration of azithromycin and pyrimethamine was evaluated in mice experimentally infected with cystogenic strain of Toxoplasma gondii. The animals were intraperitoneally inoculated with one cyst of T. gondii and after 20 days were allocated into four groups: GI, infected without treatment; GII, infected and treated with the association of pyrimethamine (12.5mg/kg/day) and azithromycin (100mg/kg/day); GIII, infected and treated with the same dose of pyrimethamine; and GIV, infected and treated in the same way with azithromycin. The oral treatment lasted 120 days, after this period all the animals were sacrificed and the count of cysts in the brain was done. The association of both drugs provided the best results, by diminishing the cyst count in the brain of the animals treated in this way
