    A lesson not learned: allele misassignment

    Misassigned alleles can annihilate efforts to control quality in otherwise well-designed genetic association analyses. To date, the issue remains underreported, as is exemplified by studies of a diallelic DRD2 missense variant in schizophrenia. For this variant, allele frequency data have been either misassigned, or incorrectly cited on four consecutive occasions. Contrary to conjecture, low heterozygosity has not guarded against the error with regard to rs1801028, a SNP that features a canonical base pair transversion, G:C. Measures are discussed that may help to identify misassigned alleles, and to avoid related perils pending more systematic investigation of this confounder in genotype-phenotype associations

    To achieve true sustainability, planners must balance the interests of environmental protection, economic development, and social equity. In a critically changing, complex ecosystem such as Louisiana’s coast, challenges to achieve the perfect equilibrium are further compounded. Following the logic of emerging adaptive co-management literature, the planning framework for Louisiana’s Coastal Master Plan could be transformed into a more collaborative, democratic process. Adaptive co-management is a regime which provides power to local citizens, often in possession of invaluable traditional ecological knowledge. In general, it focuses on constant learning and collaboration. Through power-sharing and participatory action, we embrace science, but step back from technocracy.     Irregular orbits generate higher harmonics

    The spectrum of higher harmonics in atoms calculated with a uniformized semiclassical propagator is presented and it is shown that higher harmonic generation is an interference phenomenon which can be described semiclassically. This can be concluded from the good agreement with the quantum spectrum. Moreover, the formation of a plateau in the spectrum is specifically due to the interference of irregular, time delayed, trajectories with regular orbits without a time-delay. This is proven by the absence of the plateau in an artificial semiclassical spectrum generated from a sample of trajectories from which the irregular trajectories (only a few percent) have been discarded

    Deep resequencing of the voltage-gated potassium channel subunit KCNE3 gene in chronic tinnitus

    Membrane-stabilizing drugs have long been used for the treatment of chronic tinnitus, suggesting an underlying disturbance of sensory excitability due to changes in ion conductance. The present study addresses the potassium channel subunit gene KCNE3 as a potential candidate for tinnitus susceptibility. 288 Caucasian outpatients with a diagnosis of chronic tinnitus were systematically screened for mutations in the KCNE3 open reading frame and in the adjacent region by direct sequencing. Allele frequencies were determined for 11 known variants of which two (F66F and R83H) were polymorphic but were not associated with the disorder. No novel variants were identified and only three carriers of R83H were noted. However, owing to a lack of power, our study can neither rule out effects of KCNE3 on the risk for developing chronic tinnitus, nor can it exclude a role in predicting the severity of tinnitus. More extensive investigations are invited, including tests for possible effects of variation in this ion channel protein on the response to treatment

    Analytic Gradients for Complete Active Space Pair-Density Functional Theory

    Analytic gradient routines are a desirable feature for quantum mechanical methods, allowing for efficient determination of equilibrium and transition state structures and several other molecular properties. In this work, we present analytical gradients for multiconfiguration pair-density functional theory (MC-PDFT) when used with a state-specific complete active space self-consistent field reference wave function. Our approach constructs a Lagrangian that is variational in all wave function parameters. We find that MC-PDFT locates equilibrium geometries for several small- to medium-sized organic molecules that are similar to those located by complete active space second-order perturbation theory but that are obtained with decreased computational cost
