    6'-Methoxy Raloxifene-analog enhances mouse bone properties with reduced estrogen receptor binding

    Raloxifene (RAL) is an FDA-approved drug used to treat osteoporosis in postmenopausal women. RAL suppresses bone loss primarily through its role as a selective estrogen receptor modulator (SERM). This hormonal estrogen therapy promotes unintended side effects, such as hot flashes and increased thrombosis risk, and prevents the drug from being used in some patient populations at-risk for fracture, including children with bone disorders. It has recently been demonstrated that RAL can have significant positive effects on overall bone mechanical properties by binding to collagen and increasing bone tissue hydration in a cell-independent manner. A Raloxifene-Analog (RAL-A) was synthesized by replacing the 6-hydroxyl substituent with 6-methoxy in effort to reduce the compound's binding affinity for estrogen receptors (ER) while maintaining its collagen-binding ability. It was hypothesized that RAL-A would improve the mechanical integrity of bone in a manner similar to RAL, but with reduced estrogen receptor binding. Molecular assessment showed that while RAL-A did reduce ER binding, downstream ER signaling was not completely abolished. In-vitro, RAL-A performed similarly to RAL and had an identical concentration threshold on osteocyte cell proliferation, differentiation, and function. To assess treatment effect in-vivo, wildtype (WT) and heterozygous (OIM+/-) female mice from the Osteogenesis Imperfecta (OI) murine model were treated with either RAL or RAL-A from 8 weeks to 16 weeks of age. There was an untreated control group for each genotype as well. Bone microarchitecture was assessed using microCT, and mechanical behavior was assessed using 3-point bending. Results indicate that both compounds produced analogous gains in tibial trabecular and cortical microarchitecture. While WT mechanical properties were not drastically altered with either treatment, OIM+/- mechanical properties were significantly enhanced, most notably, in post-yield properties including bone toughness. This proof-of-concept study shows promising results and warrants the exploration of additional analog iterations to further reduce ER binding and improve fracture resistance

    Fermi point in graphene as a monopole in momentum space

    We consider the effective field theory of graphene monolayer with the Coulomb interaction between fermions taken into account. The gauge field in momentum space is introduced. The position of the Fermi point coincides with the position of the corresponding monopole. The procedure of extracting such monopoles during lattice simulations is suggested.Comment: Latex, 12 page

    Interface relaxation in electrophoretic deposition of polymer chains: Effects of segmental dynamics, molecular weight, and field

    Using different segmental dynamics and relaxation, characteristics of the interface growth is examined in an electrophoretic deposition of polymer chains on a three (2+1) dimensional discrete lattice with a Monte Carlo simulation. Incorporation of faster modes such as crankshaft and reptation movements along with the relatively slow kink-jump dynamics seems crucial in relaxing the interface width. As the continuously released polymer chains are driven (via segmental movements) and deposited, the interface width WW grows with the number of time steps tt, Wtβ,W \propto t^{\beta}, (β0.4\beta \sim 0.4--0.8)0.8), which is followed by its saturation to a steady-state value WsW_s. Stopping the release of additional chains after saturation while continuing the segmental movements relaxes the saturated width to an equilibrium value (WsWrW_s \to W_r). Scaling of the relaxed interface width WrW_r with the driving field EE, WrE1/2W_r \propto E^{-1/2} remains similar to that of the steady-state WsW_s width. In contrast to monotonic increase of the steady-state width WsW_s, the relaxed interface width WrW_r is found to decay (possibly as a stretched exponential) with the molecular weight.Comment: 5 pages, 7 figure

    Sum Rules for the Optical and Hall Conductivity in Graphene

    Graphene has two atoms per unit cell with quasiparticles exhibiting the Dirac-like behavior. These properties lead to interband in addition to intraband optical transitions and modify the ff-sum rule on the longitudinal conductivity. The expected dependence of the corresponding spectral weight on the applied gate voltage VgV_g in a field effect graphene transistor is constVg3/2\sim {const}- |V_g|^{3/2}. For Vg=0V_g =0, its temperature dependence is T3T^3 rather than the usual T2T^2. For the Hall conductivity, the corresponding spectral weight is determined by the Hall frequency ωH\omega_H which is linear in the carrier imbalance density ρ\rho, and hence proportional to VgV_g, and is different from the cyclotron frequency for Dirac quasiparticles.Comment: 16 pages, RevTeX4, 4 EPS figures; v2: to match PRB versio
