    Structural stability of adenylate kinase from the sulfate-reducing bacteria Desulfovibrio gigas

    Biophysical Chemistry 110 (2004) 83–92A novel adenylate kinase (AK) has recently been purified from Desulfovibrio gigas and characterized as a Co2+/Zn2+-containing enzyme: this is an unusual characteristic for AKs from Gram-negative bacteria, in which these enzymes are normally devoid of metals. Here, we studied the conformational stability of holo- and apo-AK as a function of temperature by differential scanning calorimetry (DSC), circular dichroism (CD), and intrinsic fluorescence spectroscopy. The thermal unfolding of AK is a cooperative two-state process, and is sufficiently reversible in the 9–11 pH range, that can be correctly interpreted in terms of a simple two-state thermodynamic model. The spectral parameters as monitored by ellipticity changes in the CD spectra of the enzyme as well as the decrease in tryptophan intensity emission upon heating were seen to be good complements to the highly sensitive but integral DSC-method.supported in part by the Fundação para a Ciência e a Tecnologia, Portugal, fellowships BD/13775/97 to OYG, BPD/3518/00 to SAB, SFRH/BD/1067/2000 to DGP, and NATO Scientific Programme Fellowships for Spain, call 2002 to GGZ

    Thermostability of cardosin A from Cynara cardunculus L.

    The structural stability of cardosin A, a plant aspartic proteinase (AP) from Cynara cardunculus L., has been investigated by high-sensitivity differential scanning calorimetry, intrinsic fluorescence and circular dichroism spectroscopy, and enzymatic activity assays. Even though the thermal denaturation of cardosin A is partially irreversible, valid thermodynamic data can be obtained within a wide pH region. Also, although cardosin A is a heterodimeric enzyme its thermal denaturation occurs without simultaneous dissociation to unfolded monomers. Moreover, in the 3-7 pH region the excess heat capacity can be deconvoluted into two components corresponding to two elementary two-state transitions, suggesting that the two polypeptide chains of cardosin A unfold independently. Detailed thermodynamic and structural investigations of cardosin A at pH=5.0, at which value the enzyme demonstrates maximum stability and enzymatic activity, revealed that after thermal denaturation the polypeptide chains of this protein retain most of their secondary structure motifs and are not completely hydrated.http://www.sciencedirect.com/science/article/B6THV-47P1SF6-4/1/edc14f851e47459fcd87b748d068439

    Structure, optical, electrical and thermoelectric properties of solution-processed Li-doped NiO films grown by SILAR

    The article presents a new facial synthesis of Li-doped NiO films (NiO:Li) via an easy and cost-effective method Successive Ionic Layer Adsorption and Reaction (SILAR) with the processing of the obtained NiO films in a lithium-containing aqueous solution for their transformation after annealing into NiO:Li layers. Comparative analysis of crystal structure, optical, electrical and thermoelectric properties of the obtained NiO and NiO:Li 420-1050 nm thick films have reveiled a cubic rock-salt NiO structure, at that, NiO:Li samples are nanocrystalline single phased Li-NiO solid solutions. The fabricated NiO and NiO:Li films are p-type semiconductors with activation energy Ea = 0.1 eV and Ea = 0.25‒0.31 eV, respectively. The obtained in-plane Seebeck coefficients Z are in the range 0.20–0.33 mV/К. Notwithstanding the fact that the maximum values of the thermoelectric power factors P=2.2 μW/K2·m, are rather small, they were achieved if the hot end of the NiO:Li film was heated only to 115 °C. Thus, the produced in this work new low cost thermoelectric thin film material is suitable for a production of electrical energy for low-power devices due to absorption of low-potential heat

    Due to its asymptomatic nature, orthostatic hypotension can be diagnosed only by means of orthostatic challenge. One should try to define the causes of orthostatic hypotension in every single case because treating illness that lead to orthostatic hypotension will improve patient’s health outcomes. The main target for therapy should be improving patient’s functional status, reducing orthostatic symptoms as well as risk for falls and syncopes, but not maintaining arterial blood pressure within certain limits. Doctors need to be aware of the methods for diagnosing the orthostatic hypotension to provide patients with the better quality of life.     Measurement of the e+eπ+πe^+e^-\to\pi^+\pi^- cross section from threshold to 1.2 GeV with the CMD-3 detector

    The cross section of the process e+eπ+πe^+e^-\to\pi^+\pi^- has been measured in the center of mass energy range from 0.32 to 1.2 GeV with the CMD-3 detector at the electron-positron collider VEPP-2000. The measurement is based on a full dataset collected below 1 GeV during three data taking seasons, corresponding to an integrated luminosity of about 62 pb1^{-1}. In the dominant ρ\rho-resonance region, a systematic uncertainty of 0.7% has been reached. At energies around ϕ\phi-resonance the π+π\pi^+\pi^- production cross section was measured for the first time with high energy resolution. The forward-backward charge asymmetry in the π+π\pi^+\pi^- production has also been measured. It shows the strong deviation from the theoretical prediction based on conventional sQED framework and is in good agreement with GVDM and dispersive-based predictions.     The cross section of the process e+eπ+πe^+e^-\to\pi^+\pi^- has been measured in the center of mass energy range from 0.32 to 1.2 GeV with the CMD-3 detector at the electron-positron collider VEPP-2000. The measurement is based on an integrated luminosity of about 88 pb1^{-1} out of which 62 pb1^{-1} constitutes a full dataset collected by CMD-3 at center-of-mass energies below 1 GeV. In the dominant region near ρ\rho-resonance a systematic uncertainty of 0.7% has been reached.     Christian–Weber disease refers to the group of panniculitis, which present an inflammation of the subcutaneous adipose tissue with the involvement of neutrophils, white blood cells and histiocytics, which leads to fibrosis in adipose tissue and sometimes to granulomatous changes. Clinically this pathology manifests from the formation of nodes in the subcutaneous adipose tissue, which appear suddenly, sporadically in some areas or multiple, sometimes forming large aggregations (plaques) or strands, shaped like a rosary. Consistency of nodes initially soft, then compacted, can form adhesions with the skin, the skin over them is not changed initially, followed by a color change from bright pink to brown-purple. Ends the process of atrophy of the affected area, sometimes with a depression in the center. Localization sites in the body are different; they are placed often on the buttocks, arms and legs. The duration of the process of the skin varies from a few days to a few months, after involution of old nodes may appear new ones, it are the cause of increasing the duration of the disease, having a relapsing course. It can be rather difficult to do differential diagnosis, which requires a great deal of experience of experts and a team approach to the study and treatment of this pathology. To confirm the diagnosis requires a biopsy of the skin. Prognosis depends on the nature of the disease and timely diagnosis.Болезнь Крисчена–Вебера относится к группе панникулитов, которые представляют собой воспаление подкожно-жировой клетчатки с вовлечением нейтрофилов, лейкоцитов и гистиоцитов, приводящее к фиброзу и иногда к гранулематозным изменениям. Клинически данная патология проявляется формированием узлов в подкожно-жировой клетчатке, которые возникают внезапно, единично или множественно, иногда образуя крупные скопления (бляшки) или тяжи, по форме напоминающие четки. Консистенция узлов вначале мягкая, затем они уплотняются, могут образовываться спайки с кожей, цвет кожных покровов над ними первоначально не изменен, но в последствии меняется от ярко-розового до коричнево-багрового. Заканчивается данный процесс атрофией пораженного участка, нередко с вдавлением в центре. Локализация узлов на теле различна, чаще они располагаются на ягодицах, руках и ногах. Длительность кожного процесса варьируется от нескольких дней до нескольких месяцев, при инволюции старых узлов могут появляться новые, что служит причиной увеличения продолжительности самого заболевания, имеющего рецидивирующее течение. Достаточно сложной может быть дифференциальная диагностика, требующая большого опыта специалистов и коллективного подхода к изучению и лечению данной патологии. Для подтверждения диагноза необходима биопсия кожи. Прогноз зависит от характера течения заболевания и своевременно установленного диагноза
