    Design aspects of a CMC coating-like system for hot surfaces of aero engine components

    Ceramic Matrix Composite (CMC) is an emerging material system that can be a game changer in the aerospace industry, both civil and military. CMCs components are, in fact, lighter and less prone to fatigue failure in a high temperature environment. However, at high temperatures, the diffusion of oxygen and water vapour inside the CMC can have detrimental effects. Therefore, the presence of protective coating is necessary to extend the life of CMC components. In the present work, a three-layers coating, consisting of a silicon bond (BND), adhesively bonded to the CMC, an Environment Barrier Coating (EBC) and a softer layer 3 (LAY3), is investigated for a CMC component. An aero-engine high pressure turbine seal segment was considered. Two design aspects are covered: (i) creep law is determined and calibrated in environment Abaqus from the experimental data of each coating layer available in the open literature, to provide a suitable instrument for the creep relaxation analyses of hot components; (ii) thickness sensitivity study of each layer of the coating is conducted to minimise the interface stresses of coating with substrate in order to mitigate cracking and removal/spalling phenomena when exposed to temperature gradients and to increase their service life. These two different aspects are combined together to predict the coating stress field as a function of service time

    Coupled FEM-DBEM Simulation of 3D Crack Growth under Fatigue Load Spectrum

    Abstract Numerical predictions of fatigue crack growth under load spectrum are obtained by coupled FEM-DBEM approach. An initial part-through corner crack, in a pre-notched specimen undergoing a traction fatigue load, propagates becoming through the thickness. A two parameter crack growth law ("Unified Approach") is calibrated by in house made constant amplitude experimental tests and the crack growth retardation after an overload application is reproduced. The residual stresses responsible for such retardation are calculated by a sequence of elastic-plastic static FEM analysis; such stresses are then applied to the crack faces for the propagation simulation in a DBEM environment. A satisfactory agreement between numerical and experimental crack growth rates are displayed, for both part-through crack and through the thickness crack. This approach provide general modeling capabilities, with allowance for general crack front shape and fully automatic propagation

    Experimental investigation on flow boiling heat transfer and pressure drop of refrigerants R32 and R290 in a stainless steel horizontal tube

    The purpose of this paper is to present new flow boiling heat transfer and pressure drop data in a single, horizontal smooth stainless steel tube of 6.0 mm internal diameter, in which R32 and R290 (propane) are employed as working fluids. The cross sectional average heat transfer coefficients are obtained by measuring the temperatures at the top, bottom, left and right sides of the channel. The experimental trends are analyzed for different operating conditions in terms of mass velocity (from 150 to 300 kg/(m2 s)) and heat flux (from 10 to 40 kW/m2). The saturation temperature is fixed to 25 °C for the heat transfer data and to 25 and 35 °C for the pressure drop experiments. The effects of the operative parameters and of the working fluids on local heat transfer coefficients and frictional pressure drop are discussed and the experimental data are finally compared with some of the available correlations taken from scientific literature

    FEM Substructuring for the Vibrational Characterization of a Petrol Engine

    In this work the vibrational behavior of a 4-cylinder, 4-stroke, petrol engine has been simulated by leveraging on a reduced modelling strategy, based on the Component Mode Synthesis (CMS), adopted to reduce the size of the full FEM model of the engine. The FEM model of the engine, comprising all of its sub-components, has been preliminary characterized from the vibrational standpoint; subsequently, the CMS has been adopted in order to reduce the FEM model size. Frequency Response Function (FRF) analyses have been used to identify the resonant frequencies and mode shapes of the different FEM models, and the so-obtained results have been compared showing a very good agreement. The reduced model has been able to reproduce with a high accuracy the vibration response at the engine mounts. The adopted reduced modelling strategy turned out to be effective in lowering the computational burden, keeping, at the same time, an accurate replication of the engine vibrational behavior. Runtimes have been significantly reduced from 24 hours for the full FEM model to less than 2 hours for the reduced model

    Prevention of Dermatologic Side Effects of Bimatoprost 0.03% Topical Therapy

    PURPOSE: To investigate the efficacy of reducing the drop-skin contact to prevent dermatologic side effects of bimatoprost 0.03% topical therapy. DESIGN: Prospective, randomized, single-blinded, internally controlled study. METHODS: Enrolled subjects started bimatoprost 0.03% therapy once at night in both eyes and were instructed to wipe selectively only one eye (eye 1) with an adsorbent pad during and after drops administration for four months. The fellow eye acted as the internal control. Eyelash growth, regional skin hypertrichosis, and pigmentation on the periocular skin were assessed at baseline and during the four months of follow-up. RESULTS: A lower incidence of eyelash growth and skin pigmentation in the inferonasal pericanthal region were observed in eye 1. The incidence of pigmentation in the inferotemporal skin region and skin hypertrichosis were similar in the two eyes. CONCLUSION: The reduction of the drop,skin contact affects the regional incidence and the extent of dermatologic skin changes that are related to bimatoprost 0.03% topical therapy

    Passive noise control oriented design of aircraft headrests

    Two Passive Noise Control (PNC) concepts were numerically evaluated in terms of their impact on the Sound Pressure Level (SPL) perceived by passengers of an aircraft flight. A concept was based on the shape optimization of the headrests, whereas the second one was based on the adoption of a high absorbing material, i.e. a nanofiber textile, to improve the acoustic performances of the headrests. To this aim, an aircraft seat was modelled with the Boundary Element Method (BEM) and loaded with a spherical distribution of monopole sources surrounding the seat. Different configurations of headrest shape and covering textiles were then compared in terms of the SPL calculated at passengers’ ears. The work shows how an acoustic-oriented design of the aircraft headrests could achieve an average SPL reduction for passengers up to 3 dBA

    Introduction: the PROVE-IT (“Intensive versus moderate lipid lowering with statins after acute coronary syndromes”) was a comparison of pravastatin 40 mg/die versus atorvastatin 80 mg/die in patients with an acute coronary syndrome (ACS). Aim: our aim was to investigate the economic consequence of high dose of atorvastatin vs usual-dose of pravastatin in Italian patients with a history of acute coronary syndrome. Methods: the analysis is conducted on the basis of clinical outcomes of the PROVE-IT study. We conducted a cost-effectiveness analysis, comparing high dose of atorvastatin (80 mg/die) versus usual-dose of pravastatin (40 mg/die) in the perspective of the Italian National Health Service. We identified and quantified medical costs: drug costs according to the Italian National Therapeutic Formulary and hospitalizations were quantified based on the Italian National Health Service tariffs (2006). Effects were measured in terms of mortality and morbidity reduction (number of deaths, life years gained and frequency of hospitalizations). We considered an observation period of 24 months. The costs borne after the first 12 months were discounted using an annual rate of 3%. We conducted one and multi-way sensitivity analyses on unit cost and effectiveness. We also conducted a threshold analysis. Results: the cost of pravastatin or atorvastatin therapy over the 2 years period amounted to approximately 1.3 millions euro and 870,000 euro per 1,000 patients respectively. Atorvastatin was more efficacious compared to pravastatin and the overall cost of care per 1,000 patients over 24 months of follow-up was estimated at 3.2 millions euro in the pravastatin and 2.5 millions euro in the atorvastatin group, resulting into a cost saving of about 700,000 euro that is 27% of total costs occurred in the pravastatin group. Discussion: this study demonstrates that high does atorvastatin treatment leads to a reduction of direct costs for the National Health System. Atorvastatin therapy is dominant since it is both less costly and more effective than pravastatin. Results of sensitivity analysis showed that atorvastatin therapy remains dominant even in the most unfavourable hypotheses

    Pseudo-dipeptide bearing α,α-difluoromethyl ketone moiety as electrophilic warhead with activity against coronaviruses

    The synthesis of α-fluorinated methyl ketones has always been challenging. New methods based on the homologation chemistry via nucleophilic halocarbenoid transfer, carried out recently in our labs, allowed us to design and synthesize a target-directed dipeptidyl α,α-difluoromethyl ketone (DFMK) 8 as a potential antiviral agent with activity against human coronaviruses. The ability of the newly synthesized compound to inhibit viral replication was evaluated by a viral cytopathic effect (CPE)-based assay performed on MCR5 cells infected with one of the four human coronaviruses associated with respiratory distress, i.e., hCoV-229E, showing antiproliferative activity in the micromolar range (EC50 = 12.9 ± 1.22 µM), with a very low cytotoxicity profile (CC50 = 170 ± 3.79 µM, 307 ± 11.63 µM, and 174 ± 7.6 µM for A549, human embryonic lung fibroblasts (HELFs), and MRC5 cells, respectively). Docking and molecular dynamics simulations studies indicated that 8 efficaciously binds to the intended target hCoV-229E main protease (Mpro). Moreover, due to the high similarity between hCoV-229E Mpro and SARS-CoV-2 Mpro, we also performed the in silico analysis towards the second target, which showed results comparable to those obtained for hCoV-229E Mpro and promising in terms of energy of binding and docking pose

    Hedgehog-GLI signalling promotes chemoresistance through the regulation of ABC transporters in colorectal cancer cells

    Colorectal cancer (CRC) is a leading cause of cancer death. Chemoresistance is a pivotal feature of cancer cells leading to treatment failure and ATP-binding cassette (ABC) transporters are responsible for the efflux of several molecules, including anticancer drugs. The Hedgehog-GLI (HH-GLI) pathway is a major signalling in CRC, however its role in chemoresistance has not been fully elucidated. Here we show that the HH-GLI pathway favours resistance to 5-fluorouracil and Oxaliplatin in CRC cells. We identified potential GLI1 binding sites in the promoter region of six ABC transporters, namely ABCA2, ABCB1, ABCB4, ABCB7, ABCC2 and ABCG1. Next, we investigated the binding of GLI1 using chromatin immunoprecipitation experiments and we demonstrate that GLI1 transcriptionally regulates the identified ABC transporters. We show that chemoresistant cells express high levels of GLI1 and of the ABC transporters and that GLI1 inhibition disrupts the transporters up-regulation. Moreover, we report that human CRC tumours express high levels of the ABCG1 transporter and that its expression correlates with worse patients’ prognosis. This study identifies a new mechanism where HH-GLI signalling regulates CRC chemoresistance features. Our results indicate that the inhibition of Gli1 regulates the ABC transporters expression and therefore should be considered as a therapeutic option in chemoresistant patients
