Synthesis, reactivity studies, and cytotoxicity of two trans-Iodidoplatinum(II) complexes. Does photoactivation work?

trans-Platinum complexes have been the landmark in unconventional drugs prompting the development of innovative structures that might exhibit chemical and biological profiles different to cisplatin. Iodido complexes signaled a new turning point in the platinum drug design field when their cytotoxicity was reevaluated and reported. In this new study, we have synthesized and evaluated diodidoplatinum complexes trans-[PtI2(amine)(pyridine)] bearing aliphatic amines (isopropylamine and methylamine) and pyridines in trans configuration. X-ray diffraction data support the structural characterization. Their cytotoxicity has been evaluated in tumor cell lines such as SAOS-2, A375, T-47D, and HCT116. Moreover, we report their solution behavior and reactivity with biological models. Ultraviolet-a (UVA) irradiation induces an increase in their reactivity towards model nucleobase 5′-GMP in early stages, and promotes the release of the pyridine ligand (spectator ligand) at longer reaction times. Density Functional calculations have been performed and the results are compared with our previous studies with other iodido derivatives.MINECO CTQ-2015-68779

Bgs4 is essential for cytokinesis and cell growth

[EN]Schizosaccharomyces pombe contains four putative (1,3)β-D-glucan synthase (GS) catalytic subunits, Bgs1p to Bgs4p. In this work, we cloned bgs4+ and show that Bgs4p is the only subunit 1) essential for maintaining cell integrity during both cytokinesis and polarized growth, and 2) found to be part of the GS enzyme. Here we show that bgs4+, cwg1+ (cwg1-1 shows reduced cell-wall β-glucan and GS catalytic activity) and orb11+ (orb11-59 is defective in cell morphogenesis) are the same gene. bgs4+ is essential during spore germination. bgs4+ shut-off produces cell lysis at growing poles and mainly at the septum prior to cytokinesis, suggesting that Bgs4p is essential for cell wall growth and for compensating an excess of cell wall degradation during cytokinesis. Shut-off and overexpression analysis suggest that 1) Bgs4p forms part of a GS catalytic multiprotein complex, and 2) Bgs4p-promoted cell-wall β-glucan alterations induce compensatory mechanisms from other Bgs subunits and (1,3)α-D-glucan synthase. Physiological localization studies showed that Bgs4p localizes to the growing ends, the medial ring and septum, and in each process of wall synthesis or remodeling that occurs during sexual differentiation: mating, zygote and spore formation, and spore germination. Bgs4p timing and requirements for proper positioning during cytokinesis and its localization pattern during spore maturation differ from those of Bgs1p. Bgs4p localizes overlapping the contractile ring once Bgs1p is present and a Calcofluor white-stained septum material is detected, suggesting that Bgs4p is involved in a late process of secondary or general septum synthesis. Unlike Bgs1p, Bgs4p needs the medial ring but not the Septation Initiation Network proteins to localize with the other septation components. Furthermore, Bgs4p localization depends on the polarity establishment proteins. Finally, F-actin is necessary for Bgs4p delocalization from and relocalization to the growing regions, but it is not needed for its stable maintenance at the growing sites, poles and septum. All these data show for the first time an essential role for a Bgs subunit in the synthesis of a (1,3)β-D-glucan necessary to preserve cell integrity when cell wall synthesis or repair are needed

On negative flows of the AKNS hierarchy and a class of deformations of bihamiltonian structure of hydrodynamic type

A deformation parameter of a bihamiltonian structure of hydrodynamic type is shown to parameterize different extensions of the AKNS hierarchy to include negative flows. This construction establishes a purely algebraic link between, on the one hand, two realizations of the first negative flow of the AKNS model and, on the other, two-component generalizations of Camassa-Holm and Dym type equations. The two-component generalizations of Camassa-Holm and Dym type equations can be obtained from the negative order Hamiltonians constructed from the Lenard relations recursively applied on the Casimir of the first Poisson bracket of hydrodynamic type. The positive order Hamiltonians, which follow from Lenard scheme applied on the Casimir of the second Poisson bracket of hydrodynamic type, are shown to coincide with the Hamiltonians of the AKNS model. The AKNS Hamiltonians give rise to charges conserved with respect to equations of motion of two-component Camassa-Holm and two-component Dym type equations.Comment: 20 pages, Late

Automated calibration of FEM models using LiDAR point clouds

In present work it is pretended to estimate elastic parameters of beams through the combined use of precision geomatic techniques (laser scanning) and structural behaviour simulation tools. The study has two aims, on the one hand, to develop an algorithm able to interpret automatically point clouds acquired by laser scanning systems of beams subjected to different load situations on experimental tests; and on the other hand, to minimize differences between deformation values given by simulation tools and those measured by laser scanning. In this way we will proceed to identify elastic parameters and boundary conditions of structural element so that surface stresses can be estimated more easily.Ministerio de Interior | Ref. Vertex Operators and Soliton Solutions of Affine Toda Model with U(2) Symmetry

The symmetry structure of non-abelian affine Toda model based on the coset SL(3)/SL(2)U(1)SL(3)/SL(2)\otimes U(1) is studied. It is shown that the model possess non-abelian Noether symmetry closing into a q-deformed SL(2)U(1)SL(2)\otimes U(1) algebra. Specific two vertex soliton solutions are constructed.Comment: 17 pages, latex, misprints corrected, version to appear in J.Phys
