    Characteristics of P2X7 receptors from human B lymphocytes expressed in Xenopus oocytes

    AbstractHuman B lymphocytes express an ATP-gated ion channel (P2Z receptor), which shares similarities with the recently identified P2X7 receptor. Using gene specific primers, we have now isolated P2X7 cDNA from the total RNA of human B lymphocytes. This hP2X7 receptor subtype was expressed in Xenopus oocytes and electrophysiologically characterized. The hP2X7 receptor is similar to, but does not completely match, P2Z of human B cells. The hP2X7 receptors resemble the P2Z receptors with regard to the ATP concentration of half maximal activation, reproducibility, permeation characteristics and lack of desensitization of the ATP-evoked currents. However, in contrast to the native lymphocytic P2Z receptor, the time course of activation of hP2X7 displayed an additional linearly increasing current component. Furthermore, a second, small and slowly deactivating current component exists only in hP2X7 expressed in oocytes. The activation and deactivation kinetics as well as permeation characteristics of hP2X7 are different from rat P2X7 recently expressed in oocytes.     Toll‐like receptors (TLRs) build the first barrier in the innate immune response and therefore represent promising targets for the modulation of inflammatory processes. Recently, the pyrogallol‐containing TLR2 antagonists CU‐CPT22 and MMG‐11 were reported; however, their 1,2,3‐triphenol motif renders them highly susceptible to oxidation and excludes them from use in extended experiments under aerobic conditions. Therefore, we have developed a set of novel TLR2 antagonists (1 –9 ) based on the systematic variation of substructures, linker elements, and the hydrogen‐bonding pattern of the pyrogallol precursors by using chemically robust building blocks. The novel series of chemically stable and synthetically accessible TLR2 antagonists (1 –9 ) was pharmacologically characterized, and the potential binding modes of the active compounds were evaluated structurally. Our results provide new insights into structure‐activity relationships and allow rationalization of structural binding characteristics. Moreover, they support the hypothesis that this class of TLR ligands bind solely to TLR2 and do not directly interact with TLR1 or TLR6 of the functional heterodimer. The most active compound from this series (6 ), is chemically stable, nontoxic, TLR2‐selective, and shows a similar activity with regard to the pyrogallol starting points, thus indicating the variability of the hydrogen bonding pattern

    Occlusive dressing-induced secretomes influence the migration and proliferation of mesenchymal stem cells and fibroblasts differently

    Background: Fingertip injuries treated with occlusive dressings (ODs) lead to nearly scar-free, functionally, and aesthetically pleasing results. We hypothesized that paracrine factors in the wound fluid (secretome) may influence migration and proliferation of mesenchymal stem cells (MSCs) and fibroblasts and modulate the wound-healing process. Methods: We could collect wound fluid samples from 4 fingertip injuries and 7 split skin donor sites at the 5th day during dressing change. Blood serum samples served as controls. The proliferation rate of MSCs and fibroblasts (HS27) was continuously measured through impedance analysis for 60h and by Alamarblue analysis after 72h. Cell migration was evaluated continuously for 15h and confirmed by the in vitro wound-healing assay. Results: Migration of MSCs under the influence of both wound fluids was significantly faster than controls from 4 to 6h after incubation and reversed after 9h. MSC proliferation in wound fluid groups showed a significant increase at 5 and 10h and was significantly decreased after 45h. Fibroblasts in wound fluid groups showed overall a significant increase in migration and a significant decrease in proliferation compared to controls. Conclusion: OD-induced secretomes influence MSCs and fibroblasts and thereby possibly modulate wound healing and scar tissue formation

    Das neue Schulfach Digitale Grundbildung (DGB) wurde als Pflichtfach in der Sekundarstufe I mit überarbeitetem Lehrplan verankert – ein Grund zur Freude, dass der Digitalisierung auch innerhalb der schulische Bildung endlich die nötige Aufmerksamkeit gegeben wird. Das Fach stellt, besonders vor dem Hintergrund der Initiativen, die im Rahmen des 8-Punkte Plans der Bundesregierung gesetzt wurden, die notwendige Brücke zwischen Infrastruktur-Maßnahmen, Inhalten und der tatsächlichen Nutzung dieser Ressourcen dar. Das neue Schulfach Digitale Grundbildung – eine Projektionsfläche für mannigfaltige Bedürfnislagen. Aus Informatikperspektive und Sicht der Wirtschaft bietet es eine Gelegenheit, kompetenzorientierten Informatikunterricht schon in der Sekundarstufe I beginnen zu lassen. Aus medienpädagogischem Betrachtungswinkel ist das Fach ein lang gewünschtes Gefäß, um jungen Menschen ein besseres Verständnis von Medien und eine kompetente Mediennutzung nahezubringen. Aus der im Folgenden abgebildeten Perspektive des Zentrums für Lehrer*innenbildung der Universität Wien bietet das Schulfach den notwendigen Rahmen, um zumindest ansatzweise inklusive, diversitätsgerechte und interdisziplinäre Perspektiven auf die Digitalisierung und ihre sozialen, kulturellen, politschen und gesellschaftlichen Aus- und Wechselwirkungen zusammenzuführen

    The fundamental left-right asymmetry in the Germanic verb cluster

    Cinque (2005, 2009, 2014a) observes that there is an asymmetry in the possible ordering of dependents of a lexical head before versus after the head. A reflection on some of the concepts needed to develop Cinque’s ideas into a theory of neutral word order reveals that dependents need to be treated separately by class. The resulting system is applied to the problem of word order in the Germanic verb cluster. It is shown that there is an extremely close match between theoretically derived expectations for clusters made up of auxiliaries, modals, causative ‘let’, a main verb, and verbal particles. The facts point to the action of Cinque’s fundamental left-right asymmetry in language in the realm of the verb cluster. At the same time, not all verb clusters fall under Cinque’s generalization, which, therefore, argues against treating all cases of restructuring uniformly
