    Regulatory effects of PRF and titanium surfaces on cellular adhesion, spread, and cytokine expressions of gingival keratinocytes

    Dental implant material has an impact on adhesion and spreading of oral mucosal cells on its surface. Platelet-rich fibrin (PRF), a second-generation platelet concentrate, can enhance cell proliferation and adhesion. The aim was to examine the regulatory effects of PRF and titanium surfaces on cellular adhesion, spread, and cytokine expressions of gingival keratinocytes. Human gingival keratinocytes were cultured on titanium grade 4, titanium grade 5 (Ti5), and HA discs at 37 °C in a CO2 incubator for 6 h and 24 h, using either elutes of titanium-PRF (T-PRF) or leukocyte and platelet-rich fibrin (L-PRF), or mammalian cell culture medium as growth media. Cell numbers were determined using a Cell Titer 96 assay. Interleukin (IL)-1β, IL-1Ra, IL-8, monocyte chemoattractant protein (MCP)-1, and vascular endothelial growth factor (VEGF) expression levels were measured using the Luminex® xMAP™ technique, and cell adhesion and spread by scanning electron microscopy. Epithelial cell adhesion and spread was most prominent to Ti5 surfaces. L-PRF stimulated cell adhesion to HA surface. Both T-PRF and L-PRF activated the expressions of IL-1 β, IL-8, IL-1Ra, MCP-1, and VEGF, T-PRF being the strongest activator. Titanium surface type has a regulatory role in epithelial cell adhesion and spread, while PRF type determines the cytokine response.</p

    Increased mean platelet volume in patients with infective endocarditis and embolic events

    Background: Platelet activation appears to play an important role in thromboembolic com­plications of infective endocarditis (IE). Mean platelet volume (MPV) is a potentially useful marker of platelet activity and a quick and easy determinant of thrombotic risk. Hence the aim of this study was to investigate the baseline platelet volume indices (MPV and platelet distribution width [PDW]) in IE patients who developed embolic events in the follow-up period and who did not. Methods: The study group consisted of 76 consecutive patients (female: 55, male: 21, mean age: 26 years old, ranged: 8–64 years) with definite IE according to Duke Criteria. Thirty four healthy subjects, who were age and gender adjusted, served as the control group. The mean duration of hospital stay was 44 days. Results: Among the IE patients, 13 (13/76, 17.1%) had major embolic events. Significantly larger vegetations were observed in patients with embolic events as compared to non-embolic group (1.4 vs. 1.0 cm, p = 0.03). MPV at hospital admission was higher in patients who had embolic events in the follow-up period compared to both those who did not and the control subjects (10.62 ± 1.13 vs. 9.25 ± 0.97 and 8.93 ± 0.82 fL, p &lt; 0.001, respectively). Similarly, the patients with embolic events had increased PDW compared to the non-embolic ones and the control group (16.31 ± 2.42 vs. 14.35 ± 1.97 and 14.04 ± 1.82%, p &lt; 0.001, respectively). Conclusions: The present study demonstrated that IE patients with embolic events had in­creased MPV and PDW values, compared to non-embolics. Future prospective studies with standardized measurements may clarify the clinical role of platelet volume indices in thrombo­embolic complications of IE

    Periodontitis and peri-implantitis tissue levels of Treponema denticola-CTLP and its MMP-8 activating ability

    Background and Aims: Chymotrypsin-like-proteinase of Treponema denticola (Td-CTLP) can stimulate the protein expression and activation of matrix metalloproteinase (MMP)-8 (or collagenase-2), a potent tissue destructive enzyme from gingival cells in vitro. The aims of this study were 1) to demonstrate the proMMP-8 (or latent MMP-8) activation by Td-CTLP in vitro and 2) to detect Td-CTLP and MMP-8 protein levels in the tissue samples of peri-implantitis and periodontitis patients.Materials and Methods: proMMP-8 activation by Td-CTLP was analyzed by immunoblots. Tissue specimens were collected from 38 systemically healthy and non-smoking patients; 14 of whom had moderate to severe periodontitis, 10 of whom were suffering from peri-implantitis, and finally 14 of whom showed no sign of periodontal inflammation nor radiological bone decay (control group). The immune-expression levels of MMP-8 and Td-CTLP in the epithelium and the connective tissue were analyzed immunohistochemically. A pixel color-intensity analyze was performed with ImageJ software (version 1.46c; Rasband WS, National Institutes of Health, Bethesda, MD, USA) to obtain a comparable numeral score for each patient's epithelium and connective tissue MMP-8 and Td-CTLP enzyme level.Results: Td-CTLP activated proMMP-8 in vitro by converting the 70 75 kDa proMMP-8 to 65 kDa active MMP-8. Also, lower molecular size 25 50 kDa parts of MMP-8 were formed. There was no statistically significant difference between the study groups in terms of their MMP-8 and Td-CTLP levels in the epithelium or in the connective tissue.Conclusion: Regarding the limits of this study, it can thus be said that the Td-CTLP enzyme can activate the host proMMP-8 enzyme. Tissue protein levels of MMP-8 and Td-CTLP do not seem to be changed in peri-implantitis and in periodontitis.</p

    The frequency of Duchenne muscular dystrophy/Becker muscular dystrophy and Pompe disease in children with isolated transaminase elevation: results from the observational VICTORIA study

    IntroductionElevated transaminases and/or creatine phosphokinase can indicate underlying muscle disease. Therefore, this study aims to determine the frequency of Duchenne muscular dystrophy/Becker muscular dystrophy (DMD/BMD) in male children and Pompe disease (PD) in male and female children with isolated hypertransaminasemia.MethodsThis multi-center, prospective study enrolled patients aged 3–216 months with serum alanine transaminase (ALT) and/or aspartate transaminase (AST) levels &gt;2× the upper limit of normal (ULN) for ≥3 months. Patients with a known history of liver or muscle disease or physical examination findings suggestive of liver disease were excluded. Patients were screened for creatinine phosphokinase (CPK) levels, and molecular genetic tests for DMD/BMD in male patients and enzyme analysis for PD in male and female patients with elevated CPK levels were performed. Genetic analyses confirmed PD. Demographic, clinical, and laboratory characteristics of the patients were analyzed.ResultsOverall, 589 patients [66.8% male, mean age of 63.4 months (standard deviation: 60.5)] were included. In total, 251 patients (188 male and 63 female) had CPK levels above the ULN. Of the patients assessed, 47% (85/182) of male patients were diagnosed with DMD/BMD and 1% (3/228) of male and female patients were diagnosed with PD. The median ALT, AST, and CPK levels were statistically significantly higher, and the questioned neurological symptoms and previously unnoticed examination findings were more common in DMD/BMD patients than those without DMD/BMD or PD (p &lt; 0.001).DiscussionQuestioning neurological symptoms, conducting a complete physical examination, and testing for CPK levels in patients with isolated hypertransaminasemia will prevent costly and time-consuming investigations for liver diseases and will lead to the diagnosis of occult neuromuscular diseases. (İSDEMİR) fabrika sahasında bulunan; Yüksek Fırın Soba Bacası, Haddehane Bacaları, Su Kulesi ve Yakma Bacası gibi kule tipi yapıların dinamik davranışları incelenmiştir. Farklı standartlara göre inşa edilmiş yüksek yapıların gerçek davranışları ile yapı analiz program sonuçları karşılaştırılarak bu özel yapı türleri için literatürdeki boşluğu doldurmak amaçlanmıştır. Yapının mevcut dinamik davranışı tespit için yapı izleme sistemlerinden mevcut titreşim testi (ambient vibration) kullanılmıştır. Kule tipi yapılarda ivme ölçerler ile kayıt alınarak bilgisayara kaydedilmiştir. Analiz programları ile çözülen yapının dinamik karakteristik davranışları karşılaştırılmış ve mevcut veriler ışığında model kalibrasyonu yapılmıştır.İskenderun Iron and Steel Factory is one of the oldest iron and steel factory in The Town of İskenderun of Hatay, Turkey. The dynamic behavior of tower structures such as Blast Furnace Stove Chimney, Rolling Mill Chimney, Water Towers and Flare Tower in İSDEMİR was examined in this study. Structural analysis program results were compared with the actual behavior of tall structures which were built according to different codes. It’s aimed to fill the gap in the literature for this specific building types. Ambient vibration test which is the one of the structural monitoring system is used for determining the existing dynamic behavior of structure. Ambient vibrations are recorded on computer by the help of accelerometers. Dynamic characteristic behavior of the structure were analyzed with structural analysis programs and the results were compared with the behavior of the existing structure.     Investigation of effects of low level laser therapy in distraction osteogenesis

    WOS: 000456954700003PubMed ID: 29885471Purpose: The purpose of the study was to investigate the histopathological effects of low level laser therapy (LLLT) application in distraction osteogenesis (DO). Materials and Methods: Twenty adult female New Zealand white rabbits were included in the study. Under general anesthesia unilateral mandibular corpus osteotomy was performed. Custom made external distractors were placed to right mandibles of rabbits. After five days of latency period, distractors were activated once a day for 5 days with 1 mm/day frequency. Animals in the study group were exposed to LLLT from six different points transcutanously after each distractor activation. Control group was not exposed to laser irradiation. Animals were sacrificed after 15 and 30 days of consolidation periods and mandibles were processed for histopathological investigation under light microscope. Bone healing was analyzed with a semi-quantitative 4 point scale. Results: Osteoblastic activity and vascularization were found higher in the study group than control group after 15 days consolidation. Chondroblastic activity of the control group was significantly higher than the study group in both 15 and 30 days of consolidation groups. Osteoblastic activity and trabecular bone formation were found significantly higher in the study group than the control group after 15 days consolidation. Conclusions: The use of LLLT in activation period of distraction osteogenesis stimulates bone repair in the early stages of distraction osteogenesis by inducing intramembranous healing and less cartilage tissue formation in the bone callus.Marmara University Scientific Research Project Commission [SAG-A-060308-0025]This research was funded by Marmara University Scientific Research Project Commission with project number 'SAG-A-060308-0025'

    Depreme karşı güçlendirme çalışmalarında mevcut yapıların modellenebilmesi için tahribatlı testler büyük önem arz etmektedir. Mevcut yapı malzeme özellikleri modelleme çalışmalarının doğru sonuç vermesini sağlamaktadır. Bu çalışmada Rus standartlarına göre tasarlanmış ve 1974 yılında yapımı tamamlanmış olan pompa binası ele alınmıştır. Bina; denizden su alma yapısı, su kamaraları, pompa temelleri, betonarme taşıyıcı sistem ve çelik çatıdan teşkil edilmiştir. Mevcut yapı analizi için zemin sondaj çalışmaları yapılmış, dalgıç marifeti ile video ve fotoğraflar çekilmiş, karot numuneleri alınmış, donatıların örnekleme metodu ile çap ve adet kontrolleri yapılmıştır. Alınan ölçüm sonuçlarından hareketle yapının mevcut durumu paket programlar ile modellenerek analiz edilmiştir. Yapılan analizden elde edilen sonuçlara göre gerekli kesit artışları tespit edilmiş ve uygulanması gereken güçlendirmeler projelendirilerek gerekli yapı elemanlarında uygulanmıştırDestructive tests have much importance for modelling existing structures of the studies of the strengthening for the earthquake. Existing building materials properties provide the accuracy of the modeling study. In this study, The Pump House which was designed according to the Russian standarts and which was completed construction in 1974 is discussed. Sea Water Intake Buildings has been formed from waterpools, pumps, reinforced concrete system and steel roof. For analysis of the existing structure; ground drilling work was done, video and photos taken with divers ingenuity, core samples were taken and the pieces of rebar diameter were examined. The structure was modelled according to the results by using computer softwares. Obtained results from the analysis determined by increases in the required section. According to the results the necessary work to strengthen the structure were applied and described by prepared project
