    Phylogenetic analysis reveals an ancient gene duplication as the origin of the MdtABC efflux pump.

    The efflux pumps from the Resistance-Nodulation-Division family, RND, are main contributors to intrinsic antibiotic resistance in Gram-negative bacteria. Among this family, the MdtABC pump is unusual by having two inner membrane components. The two components, MdtB and MdtC are homologs, therefore it is evident that the two components arose by gene duplication. In this paper, we describe the results obtained from a phylogenetic analysis of the MdtBC pumps in the context of other RNDs. We show that the individual inner membrane components (MdtB and MdtC) are conserved throughout the Proteobacterial species and that their existence is a result of a single gene duplication. We argue that this gene duplication was an ancient event which occurred before the split of Proteobacteria into Alpha-, Beta- and Gamma- classes. Moreover, we find that the MdtABC pumps and the MexMN pump from Pseudomonas aeruginosa share a close common ancestor, suggesting the MexMN pump arose by another gene duplication event of the original Mdt ancestor. Taken together, these results shed light on the evolution of the RND efflux pumps and demonstrate the ancient origin of the Mdt pumps and suggest that the core bacterial efflux pump repertoires have been generally stable throughout the course of evolution

    Cu-Mg-Fe-O-(Ce) complex oxides as catalysts of selective catalytic oxidation of ammonia to dinitrogen (NH3-SCO)

    Multicomponent oxide systems 800-Cu-Mg-Fe-O and 800-Cu-Mg-Fe-O-Ce were tested as catalysts of selective catalytic oxidation of ammonia to dinitrogen (NH3-SCO) process. Materials were obtained by calcination of hydrotalcite-like compounds at temperature 800 degrees C. Some catalysts were doped with cerium by the wet impregnation method. Not only simple oxides, but also complex spinel-like phases were formed during calcination. The influence of chemical composition, especially the occurrence of spinel phases, copper loading and impregnation by cerium, were investigated. Materials were characterized by several techniques: X-ray diffraction (XRD), Fourier-transform infrared spectroscopy (FT-IR), low-temperature nitrogen adsorption (BET), cyclic voltammetry (CV), temperature programmed reduction (H-2-TPR), UV-vis diffuse reflectance spectroscopy and scanning electron microscopy (SEM). Examined oxides were found to be active as catalysts of selective catalytic oxidation of ammonia with high selectivity to N-2 at temperatures above 300 degrees C. Catalysts with low copper amounts (up to 12 wt %) impregnated by Ce were slightly more active at lower temperatures (up to 350 degrees C) than non-impregnated samples. However, when an optimal amount of copper (12 wt %) was used, the presence of cerium did not affect catalytic properties. Copper overloading caused a rearrangement of present phases accompanied by the steep changes in reducibility, specific surface area, direct band gap, crystallinity, dispersion of CuO active phase and Cu2+ accessibility leading to the decrease in catalytic activity.Web of Science102art. no. 15

    Effect of high-temperature exposure on the microstructure and mechanical properties of the Al5Ti5Co35Ni35Fe20 high-entropy alloy

    Effect of high-temperature exposure on the microstructure and mechanical properties of the Al5Ti5Co35Ni35Fe20 High-Entropy Alloy was studied. High-entropy alloys belong to the group of multi-principal element alloys composed of at least five elements with a concentration between 5 and 35 atomic pct. Despite the multicomponent composition, the goal is to achieve a simple solid solution. The alloy was exposed to a temperature range of 650-900 degrees C for 168 h (7 days), after hot rolling and air-quenching. After hot rolling the investigated alloy was not fully supersaturated. The application of air cooling caused gamma' precipitation, which was uniformly distributed within the microstructure. High-temperature exposure caused the growth of gamma' particles. At 700 degrees C and higher, additional continuous and discontinuous precipitation of gamma' was observed. Finally, the microstructure of the investigated alloy consisted of gamma matrix and three types of gamma' precipitates, i.e. spherical gamma' phases with bimodal distribution within the grains and elongated DP gamma' phases present at the grain boundaries. The highest mechanical properties were obtained after exposure to 650 degrees C, which was caused by gamma' precipitation.     Catalytic oxidation of ammonia over cerium-modified copper aluminium zinc mixed oxides

    Copper-containing mixed metal oxides are one of the most promising catalysts of selective catalytic oxidation of ammonia. These materials are characterized by high catalytic efficiency; however, process selectivity to dinitrogen is still an open challenge. The set of Cu-Zn-Al-O and Ce/Cu-Zn-Al-O mixed metal oxides were tested as catalysts of selective catalytic oxidation of ammonia. At the low-temperature range, from 250 & DEG;C up to 350 & DEG;C, materials show high catalytic activity and relatively high selectivity to dinitrogen. Samples with the highest Cu loading 12 and 15 mol.% of total cation content were found to be the most active materials. Additional sample modification by wet impregnation of cerium (8 wt.%) improves catalytic efficiency, especially N-2 selectivity. The comparison of catalytic tests with results of physicochemical characterization allows connecting the catalysts efficiency with the form and distribution of CuO on the samples' surface. The bulk-like well-developed phases were associated with sample activity, while the dispersed CuO phases with dinitrogen selectivity. Material characterization included phase composition analysis (X-ray powder diffraction, UV-Vis diffuse reflectance spectroscopy), determination of textural properties (low-temperature N-2 sorption, scanning electron microscopy) and sample reducibility analysis (H-2 temperature-programmed reduction).Web of Science1421art. no. 658

    FInvestigation of Hot-Dip Galvanising Influence on the Buckling Resistance of Steel Angles

    The buckling curves, which enable the prediction of buckling resistance of steel structural elements, are physically connected with their type of cross-section, initial out-of-straightness, and the magnitude of residual stresses due to the different manufacturing technologies. It is known that hot-dip galvanisation changes and relives residual stresses, but so far the current design provisions do not take directly into account the impact of hot-dip galvanising on the reduction of residual stresses, and thus the reduction the generalised imperfection in the Ayrton-Perry model. The paper presents the difference between the relative buckling resistance of steel angles with residual stresses resulting from hot rolling and the same elements in which the magnitude of residual stresses was decreased by the hot-dip galvanising process. Carried out tests and geometrically and materially non-linear analyses with imperfections (GMNIA) have shown that angles with residual stresses reduced by heat treatment caused by hot-dip galvanising have higher buckling resistance compared to those with residual stresses after hot rolling. This increase ranges from 2 to 7%. The analyses carried out confirmed that the predicted reduction factors exhibit values closer to the buckling curve ‘a’, but these values do not reach the ‘a0’ curve recommended by the EN 50341-1 standard

    Water wells play an increasingly important role in providing water for the civilian population all over the world. Like other engineering structures, wells are subject to ageing processes resulting in degradation, which is observed as a reduction in hydraulic efficiency throughout their lifespan. To date, it has been found that the ageing process of a well is determined by a number of factors. The mathematical description of this process can be simplified. Drawing on Jacob’s equation, this paper presents the course of the degradation process as a variable depending on operation time, well loss and flow rate. To apply the determined relationships in practice, simplifying assumptions were adopted, which make it possible to determine the moment of ageing compensations of the degradation processes. It was also demonstrated that the degradation process may be slowed down by the appropriate selection of initial operating parameters. The presented discussion highlights the significance of parameters α, δ and exponent β. The relation between hydraulic resistances in an aquifer and in the engineering structure is closely connected with these values. The presented arguments indicate that step drawdown tests provide the necessary information which allows tracking changes in the ageing processes occurring in the engineering structure. The analysis of the drawdown test results makes it possible to determine the moment when the necessary adjustments in the operating parameters of a water well should be performed. Eventually, it allows maintaining the high hydraulic efficiency of the intake and extending the lifespan of the well in accordance with the principle of sustainability

    Functional differentiation of antiporter-like polypeptides in complex I; a site-directed mutagenesis study of residues conserved in MrpA and NuoL but Not in MrpD, NuoM, and NuoN

    It has long been known that the three largest subunits in the membrane domain (NuoL, NuoM and NuoN) of complex I are homologous to each other, as well as to two subunits (MrpA and MrpD) from a Na+ /H+ antiporter, Mrp. MrpA and NuoL are more similar to each other and the same is true for MrpD and NuoN. This suggests a functional differentiation which was proven experimentally in a deletion strain model system, where NuoL could restore the loss of MrpA, but not that of MrpD and vice versa. The simplest explanation for these observations was that the MrpA and MrpD proteins are not antiporters, but rather single subunit ion channels that together form an antiporter. In this work our focus was on a set of amino acid residues in helix VIII, which are only conserved in NuoL and MrpA (but not in any of the other antiporter-like subunits.) and to compare their effect on the function of these two proteins. By combining complementation studies in B. subtilis and 23Na-NMR, response of mutants to high sodium levels were tested. All of the mutants were able to cope with high salt levels; however, all but one mutation (M258I/M225I) showed differences in the efficiency of cell growth and sodium efflux. Our findings showed that, although very similar in sequence, NuoL and MrpA seem to differ on the functional level. Nonetheless the studied mutations gave rise to interesting phenotypes which are of interest in complex I research
