    Yield loss of Oryza glaberrima caused by grain shattering under rainfed upland conditions

    The general perception of Oryza glaberrima as low yielding is a potential drawback to intraspecific breeding of this species. Yields of 21 O. glaberrima accessions were evaluated alongside four O. sativa check varieties under two conditions in which panicles were both covered by paper sacks to avoid grain shattering (potential yield plot) and also not covered (control plot) in a rainfed upland field of Benin in 2010 and 2011. Average potential yields of O. glaberrima with panicles covered were 2.42 and 3.65 t ha-1 in 2010 and 2011, respectively, while those of O. sativa were respectively 2.99 and 3.49 t ha-1. Although there was no significant difference between potential yield and control yield in O. sativa, 19 O. glaberrima accessions showed significant yield reduction by grain shattering — the average percentages of lost yield by shattering were 57.88% in 2010 and 66.76% in 2011. These results suggest that low yield of O. glaberrima is due to grain shattering and that its potential yield would not be inferior to O. sativa if the grain shattering problems were overcome. Two O. glaberrima accessions, TOG 12303 and TOG 9300, possessing strong resistance to grain shattering comparable to O. sativa checks, were identified.Keywords: Panicles, paper sacks, potential yield, yield reduction

    Study on Static Electrification of Palm Fatty Acid Ester (PFAE) Oil Using Mini Static Tester (Archive)

    Palm fatty acid ester (PFAE) oil is an alternative insulating liquid of power transformer that has good biodegradable properties. As a part of insulating system of power transformer, PFAE should also has a low risk in term of static electrification that may degrade electrical strength of solid insulation due to charge accumulation at its surface during oil circulating. Charge potential value could describe the possibility of insulating oil in involving on charge accumulation at the surface of solid insulation. This can be calculated from charge density through electrostatic charge tendency (ECT) and volume resistivity. This study using mini-static-tester to measure ECT since it is easy to reproduce the specimen and suitable for standardization. Since PFAE has higher polarity than mineral oil, PFAE has larger charges than mineral oil. Even though static charges are estimated to generate on the pressboard surface easily, generated charges are estimated to escape from pressboard surface because of its lower resistivity. Variation of charge density and volume resistivity of several types (i.e. PFAE and mineral) and conditions (i.e. with-additives, non-additives, unused, and aged) of oil will influence to its charge potentia

    This paper responds to ‘Funding international agricultural research and the need to be noticed: a case study of NERICA rice’ by Stuart Orr, James Sumberg, Olaf Erenstein and Andreas Oswald, published in this issue of Outlook on Agriculture. In summary, the article by Orr et al, based on an internal WARDA document written in November 2003 and augmented with results from Internet searches, is outdated and does not seem to be fair, objective or useful. We invite the authors to visit WARDA or any of its partners in Sub-Saharan Africa for evidence of the impact of NERICA varieties or the other improved varieties and technologies that have been developed and disseminated by WARDA in recent years

    A Cross-Species Analysis of a Mouse Model of Breast Cancer-Specific Osteolysis and Human Bone Metastases Using Gene Expression Profiling

    <p>Abstract</p> <p>Background</p> <p>Breast cancer is the second leading cause of cancer-related death in women in the United States. During the advanced stages of disease, many breast cancer patients suffer from bone metastasis. These metastases are predominantly osteolytic and develop when tumor cells interact with bone. <it>In vivo </it>models that mimic the breast cancer-specific osteolytic bone microenvironment are limited. Previously, we developed a mouse model of tumor-bone interaction in which three mouse breast cancer cell lines were implanted onto the calvaria. Analysis of tumors from this model revealed that they exhibited strong bone resorption, induction of osteoclasts and intracranial penetration at the tumor bone (TB)-interface.</p> <p>Methods</p> <p>In this study, we identified and used a TB microenvironment-specific gene expression signature from this model to extend our understanding of the metastatic bone microenvironment in human disease and to predict potential therapeutic targets.</p> <p>Results</p> <p>We identified a TB signature consisting of 934 genes that were commonly (among our 3 cell lines) and specifically (as compared to tumor-alone area within the bone microenvironment) up- and down-regulated >2-fold at the TB interface in our mouse osteolytic model. By comparing the TB signature with gene expression profiles from human breast metastases and an <it>in vitro </it>osteoclast model, we demonstrate that our model mimics both the human breast cancer bone microenvironment and osteoclastogenesis. Furthermore, we observed enrichment in various signaling pathways specific to the TB interface; that is, TGF-β and myeloid self-renewal pathways were activated and the Wnt pathway was inactivated. Lastly, we used the TB-signature to predict cyclopenthiazide as a potential inhibitor of the TB interface.</p> <p>Conclusion</p> <p>Our mouse breast cancer model morphologically and genetically resembles the osteoclastic bone microenvironment observed in human disease. Characterization of the gene expression signature specific to the TB interface in our model revealed signaling mechanisms operative in human breast cancer metastases and predicted a therapeutic inhibitor of cancer-mediated osteolysis.</p

    Conjugated docosahexaenoic acid suppresses KPL-1 human breast cancer cell growth in vitro and in vivo: potential mechanisms of action

    Introduction The present study was conducted to examine the effect of conjugated docosahexaenoic acid (CDHA) on cell growth, cell cycle progression, mode of cell death, and expression of cell cycle regulatory and/or apoptosis-related proteins in KPL-1 human breast cancer cell line. This effect of CDHA was compared with that of docosahexaenoic acid (DHA). Methods KPL-1 cell growth was assessed by colorimetric 3- (4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay; cell cycle progression and mode of cell death were examined by flow cytometry; and levels of expression of p53, p21Cip1/Waf1, cyclin D1, Bax, and Bcl-2 proteins were examined by Western blotting analysis. In vivo tumor growth was examined by injecting KPL-1 cells subcutaneously into the area of the right thoracic mammary fat pad of female athymic mice fed a CDHA diet. Results CDHA inhibited KPL-1 cells more effectively than did DHA (50% inhibitory concentration for 72 hours: 97 μmol/l and 270 μmol/l, respectively). With both CDHA and DHA growth inhibition was due to apoptosis, as indicated by the appearance of a sub-G1 fraction. The apoptosis cascade involved downregulation of Bcl-2 protein; Bax expression was unchanged. Cell cycle progression was due to G0/G1 arrest, which involved increased expression of p53 and p21Cip1/Waf1, and decreased expression of cyclin D1. CDHA modulated cell cycle regulatory proteins and apoptosis-related proteins in a manner similar to that of parent DHA. In the athymic mouse system 1.0% dietary CDHA, but not 0.2%, significantly suppressed growth of KPL-1 tumor cells; CDHA tended to decrease regional lymph node metastasis in a dose dependent manner. Conclusion CDHA inhibited growth of KPL-1 human breast cancer cells in vitro more effectively than did DHA. The mechanisms of action involved modulation of apoptosis cascade and cell cycle progression. Dietary CDHA at 1.0% suppressed KPL-1 cell growth in the athymic mouse system.</p

    Valuation of rice postharvest losses in Sub-Saharan Africa and its mitigation strategies

    Data on rice harvest and postharvest loss in Sub-Sahara Africa (SSA) is scanty making it difficult for stakeholders to appreciate the loss and set priority areas for loss reduction along the value chain. To address this problem, a protocol was developed and validated for postharvest loss (PHL) quantification in SSA. Quantitative losses at each segment were determined by field measurements. Interactive effect of origin of rice (domestic versus imported) and type of processing (white versus parboiled milled) on rice price in 33 markets in Africa was used to estimate qualitative loss for both white and parboiled milled rice. Total PHL for rice in SSA in 2018 is estimated at about US$ 10.24 billion, representing 47.63% of the expected total production. The highest loss recorded was quantitative loss before and during harvesting, followed by qualitative loss along the entire value chain, quantitative loss during milling, parboiling, threshing in that order, with the lowest being quantitative loss during drying. Priority areas to be targeted for PHL reduction in SSA and some loss mitigation tools and technologies piloted or suitable for SSA are proposed

    Predicting Rice Yield Under Salinity Stress Using K/Na Ratio Variable in Plant Tissue

    Estimation of yield reduction in crop caused by the salinity stress is mostly based on variations of soil electrical conductivity and the severity of water stress. Crop response curves to salinity were developed without considering ion toxicity and nutritional imbalance in the plant. The objective of this study was to explore the possibility of using the ratio of the concentration of potassium by sodium in rice leaf (leaf-K/Na) to predict yield under the salinity stress. The rice (Oryza sativa L.) yield under fresh and saline condition and the leaf-K/Na related database was created. Data were collected from consecutive three seasons of a field experiment in the Africa Rice Center experimental farm in Senegal (16° 11ʹ N, 16° 15ʹW). We studied the relationship between the relative yield (Yr), a ratio of yield under the salinity stress to the potential yield and the leaf-K/Na (x). Furthermore, we did regression analyses and F-test to determine the best fitting function. Results indicate that the exponential function [i.e. Yr = 100 exp (-b x)] was the best fitting model with the lowest root mean square error (9.683) and the highest R2 value (0.90). Example applications on independent data from published papers showed relatively good predictions, suggesting that the model can be used to predict rice yield in saline soils

    Clinical and immunological evaluation of anti-apoptosis protein, survivin-derived peptide vaccine in phase I clinical study for patients with advanced or recurrent breast cancer

    <p>Abstract</p> <p>Background</p> <p>We previously reported that survivin-2B, a splicing variant of survivin, was expressed in various types of tumors and that survivin-2B peptide might serve as a potent immunogenic cancer vaccine. The objective of this study was to examine the toxicity of and to <b>c</b>linically and immunologically evaluate survivin-2B peptide in a phase I clinical study for patients with advanced or recurrent breast cancer.</p> <p>Methods</p> <p>We set up two protocols. In the first protocol, 10 patients were vaccinated with escalating doses (0.1–1.0 mg) of survivin-2B peptide alone 4 times every 2 weeks. In the second protocol, 4 patients were vaccinated with the peptide at a dose of 1.0 mg mixed with IFA 4 times every 2 weeks.</p> <p>Results</p> <p>In the first protocol, no adverse events were observed during or after vaccination. In the second protocol, two patients had induration at the injection site. One patient had general malaise (grade 1), and another had general malaise (grade 1) and fever (grade 1). Peptide vaccination was well tolerated in all patients. In the first protocol, tumor marker levels increased in 8 patients, slightly decreased in 1 patient and were within the normal range during this clinical trial in 1 patient. With regard to tumor size, two patients were considered to have stable disease (SD). Immunologically, in 3 of the 10 patients (30%), an increase of the peptide-specific CTL frequency was detected. In the second protocol, an increase of the peptide-specific CTL frequency was detected in all 4 patients (100%), although there were no significant beneficial clinical responses. ELISPOT assay showed peptide-specific IFN-γ responses in 2 patients in whom the peptide-specific CTL frequency in tetramer staining also was increased in both protocols.</p> <p>Conclusion</p> <p>This phase I clinical study revealed that survivin-2B peptide vaccination was well tolerated. The vaccination with survivin-2B peptide mixed with IFA increased the frequency of peptide-specific CTL more effectively than vaccination with the peptide alone, although neither vaccination could induce efficient clinical responses. Considering the above, the addition of another effectual adjuvant such as a cytokine, heat shock protein, etc. to the vaccination with survivin-2B peptide mixed with IFA might induce improved immunological and clinical responses.</p
