    Structural evaluation of candidate designs for the large space telescope primary mirror

    Structural performance analyses were conducted on two candidate designs (Itek and Perkin-Elmer designs) for the large space telescope three-meter mirror. The mirror designs and the finite-element models used in the analyses evaluation are described. The results of the structural analyses for several different types of loading are presented in tabular and graphic forms. Several additional analyses are also reported: the evaluation of a mirror design concept proposed by the Boeing Co., a study of the global effects of local cell plate deflections, and an investigation of the fracture mechanics problems likely to occur with Cervit and ULE. Flexibility matrices were obtained for the Itek and Perkin-Elmer mirrors to be used in active figure control studies. Summary, conclusions, and recommendations are included

    Red Sea Outflow Experiment (REDSOX) : DLD2 RAFOS float data report February 2001 - March 2003

    This is the final data report of all acoustically tracked second-generation Deep Lagrangian Drifter (DLD2) RAFOS float data collected by the Woods Hole Oceanographic Institution in 2001-2003 during the Red Sea Outflow Experiment (REDSOX). The float component of REDSOX was comprised of two deployments on the R/V Knorr and R/V Ewing: the first in February-March 2001, with 26 floats, and the second in August-September 2001, with 27 floats. The isobaric floats were ballasted for 650 decibars to target the intermediate-depth, high-salinity outflow waters from the Red Sea. The objectives of the Lagrangian float study were (1) to identify the spreading pathways of the equilibrated Red Sea outflow, and to quantify the velocities and eddy variability typical of this outflow and of the background oceanic environment in the Gulf of Aden, and (2) to identify and describe the mesoscale processes which contribute to the seaward transport of Red Sea Overflow Water properties through the Gulf of Aden and into the western Indian Ocean. In addition to floats activated and launched during the two cruises, four time-series sites were chosen for dual-release float moorings. The dual-release floats were released every two months between cruises and every two months after the second cruise, with the final release in March 2002. A pirate attack on the R/V Ewing forced some modification of the float deployment plan during the second cruise.Funding was provided by the National Science Foundation under Grant Number OCE-9818464

    Genome-wide sequence and functional analysis of early replicating DNA in normal human fibroblasts

    BACKGROUND: The replication of mammalian genomic DNA during the S phase is a highly coordinated process that occurs in a programmed manner. Recent studies have begun to elucidate the pattern of replication timing on a genomic scale. Using a combination of experimental and computational techniques, we identified a genome-wide set of the earliest replicating sequences. This was accomplished by first creating a cosmid library containing DNA enriched in sequences that replicate early in the S phase of normal human fibroblasts. Clone ends were then sequenced and aligned to the human genome. RESULTS: By clustering adjacent or overlapping early replicating clones, we identified 1759 "islands" averaging 100 kb in length, allowing us to perform the most detailed analysis to date of DNA characteristics and genes contained within early replicating DNA. Islands are enriched in open chromatin, transcription related elements, and Alu repetitive elements, with an underrepresentation of LINE elements. In addition, we found a paucity of LTR retroposons, DNA transposon sequences, and an enrichment in all classes of tandem repeats, except for dinucleotides. CONCLUSION: An analysis of genes associated with islands revealed that nearly half of all genes in the WNT family, and a number of genes in the base excision repair pathway, including four of ten DNA glycosylases, were associated with island sequences. Also, we found an overrepresentation of members of apoptosis-associated genes in very early replicating sequences from both fibroblast and lymphoblastoid cells. These data suggest that there is a temporal pattern of replication for some functionally related genes

    Wildlife populations in Southeast Asia are subject to increasing pressure from climate change, habitat loss and human disturbance. Cave-roosting bats are particularly vulnerable to all three factors. Because of the ecological services they provide, it is important to assess specific vulnerabilities to inform their conservation management. We evaluated the reproductive phenology and body condition of Chaerephon plicatus for 14 months in 2015–2016 and quantified guano harvesting at the largest colony in Cambodia in 2011–2016. As in Thailand and Myanmar, two annual breeding cycles were recorded, characterized as continuous bimodal polyoestry, with parturition primarily occurring in April and October. Significant declines occurred in body condition between the late wet season and the late dry season, suggesting that bats experience increasing energetic stress as the dry season progresses. Annual guano harvests increased over the study period but could not be used as a proxy for monitoring population size due to the loss of unknown amounts during the wet season and unquantified movements of bats between C. plicatus colonies in the region. We recommend studies to determine the scale and drivers of such movements and creation of sustainable guano harvesting and population monitoring initiatives to ensure the conservation of C. plicatus colonies in Cambodia

    Eddy Seeding in the Labrador Sea: A Submerged Autonomous Launch Platform Application

    Abstract Two submerged autonomous launch platforms (SALPs) were deployed at 500-m depth on a deep-water mooring in the northeastern Labrador Sea from 2007 to 2009 to automatically release profiling floats into passing warm-core anticyclonic Irminger Rings (IRs). The objective was to investigate the rings’ vertical structure and evolution as they drifted from their formation site near the western coast of Greenland to the area of deep convection in the south-central part of the basin. Mechanically and electronically, the SALP worked well: 10 out of 11 floats were successfully released from the mooring over 2 years. However, getting floats trapped in eddy cores using a preprogrammed release algorithm based on temperature and pressure (a proxy for current speed) measured by the SALPs met with limited success mainly because 1) the floats settled at a park pressure that was initially too deep, below the volume of water trapped in the eddy core; 2) the eddies translated past the mooring much more quickly than anticipated; and 3) there is a seasonal cycle in both background and eddy core temperature that was not known a priori and therefore not accounted for in the release algorithm. The other mooring instruments (100–3000 m) revealed that 12 anticyclones passed by the mooring in the 2-yr monitoring period. Using this independent information, the authors assessed and improved the release algorithm, still based on ocean conditions measured at one depth, and found that much better performance could have been achieved with an algorithm that allowed for faster eddy translation rates and the seasonal temperature cycle.</jats:p

    CLIMODE bobber data report : July 2005 - May 2009

    This report summarizes direct observations of Eighteen Degree Water (EDW) subduction and dispersal within the subtropical gyre of the North Atlantic Ocean. Forty acoustically-tracked bobbing, profiling floats (“bobbers”) were deployed to study the formation and dispersal of EDW in the western North Atlantic. The unique bobber dataset described herein provides insight into the evolution of EDW by means of direct, eddy-resolving measurement of EDW Lagrangian dispersal pathways and stratification. Bobbers are modified Autonomous Profiling Explorer (APEX) profiling floats which actively servo their buoyancy control mechanism to follow a particular isothermal surface. The CLIVAR Mode Water Dynamics Experiment (CLIMODE) bobbers tracked the 18.5°C temperature surface for 3 days, then bobbed quickly between the 17°C and 19°C isotherms. This cycle was repeated for one month, after which each bobber profiled to 1000 m before ascending to the surface to transmit data. The resulting dataset (37/40 tracked bobbers; more than half still profiling as of January 2010) yields well-resolved trajectories, unprecedented velocity statistics in the core of the subducting and spreading EDW, and detailed information about the Lagrangian evolution of EDW thickness and vertical structure.
