    Characterization and Improvement of the Image Quality of the Data Taken with the Infrared Camera (IRC) Mid-Infrared Channels onboard AKARI

    Mid-infrared images frequently suffer artifacts and extended point spread functions (PSFs). We investigate the characteristics of the artifacts and the PSFs in images obtained with the Infrared Camera (IRC) onboard AKARI at four mid-infrared bands of the S7 (7{\mu}m), S11 (11{\mu}m), L15 (15{\mu}m), and L24 (24 {\mu}m). Removal of the artifacts significantly improves the reliability of the ref- erence data for flat-fielding at the L15 and L24 bands. A set of models of the IRC PSFs is also constructed from on-orbit data. These PSFs have extended components that come from diffraction and scattering within the detector arrays. We estimate the aperture correction factors for point sources and the surface brightness correction factors for diffuse sources. We conclude that the surface brightness correction factors range from 0.95 to 0.8, taking account of the extended component of the PSFs. To correct for the extended PSF effects for the study of faint structures, we also develop an image reconstruction method, which consists of the deconvolution with the PSF and the convolution with an appropriate Gaussian. The appropriate removal of the artifacts, improved flat-fielding, and image reconstruction with the extended PSFs enable us to investigate de- tailed structures of extended sources in IRC mid-infrared images.Comment: 35 pages, 15 figures, accepted for publication in PAS

    本研究の目的は, 小中の発達段階に照らしながら他者を学習場面に効果的に設定し, 他者とのかかわり合いの中から学習者が自分の言葉を豊かにしていく学習活動・単元を開発するための基礎的な知見を得ることにある。他者の設定については, 大きく二つに分けて考えた。一つめは「教材としての多様な他者の言葉」であり, 二つめは「協同的な学び」である。また, 「発信型の授業構成」を設定することで教室内外の他者との出会いをつくり出し, 協同的な学びを発展させることができると考えた。検証の材料として用いるのは, 子どもの学習記録や作品, 授業記録などである。結果として, 多様な他者に出会わせることによって子どもたちはより多角的・客観的に言葉をとらえ, 興味を持って教材に向き合うことができた。また他者を設定することで, 子どもたちは言葉をとおしてかかわり合い, 自分の考えの広がりや深まりを確かめたりすることができるようになってきた。このように他者の存在を通して, 学習者はより豊かな言葉を得ることができる。今後は, 実生活においても他者とつながる言葉をつくり出す学びへと発展させていきたい

    Five-point Likert scaling on MRI predicts clinically significant prostate carcinoma

    Background: To clarify the relationship between the probability of prostate cancer scaled using a 5-point Likert system and the biological characteristics of corresponding tumor foci. Methods: The present study involved 44 patients undergoing 3.0-Tesla multiparametric MRI before laparoscopic radical prostatectomy. Tracing based on pathological and MRI findings was performed. The relationship between the probability of cancer scaled using the 5-point Likert system and the biological characteristics of corresponding tumor foci was evaluated. Results: A total of 102 tumor foci were identified histologically from the 44 specimens. Of the 102 tumors, 55 were assigned a score based on MRI findings (score 1: n = 3; score 2: n = 3; score 3: n = 16; score 4: n = 11 score 5: n = 22), while 47 were not pointed out on MRI. The tracing study revealed that the proportion of >0.5 cm3 tumors increased according to the upgrade of Likert scores (score 1 or 2: 33 %; score 3: 68.8 %; score 4 or 5: 90.9 %, χ2 test, p 7 also increased from scale 2 to scale 5 (scale 2: 0 %; scale 3: 56.3 %; scale 4: 72.7 %; 5: 90.9 %, χ2 test, p = 0.0001). On using score 3 or higher as the threshold of cancer detection on MRI, the detection rate markedly improved if the tumor volume exceeded 0.5 cm3 (<0.2 cm3: 10.3 %; 0.2-0.5 cm3: 25 %; 0.5-1.0 cm3: 66.7 %; 1.0 < cm3: 92.1 %). Conclusions: Each Likert scale favobably reflected the corresponding tumor’s volume and Gleason score. Our observations show that “score 3 or higher” could be a useful threshold to predict clinically significant carcinoma when considering treatment options

    FANTOM5 CAGE profiles of human and mouse samples

    In the FANTOM5 project, transcription initiation events across the human and mouse genomes were mapped at a single base-pair resolution and their frequencies were monitored by CAGE (Cap Analysis of Gene Expression) coupled with single-molecule sequencing. Approximately three thousands of samples, consisting of a variety of primary cells, tissues, cell lines, and time series samples during cell activation and development, were subjected to a uniform pipeline of CAGE data production. The analysis pipeline started by measuring RNA extracts to assess their quality, and continued to CAGE library production by using a robotic or a manual workflow, single molecule sequencing, and computational processing to generate frequencies of transcription initiation. Resulting data represents the consequence of transcriptional regulation in each analyzed state of mammalian cells. Non-overlapping peaks over the CAGE profiles, approximately 200,000 and 150,000 peaks for the human and mouse genomes, were identified and annotated to provide precise location of known promoters as well as novel ones, and to quantify their activities

    本研究は, 学習者が自分のまわりの他者と出会いかかわり合うことをとおして, 自分自身の言葉の世界を広げ, 自らの学びを探究していくことができるようにするための効果的な指導法を開発していくことを目的としている。そこで, 「さまざまな他者と出会い, 問いを立てる」「個の考えを深め吟味する」「他者とともによりよい方法で問題解決に取り組む」「学びの成果を適切に表現する」「学びを振り返り新たな学びにつなげる」という学習のプロセスに留意して単元構成を行った。2年生では, 自分の発想を言葉として表現したものを, 身近な他者に伝える経験を積ませる学習活動を構成することによって, 達成感を感じさせることができた。また, 子どもたちの意欲を喚起し学習に見通しをもたせることで, 発想豊かにお話作りに取り組ませることができた。3年生では, 説明文の内容を地域の行事と比べながら読ませたり, 今まで何気なく経験してきている行事の意味を改めて見つめ直させたりすることができた。また, 既習教材との比較によって, 筆者の意図や論理構成の工夫に気付かせることができた。今後も, 言葉の世界をひらき, 学びを自ら探求する学習のあり方を探っていきたい

    Involvement of β-defensin 130 (DEFB130) in the macrophage microbicidal mechanisms for killing Plasmodium falciparum

    金沢大学医薬保健研究域薬学系Understanding the molecular defense mechanism of macrophages and identifying their effector molecules against malarial parasites may provide important clues for the discovery of new therapies. To analyze the immunological responses of malarial parasite-induced macrophages, we used DNA microarray technology to examine the gene profile of differentiated macrophages phagocytizing Plasmodium falciparum-parasitized erythrocytes (iRBC). The transcriptional gene profile of macrophages in response to iRBCs represented 168 down-regulated genes, which were mainly involved in the cellular immune response, and 216 upregulated genes, which were involved in cellular proteolysis, growth, and adhesion. Importantly, the specific upregulation of β-defensin 130 (DEFB130) in these macrophages suggested a possible role for DEFB130 in malarial parasite elimination. Differentiated macrophages phagocytizing iRBCs exhibited an increase in intracellular DEFB130 levels and DEFB130 appeared to accumulate at the site of iRBC engulfment. Transfection of esiRNA-mediated knockdown of DEFB130 into macrophages resulted in a remarkable reduction in their antiplasmodial activity in vitro. Furthermore, DEFB130 synthetic peptide exhibited a modest toxic effect on P. falciparum in vitro and P. yoelii in vivo, unlike scrambled DEFB130 peptide, which showed no antiplasmodial activity. Together, these results suggest that DEFB130 might be one of the macrophage effector molecules for eliminating malarial parasites. Our data broaden our knowledge of the immunological response of macrophages to iRBCs and shed light on a new target for therapeutic intervention. © The Author(s) 2017

    本研究は, 学習者が自分のまわりの他者と出会いかかわり合うことをとおして, 自分自身の言葉の世界を広げ, 学びを探究していくことができるようにするための効果的な指導法を開発していくことを目的としている。他者とのかかわり合いをとおして, 自分の思いを表現し, 知識をもとにした的確な判断を行う学習を積み重ねることで, さまざまな場面で活躍していくための力を育み, 学びを実生活や社会の中に生かしていくことで他者とのよりよい関係を築いていくことにつながると考えた。そこで, 「言葉を学ぶ意欲の喚起」「多様な他者との出会い」「コミュニケーションを大事にした協同的な学び」「認識の広がりや深まりを実感させる場の設定」「表現を磨き合う場の設定」の5点に留意して単元構成を行った。その結果, 4年生では, 筆者の主張について, より深く考えさせ, 教材文の筆者の工夫を, 調べ学習の発表原稿やまとめ方の工夫に生かさせることができた。6年生では, 多くの他者の存在を意識して学習を進め, 本校の子どもたちに報道する事を意識し, 様々な視点からニュース番組を構成させることができた。今後も, 学習者を取り巻く環境を考慮し, 他者を意識した多様な実践を積み重ねていきたい

    CSC with and without steroids

    We investigated the rates of the use of steroids in Japanese central serous chorioretinopathy (CSC) cases and differences in the characteristics of CSC with and without steroids. A total of 538 eyes of 477 patients diagnosed with CSC, with 3 months or more of follow-up between April 2013 and June 2017 at 8 institutions. Patients with CSC with more than 3 months of follow-up were identified by OCT and fluorescein angiography at 8 institutions. Data collected included patient demographics, history of corticosteroid medication and smoking, spherical errors, findings of angiography, subfoveal choroidal thickness, and changes through the follow-up period. Differences in these findings were analyzed in cases with and without corticosteroid treatment. Among the 477 patients (344 men,133 women), 74 (15.5%) (39 men, 35 women) underwent current or prior steroid treatment. Cases with steroids were higher age (p = 0.0403) and showed no male prevalence, more bilateral involvement (p < 0.0001), and the affected eyes had multiple pigment epithelial detachment (p <0.0001), more fluorescein leakage sites (p < 0.0001), greater choroidal thickness (p = 0.0287) and a higher recurrence rate (p = 0.0412). Steroids can cause severer CSC through an effect on choroidal vessels and an impairment of retinal pigment epithelium
