Validation of the Japanese version of the CPAQ-8
This study aimed to examine the reliability and validity of the Japanese version of the Chronic Pain Acceptance Questionnaire (CPAQ-8J). A total of 108 outpatients with chronic pain completed the CPAQ-8 questionnaire, along with the Acceptance and Action Questionnaire-II, Hospital Anxiety and Depression Scale, Pain Disability Assessment Scale, Numerical Rating Scale, and EuroQol 5 dimensions 5-level. Confirmatory factor analyses examined the factor structure. Results indicated that the CPAQ-8J comprised a two-component factor structure. Correlations between the CPAQ-8J and each variable were as expected, except between the “pain willingness” subscale and other scales ; thus, the CPAQ-8J had a certain degree of convergent validity. Internal consistency and test-retest reliability suggest that the CPAQ-8J is reliable. The psychometric properties of the CPAQ-8J meet a certain standard ; meanwhile, some issues must be addressed for its practical application. Further research should consider the influence of cultural characteristics in practical application
Estimation of Completion of Dormancy in Grapevine Bud Based on Cumulative Temperature
This study was conducted to investigate the relationship between completion of dormancy of grapevine
bud and temperature. Canes of ‘Kyoho’ and ‘Pione’ grapevines (Vitis labrusca×V. vinifera) grown in 7 vineyards with different temperature conditions, in Nagano, Northern Okayama, Yamanashi, Okayama, Okayama University, Fukuoka and Miyazaki, were collected at three different chilling exposures,
December, January and February. These were then sent to Okayama University all at the same time. Cuttings with one bud were put into growth chambers kept at 25 or 30°C with 14 hours daylength, and budbreak in each cutting was surveyed at two day intervals for 60 days. Cumulative chilling hours (CCH) of exposure to below 7.2°C in each treatment time was largest in Nagano, followed in order by Northern Okayama, Yamanashi, Okayama, Okayama University, Fukuoka and Miyazaki. The CCH in Nagano was 2.5 to 4.8 times larger than in Miyazaki depending on the treatment time. The later the treatment time and the higher the temperature, the fewer were the number of days to first budbreak (NDFB) after treatment, irrespective of cultivar. A similar trend was observed in the number of days to 60% budbreak. In ‘Kyoho’ the NDFB was short in Nagano, Okayama University and Miyazaki, and longer in Okayama, Yamanashi and Fukuoka. In ‘Pione’ the NDFB was short in Fukuoka and Okayama University, and longer in Yamanashi and Okayama. The result was a weak negative correlation observed between CCH and NDFB in 4 of 7 vineyards. However, there was a strong positive correlation between NDFB and cumulative temperature (CT), a summation of temperature and hours of exposure to above 0°C from November 1 to treatment time and hours of exposure to 25 or 30°C from start of treatment to budbreak in each plot, in 6 vineyards excluding Miyazaki. The importance of estimating the completion of dormancy in grapevine bud based on CT is discussed.ブドウの芽の休眠完了と温度との関係を調査するため,温度条件の異なる7園(中信農試,山梨果試,岡山農試,岡山農試北部支場,岡山大学,福岡農試および宮崎)で栽培されている‘巨峰’と‘ピオーネ’から低温遭遇量の異なる3時期(12月,1月,2月)に結果母枝を採取した.直ちに岡山大学に送り,1芽を持つ挿し穂に調整した後,25または30℃のインキュベーター(14時間日長)に入れ,2日間隔で60日間発芽を調査した.処理開始時の7.2℃以下の遭遇量は中信農試で最も多く,次いで岡山農試北部支場,山梨果試,岡山農試,岡山大学,福岡農試,宮崎の順で,中信農試と宮崎では処理時期により2.5~4.8倍の差があった.発芽所要日数は,両品種とも処理時期が遅いほど,また温度が高いほど少なく,60%発芽所要日数もほぼ同様の傾向であった.‘巨峰’の発芽所用日数は中信農試,岡山大学および宮崎で少ない一方,岡山農試,山梨果試および福岡農試で多く,また‘ピオーネ’では福岡農試と岡山大学で少なく,山梨果試と岡山農試で多かった.7園のうち4園で低温遭遇量と発芽所要日数との間に負の相関がみられたが,相関係数は低かった.一方,11月以降処理開始までの0℃以上の温度の積算値と処理開始から発芽までの25または30℃での積算値を合計した積算温度と発芽所要日数との間には1園を除き極めて高い正の相関が認められた.これらの結果を基に,積算温度によるブドウの休眠完了予測の可能性を考察した
Classic Chromophobe Renal Cell Carcinoma Incur a Larger Number of Chromosomal Losses Than Seen in the Eosinophilic Subtype
Chromophobe renal cell carcinoma (chRCC) is a renal tumor subtype with a good prognosis, characterized by multiple chromosomal copy number variations (CNV). The World Health Organization (WHO) chRCC classification guidelines define a classic and an eosinophilic variant. Large cells with reticular cytoplasm and prominent cell membranes (pale cells) are characteristic for classic chRCC. Classic and eosinophilic variants were defined in 42 Swiss chRCCs, 119 Japanese chRCCs and in whole-slide digital images of 66 chRCCs from the Cancer Genome Atlas (TCGA) kidney chromophobe (KICH) dataset. 32 of 42 (76.2%) Swiss chRCCs, 90 of 119 (75.6%) Japanese chRCCs and 53 of 66 (80.3%) TCGA-KICH were classic chRCCs. There was no survival difference between eosinophilic and classic chRCC in all three cohorts. To identify a genotype/phenotype correlation, we performed a genome-wide CNV analysis using Affymetrix OncoScan CNV Assay (Affymetrix/Thermo Fisher Scientific, Waltham, MA, USA) in 33 Swiss chRCCs. TCGA-KICH subtypes were compared with TCGA CNV data. In the combined Swiss and TCGA-KICH cohorts, losses of chromosome 1, 2, 6, 10, 13, and 17 were significantly more frequent in classic chRCC (p < 0.05, each), suggesting that classic chRCC are characterized by higher chromosomal instability. This molecular difference justifies the definition of two chRCC variants. Absence of pale cells could be used as main histological criterion to define the eosinophilic variant of chRCC
The Effect of Long-Term Use of an Eyewash Solution on the Ocular Surface Mucin Layer
The use of eyewash solutions in Japan, especially in patients with allergic conjunctivitis and contact lens wearers, has been increasing. Our aim was to investigate how the use of preservative-free eyewash solution in healthy eyes for one month affects corneal safety and ocular surface mucin. We analyzed 42 eyes of 21 individuals (17 males, four females; mean age: 36.1 ± 7.4 years) without ocular allergies, dry eyes, or other ocular diseases through a prospective study. Eyes were randomized to a wash group (group one) and a nonwash follow up group (group two). We evaluated the dry eye-related quality-of-life score (DEQS), tear film breakup time (TBUT), fluorescein staining score, mRNA expression of MUC5AC and MUC16, MUC16 immunohistochemistry, and MUC5AC periodic acid Schiff (PAS) staining. There was a significant decrease in DEQS scores after one month of eyewash use (p < 0.05). There were no significant differences in other evaluation items that were analyzed (all p > 0.05). Furthermore, no significant differences were observed between group one and group two in all endpoints (all p > 0.05). The results suggest that one month use of a nonpreserved eyewash solution has no detrimental effects on the tear film and the ocular surface mucins
Improvement of insulation performance of solid/gas composite insulation with embedded electrode
Solid insulated switchgear (SIS) has been developed as a substitution for medium voltage sulfur hexafluoride (SF6) gas insulated switchgears. Its main circuit including a vacuum interrupter is coated with epoxy resin. Therefore, a solid/gas insulation system is composed. For further miniaturization and higher stress design of SIS, improvement of the insulation performance is required. In the solid/gas insulation system, the breakdown strength along the surface is lower than that of solid. Then, the authors investigated the surface insulation performance of the solid/gas insulation system containing an embedded electrode. In the present study, the sizes of the embedded electrode and the solid insulation material were varied and the partial discharge inception voltage and the surface breakdown voltage were measured. As a result, it was found that the surface breakdown voltage varied considerably with the radius of the embedded electrode. From the result, it was deduced that the surface insulation performance could be improved by choosing the size of the embedded electrode appropriately. Moreover, the effects of the thickness of the insulation material and of the surface distance on the surface insulation performance were clarified
Estimation of surface breakdown voltage of solid/gas composite insulation with embedded electrode

For further miniaturization and higher stress design of SIS, the authors investigated on the surface insulation of the solid/gas insulation system containing an embedded electrode. In the present study, the surface breakdown voltage was estimated based on the surface discharge propagation characteristics. From the measurement of the surface flashover voltage on a dielectric against the surface distance, the relation between the surface flashover voltage and the surface distance was found to be expressed with an equation derived from the empirical equation presented by Toepler. Then, the surface breakdown voltage was estimated based on the obtained equation. The estimated values were compared with the experimental data obtained varying a parameter of the radius of the embedded electrode, thickness of the epoxy resin plate or the radius of the plate. As a result, a good correlation was obtained. Therefore, the estimation method of the surface breakdown voltage on the solid/gas insulation with embedded electrode was established
X-ray structure of the fourth type of archaeal tRNA splicing endonuclease: insights into the evolution of a novel three-unit composition and a unique loop involved in broad substrate specificity.
Cleavage of introns from precursor transfer RNAs (tRNAs) by tRNA splicing endonuclease (EndA) is essential for tRNA maturation in Archaea and Eukarya. In the past, archaeal EndAs were classified into three types (α'2, α4 and α2β2) according to subunit composition. Recently, we have identified a fourth type of archaeal EndA from an uncultivated archaeon Candidatus Micrarchaeum acidiphilum, referred to as ARMAN-2, which is deeply branched within Euryarchaea. The ARMAN-2 EndA forms an ε2 homodimer and has broad substrate specificity like the α2β2 type EndAs found in Crenarchaea and Nanoarchaea. However, the precise architecture of ARMAN-2 EndA was unknown. Here, we report the crystal structure of the ε2 homodimer of ARMAN-2 EndA. The structure reveals that the ε protomer is separated into three novel units (αN, α and βC) fused by two distinct linkers, although the overall structure of ARMAN-2 EndA is similar to those of the other three types of archaeal EndAs. Structural comparison and mutational analyses reveal that an ARMAN-2 type-specific loop (ASL) is involved in the broad substrate specificity and that K161 in the ASL functions as the RNA recognition site. These findings suggest that the broad substrate specificities of ε2 and α2β2 EndAs were separately acquired through different evolutionary processes
