    The importance of agriculture in the development of Romanian economy

    Agriculture represent one of the most important branches of the Romanian economy. Within performing the suggested analysis we started from the premise that the economy of each country, regardless of developing degree, agriculture, by natural and human resources available, by contributing to the creation of gross domestic product, of gross value added, but also by participating at internal and external trade, obviously holds an important position. Romanian rural economy is currently dominated in a large part by agriculture. Favorable geographical conditions, topography, climate, fertile soils, in addition with workforce, a suitable administrative system, but the connection of rural population to land and animals can make the Romanian agriculture as production branch attractive and profitable to determine economic growth at national level. Thus, we considered necessary to perform an analysis concerning the importance of Romanian agriculture, analysis that represents the basis for finding the most viable solutions to determine this particularly important branch - to relaunch in the development of Romanian rural economy

    CYP450 2D6 Genotype and Flecainide Efficacy in the Treatment of Patients with Lone Atrial Fibrillation

    Background: CYP2D6 has been linked to one of four phenotypes: a) ultra-rapid metabolizers (UM), with multiple gene copies; b) extensive metabolizers (EM), with a single wild type gene copy, considered normal; c) intermediate (IM) metabolizer, with decreased enzymatic activity; and d) poor metabolizers (PM) with no detectable enzymatic activity.  By altering the drug dose-plasma concentration relationship, these differences may lead to severe toxicity and or therapeutic failure.Objectives: The aim of this study was to determine the correlation between CYP2D6 polymorphisms and both efficacy and magnitude of adverse reactions of flecainide, a class IC antiarrhythmic agent.Methods: Patients with Lone Atrial fibrillation (AF) were enrolled in a 2-arm prospective study: patients started on flecainide at the initial visit, then were followed up at 3 and 6 months intervals (arm 1) or exhibited AF recurrences on flecainide, defined as treatment failure (arm 2).  Data about recurrence of AF, side effects, and demographics were collected. Genotyping was performed using AmpliChipTM CYP450.Results: A total of 26 lone AF patients were enrolled (12 in arm 1, and 14 in arm 2).  The mean age was 47± 10.8 years and 56.2 ± 10.8 years respectively.  Among the analyzed phenotypes the following distribution was found: 1/26 (3.8%) UM, 19/26 (73%) EM, 5/26 (19%) IM, 1/26 (3.8%) PM.Conclusions: In this small series of patients with lone atrial fibrillation, most patients were found to be extensive metabolizers of flecainide.  There was no statistically significant correlation between the patients' genotype, and flecainide efficacy / side effects

    Does body mass index impact assisted reproductive technology treatment outcomes in gestational carriers

    Abstract Background The purpose of this study was to assess whether increased body mass index (BMI) negatively affects assisted reproductive technology (ART) outcomes among gestational carriers. Methods A retrospective matched case-control cohort, including all gestational carrier (GC) cycles performed at CReATe Fertility Centre (Toronto, ON, Canada) between 2003 and 2016. Setting A Canadian fertility clinic, with a large surrogacy program. Patients All gestational carriers that had undergone a cycle completed to a transfer at our clinic, and had BMI and outcome data available, were matched by BMI to infertile patients treated at our clinic during the same years provided they had undergone a cycle completed to a transfer, and had outcomes data available. Interventions None. Main outcome measures Clinical pregnancies rates, miscarriage rates and live birth rates. Results BMI was not a reliable prediction factor of any of the measured outcomes. Importantly, the gestational carrier population had better outcomes and a significantly lower overall incidence of maternal, fetal and neonatal complications when compared with infertile patients, treated at our clinic during the same years. Conclusion BMI is not a reliable predictor of outcomes among gestational carriers

    The ATR inhibitor ceralasertib potentiates cancer checkpoint immunotherapy by regulating the tumor microenvironment

    Abstract The Ataxia telangiectasia and Rad3-related (ATR) inhibitor ceralasertib in combination with the PD-L1 antibody durvalumab demonstrated encouraging clinical benefit in melanoma and lung cancer patients who progressed on immunotherapy. Here we show that modelling of intermittent ceralasertib treatment in mouse tumor models reveals CD8+ T-cell dependent antitumor activity, which is separate from the effects on tumor cells. Ceralasertib suppresses proliferating CD8+ T-cells on treatment which is rapidly reversed off-treatment. Ceralasertib causes up-regulation of type I interferon (IFNI) pathway in cancer patients and in tumor-bearing mice. IFNI is experimentally found to be a major mediator of antitumor activity of ceralasertib in combination with PD-L1 antibody. Improvement of T-cell function after ceralasertib treatment is linked to changes in myeloid cells in the tumor microenvironment. IFNI also promotes anti-proliferative effects of ceralasertib on tumor cells. Here, we report that broad immunomodulatory changes following intermittent ATR inhibition underpins the clinical therapeutic benefit and indicates its wider impact on antitumor immunity
