Fatores que influenciam a qualidade de vida em adultos com doenças inflamatórias intestinais : uma revisão integrativa
As doenças inflamatĂłrias intestinais constituem a doença de Crohn e a colite ulcerativa. SĂŁo doenças que tĂȘm incidĂȘncia em todo o mundo e sĂŁo caraterizadas por perĂodos de crises e remissĂŁo. Os principais sintomas sĂŁo dor abdominal, diarreia, fadiga. Este estudo teve como objetivo descrever os fatores clĂnicos e psicossociais que influenciam a qualidade de vida de adultos com doença inflamatĂłria intestinal. Trata-se de uma revisĂŁo integrativa segundo Cooper, cujas bases de dados utilizadas foram Literatura Latino-Americana e do Caribe em CiĂȘncia e SaĂșde (LILACS), Scientific Electronic Library Online (SCIELO), US National liberty of Medicine (PUBMED). Utilizaram-se os descritores quality of life; Crohn disease e Colitis, ulcerative e selecionaram-se 25 artigos publicados no perĂodo de 2013 a 2017. Identificou-se que fatores como depressĂŁo, ansiedade, atividade da doença, distĂșrbios gastrointestinais funcionais, baixos Ăndices de vitamina D e fadiga diminuem os Ăndices de qualidade de vida em pacientes com doença inflamatĂłria intestinal. Os pacientes possuem muitos medos e receios por causa da natureza dos sintomas da doença. Devido a necessidade da urgĂȘncia para evacuar ou pela incontinĂȘncia fecal, necessitam ter banheiros prĂłximos. TĂȘm dificuldades para fazer amigos e de ter um relacionamento Ăntimo. Pacientes tambĂ©m apresentam dificuldades no trabalho, tĂȘm seu desempenho diminuĂdo e suas relaçÔes interpessoais afetadas por causa da doença. Foi percebido a partir desse estudo que Ă© essencial que haja mais informaçÔes sobre as doenças inflamatĂłrias intestinais e de seus sintomas para a população em geral e profissionais da saĂșde
CSN5 binds to misfolded CFTR and promotes its degradation
AbstractCystic fibrosis is mainly caused by mutations that interfere with the biosynthetic folding of the cystic fibrosis transmembrane conductance regulator (CFTR) protein. The aim of this study was to find cellular proteins interacting with CFTR and regulating its processing. We have used a genetic screen in yeast to identify such proteins and identified CSN5 that interacted with the third cytoplasmic loop of CFTR. CSN5 is the 5th component of the COP9 signalosome, a complex of eight subunits that shares significant homologies to the lid subcomplex of the 26S proteasome and controls the stability of many proteins. The present study shows that CSN5 associates with the core-glycosylated form of CFTR and suggests that this association targets misfolded CFTR to the degradative pathway. Identifying CSN5 as a new component of the degradative pathway is an important step towards the goal of unraveling the sorting between misfolded and correctly folded CFTR proteins
Bone ultrastructure and x-ray microanalysis of aluminum-intoxicated hemodialyzed patients
Bone ultrastructure and x-ray microanalysis of aluminum-intoxicated hemodialyzed patients. In hemodialyzed patients aluminum (Al) intoxication may induce osteomalacic lesions which are mainly observed when plasma immunoreactive parathyroid hormone (iPTH) concentrations are low, and osteitis fibrosa absent. In this study, the bone tissue of eight hemodialyzed patients with elevated plasma and bone Al concentrations was examined by histomorphometry, electron microscopy, and x-ray microanalysis. Five patients (group 1) had osteomalacia and minimal osteitis fibrosa, three patients (group 2) had severe osteitis fibrosa. In group 1, Al was concentrated at the mineralizing front, in hexagonal structures measuring 200 to 1,000 Ă
which also contained phosphorus, but not calcium. Hydroxyapatite needles had a normal aspect. Osteoblasts appeared inactive. In group 2, Al was also present at the mineralizing layer of osteoid, but, in these cases, in small clusters next to abnormal calcium deposits. Osteoblasts appeared very active. Their mitochondria contained calcium and phosphorus granules, or amorphous material, measuring 1,500 to 2,000 Ă
, emitting x-rays characteristic for Al and phosphorus. These results suggest that secondary hyperparathyroidism, by stimulating the cellular activity, may increase the uptake and release of Al by the osteoblasts. The presence of Al within the mitochondria of these cells may be one of the factors inducing the mineralization defect.Ultrastructure et microanalyse x du tissu osseux de malades hĂ©modialysĂ©s intoxiquĂ©s par l'aluminium. Oxidative stress plays a prominent role in the pathophysiology of cystic fibrosis (CF). Eicosanoid Release Is Increased by Membrane Destabilization and CFTR Inhibition in Calu-3 Cells
The antiinflammatory protein annexin-1 (ANXA1) and the adaptor S100A10 (p11), inhibit cytosolic phospholipase A2 (cPLA2α) by direct interaction. Since the latter is responsible for the cleavage of arachidonic acid at membrane phospholipids, all three proteins modulate eicosanoid production. We have previously shown the association of ANXA1 expression with that of CFTR, the multifactorial protein mutated in cystic fibrosis. This could in part account for the abnormal inflammatory status characteristic of this disease. We postulated that CFTR participates in the regulation of eicosanoid release by direct interaction with a complex containing ANXA1, p11 and cPLA2α. We first analyzed by plasmon surface resonance the in vitro binding of CFTR to the three proteins. A significant interaction between p11 and the NBD1 domain of CFTR was found. We observed in Calu-3 cells a rapid and partial redistribution of all four proteins in detergent resistant membranes (DRM) induced by TNF-α. This was concomitant with increased IL-8 synthesis and cPLA2α activation, ultimately resulting in eicosanoid (PGE2 and LTB4) overproduction. DRM destabilizing agent methyl-ÎČ-cyclodextrin induced further cPLA2α activation and eicosanoid release, but inhibited IL-8 synthesis. We tested in parallel the effect of short exposure of cells to CFTR inhibitors Inh172 and Gly-101. Both inhibitors induced a rapid increase in eicosanoid production. Longer exposure to Inh172 did not increase further eicosanoid release, but inhibited TNF-α-induced relocalization to DRM. These results show that (i) CFTR may form a complex with cPLA2α and ANXA1 via interaction with p11, (ii) CFTR inhibition and DRM disruption induce eicosanoid synthesis, and (iii) suggest that the putative cPLA2/ANXA1/p11/CFTR complex may participate in the modulation of the TNF-α-induced production of eicosanoids, pointing to the importance of membrane composition and CFTR function in the regulation of inflammation mediator synthesis

Glia: A Neglected Player in Noninvasive Direct Current Brain Stimulation
Noninvasive electrical brain stimulation by application of direct current (DCS) promotes plasticity in neuronal networks in vitro and in in vivo. This effect has been mainly attributed to the direct modulation of neurons. Glia represents approximately 50% of cells in the brain. Glial cells are electrically active and participate in synaptic plasticity. Despite of that, effects of DCS on glial structures and on interaction with neurons are only sparsely investigated. In this perspectives article we review the current literature, present own dose response data and provide a framework for future research from two points of view: first, the direct effects of DCS on glia and second, the contribution of glia to DCS related neuronal plasticity

Safety of ipsilesional anodal transcranial direct current stimulation in acute photothrombotic stroke: implications for early neurorehabilitation
Abstract Early rehabilitation in the acute phase of stroke, that bears unique neuroplastic properties, is the current standard to reduce disability. Anodal transcranial direct current stimulation can augment neurorehabilitation in chronic stroke. Studies in the acute phase are sparse and held back by inconclusive preclinical data pointing towards potential negative interaction of the excitability increasing tDCS modality with stroke-induced glutamate toxicity. In this present study, we aimed to evaluate structural and behavioral safety of anodal tDCS applied in the acute phase of stroke. Photothrombotic stroke including the right primary motor cortex was induced in rats. 24Â h after stroke anodal tDCS was applied for 20Â min ipsilesionally at one of four different current densities in freely moving animals. Effects on the infarct volume and on stroke induced neuroinflammation were assessed. Behavioral consequences were monitored. Infarct volume and the modified Neurological Severity Score were not affected by anodal tDCS. Pasta handling, a more sensitive task for sensorimotor deficits, and microglia reactivity indicated potentially harmful effects at the highest tDCS current density tested (47.8 A/m2), which is more than 60 times higher than intensities commonly used in humans. Compared to published safety limits of anodal tDCS in healthy rats, recent stroke does not increase the sensitivity of the brain to anodal tDCS, as assessed by lesion size and neuroinflammatory response. Behavioral deficits only occurred at the highest intensity, which was associated with increased neuroinflammation. When safety limits of commonly used clinical tDCS are met, augmentation of early neurorehabilitation after stroke by anodal tDCS appears to be feasible
