    Employability Blog Series: The Big College Challenge – A Case Study Presenting Interdisciplinary Development of Employability Skills

    This forms part of a regular series of Employability Blogs for the Higher Education Policy Institute. During January 2020, students of the College of Life and Natural Sciences (CLANS) at the University of Derby took part in an interdisciplinary challenge, termed “The Big Challenge”, alongside academics in their subject specialities and several leading industry professionals. This challenge aimed to support the development of key employability skills and was staged as part of a knowledge exchange activity to encourage mutual learning for both the academics and the employers involved (Reed, 2018). Since staging the Challenge, a research project has been in progress assessing the impact and the value of the Challenge on students, employers and the academics and professional services staff involved in delivering it.     Role of large-conductance Ca2+-activated K+ channels in adenosine A1 receptor-mediated pharmacological postconditioning in H9c2 cells

    Ischaemic postconditioning is a phenomenon whereby short periods of ischaemia applied during the start of reperfusion protect the myocardium from the damaging consequences of reperfusion. As such, pharmacological-induced postconditioning represents an attractive therapeutic strategy for reducing reperfusion injury during cardiac surgery and following myocardial infarction. The primary aim of this study was to determine the role of large-conductance Ca2+-activated potassium channels (BKCa channels) in adenosine A1 receptor-induced pharmacological postconditioning in the rat embryonic cardiomyoblast-derived cell line H9c2. H9c2 cells were exposed to 6 h hypoxia (0.5% O2) followed by 18 h reoxygenation (H/R) after which cell viability was assessed by monitoring lactate dehydrogenase (LDH) release and caspase-3 activation. The adenosine A1 receptor agonist N6-cyclopentyladenosine (CPA; 100 nmol/L) or the BKCa channel opener NS1619 (10 µmol/L) were added for 30 min at the start of reoxygenation following 6 h hypoxic exposure. Where appropriate, cells were treated (15 min) before pharmacological postconditioning with the BKCa channel blockers paxilline (1 µmol/L) or iberiotoxin (100 nmol/L). Pharmacological postconditioning with CPA or NS1619 significantly reduced H/R-induced LDH release. Treatment with paxilline or iberiotoxin attenuated adenosine A1 receptor and NS1619-induced pharmacological postconditioning. These results have shown for the first time that BKCa channels are involved in adenosine A1 receptor-induced pharmacological postconditioning in a cell model system

    Emperor penguins breeding on iceshelves

    We describe a new breeding behaviour discovered in emperor penguins; utilizing satellite and aerial-survey observations four emperor penguin breeding colonies have been recorded as existing on ice-shelves. Emperors have previously been considered as a sea-ice obligate species, with 44 of the 46 colonies located on sea-ice (the other two small colonies are on land). Of the colonies found on ice-shelves, two are newly discovered, and these have been recorded on shelves every season that they have been observed, the other two have been recorded both on ice-shelves and sea-ice in different breeding seasons. We conduct two analyses; the first using synthetic aperture radar data to assess why the largest of the four colonies, for which we have most data, locates sometimes on the shelf and sometimes on the sea-ice, and find that in years where the sea-ice forms late, the colony relocates onto the ice-shelf. The second analysis uses a number of environmental variables to test the habitat marginality of all emperor penguin breeding sites. We find that three of the four colonies reported in this study are in the most northerly, warmest conditions where sea-ice is often sub-optimal. The emperor penguin’s reliance on sea-ice as a breeding platform coupled with recent concerns over changed sea-ice patterns consequent on regional warming, has led to their designation as “near threatened” in the IUCN red list. Current climate models predict that future loss of sea-ice around the Antarctic coastline will negatively impact emperor numbers; recent estimates suggest a halving of the population by 2052. The discovery of this new breeding behaviour at marginal sites could mitigate some of the consequences of sea-ice loss; potential benefits and whether these are permanent or temporary need to be considered and understood before further attempts are made to predict the population trajectory of this iconic species

    Using the unique spectral signature of guano to identify unknown seabird colonies

    Despite the threats faced by seabirds in both terrestrial and marine habitats, even basic knowledge of the locations of colonies, population sizes and trends is lacking for many remote areas of the world. Recent studies have shown that the guano of Adélie penguins can be identified from Landsat Enhanced Thematic Mapper (ETM) imagery and used to map colonies on coasts around continental Antarctica. Our study highlights a new technique based on the unique spectral signature of guano that can be used to discriminate seabird colonies from background geology and vegetation in a wider range of natural environments, including the vegetated and zoologically-diverse region of the Antarctic Peninsula; moreover, the method was effective for all densely colonial, surface-nesting seabirds. Using Landsat ETM imagery, we correctly identified all known seabird colonies of over 50 pairs in the area of Marguerite Bay. Almost all other areas with a similar spectral signature that were outside known breeding areas were single pixels that were readily distinguishable from genuine colonies. If these were excluded, only 4.1% of pixels appeared to represent unknown breeding or roosting sites, and warrant further investigation. The spatial extent of the guano provided a general guide to the number of individuals present, but further work would be required to determine the accuracy of this method for estimating population size. Spectral profiles of guano collected by satellite and hand-held spectrometers were compared with available data in spectral libraries and did not match with any known geological profile. There may also be potential for discriminating colonies of different species that differ in phenology and show seasonal changes in diet by the carefully-timed acquisition of suitable satellite imagery. We conclude that the remotely-sensed guano signature is a good indicator of the location of seabird breeding or roosting sites, with potentially wide application to other areas of the world

    Effects of Novel Chalcone Derivatives upon H9c2 and MDCK Cell Viability

    Many compounds with potential anti-cancer activity fail to reach the latter stages of clinical trials due to adverse effects, often causing cardiac and renal toxicity. Here, we synthesised a group of novel chalcone compounds, thought to have potential anti-cancer activity4 and assessed their effects upon cardiac and renal cell viability. Data revealed that all compounds produced minimal short term toxicity. Further work will be performed to assess the long term effects of these compounds on cell viability, leading to mechanistic studies and structure activity relationship analyses

    Improving rail wear and RCF performance using laser cladding

    Laser cladding has been considered as a method for improving the wear and RCF performance of standard grade rail. This paper presents results of small scale tests carried out to assess the wear and RCF performance of rail which had been laser clad. Using the laser cladding process premium metals can be deposited on to the working surface of standard rail with the aim of enhancing the wear and RCF life of the rail. Various laser clad samples were tested using a twin-disc method. The candidate metals were clad on to standard R260 grade rail discs and were tested against a disc of standard wheel material. During the tests, wear rates and RCF initiation were monitored and compared to those of a standard rail disc. Six candidate cladding materials were chosen for this test: A multi-phase Manganese Steel Variant (MMV), Martensitic Stainless Steel (MSS), TWIP Steel, NiCrBSi, Stellite 12 and Stellite 6. The MSS, Stellite 6, and Stellite 12 samples showed reduced wear rates relative to the standard R260 Grade rail discs, and also produced a reduction in wheel steel wear. The RCF initiation resistance of all of the candidate materials was superior compared to the R260 Grade material

    Krill faecal pellets drive hidden pulses of particulate organic carbon in the marginal ice zone

    The biological carbon pump drives a flux of particulate organic carbon (POC) through the ocean and affects atmospheric levels of carbon dioxide. Short term, episodic flux events are hard to capture with current observational techniques and may thus be underrepresented in POC flux estimates. We model the potential hidden flux of POC originating from Antarctic krill, whose swarming behaviour could result in a major conduit of carbon to depth through their rapid exploitation of phytoplankton blooms and bulk egestion of rapidly sinking faecal pellets (FPs). Our model results suggest a seasonal krill FP export flux of 0.039 GT C across the Southern Ocean marginal ice zone, corresponding to 17–61% (mean 35%) of current satellite-derived export estimates for this zone. The magnitude of our conservatively estimated flux highlights the important role of large, swarming macrozooplankton in POC export and, the need to incorporate such processes more mechanistically to improve model projections

    Momentum anisotropy of the scattering rate in cuprate superconductors

    We examine the momentum and energy dependence of the scattering rate of the high temperature cuprate superconductors using angle resolved photoemission spectroscopy. The scattering rate is of the form a + b*w. The inelastic coefficient b is found to be isotropic. The elastic term, a, however, is found to be highly anisotropic in the pseudogap phase of optimal doped samples, with an anisotropy which correlates with that of the pseudogap. This can be contrasted with heavily overdoped samples, which show an isotropic scattering rate in the normal state

    A comparison of baleen whale density estimates derived from overlapping satellite imagery and a shipborne survey

    As whales recover from commercial exploitation, they are increasing in abundance in habitats that they have been absent from for decades. However, studying the recovery and habitat use patterns of whales, particularly in remote and inaccessible regions, frequently poses logistical and economic challenges. Here we trial a new approach for measuring whale density in a remote area, using Very-High-Resolution WorldView-3 satellite imagery. This approach has capacity to provide sightings data to complement and assist traditional sightings surveys. We compare at-sea whale density estimates to estimates derived from satellite imagery collected at a similar time, and use suction-cup archival logger data to make an adjustment for surface availability. We demonstrate that satellite imagery can provide useful data on whale occurrence and density. Densities, when unadjusted for surface availability are shown to be considerably lower than those estimated by the ship survey. However, adjusted for surface availability and weather conditions (0.13 whales per km2, CV = 0.38), they fall within an order of magnitude of those derived by traditional line-transect estimates (0.33 whales per km2, CV = 0.09). Satellite surveys represent an exciting development for high-resolution image-based cetacean observation at sea, particularly in inaccessible regions, presenting opportunities for ongoing and future research
