Nellore bull selection alternatives using multiple traits of economic importance
Os dados são provenientes de 234 touros da raça Nelore participantes de um teste de progênie, no período de 1996 a 2003. A diferença esperada na progênie (DEP) de sete características: peso aos 120 e 210 dias, efeito materno (DMPP120 e DMPP210), peso e perímetro escrotal aos 365 e 450 dias, efeito direto (DDP365, DDP450, DDPE365 e DDPE450) e idade ao primeiro parto (DDIPP) foi utilizada para classificar os animais em três grupos, assim como identificar quais as características possuíram maior poder discriminatório na formação de cada grupo. Para tanto, foram utilizados procedimentos estatísticos multivariados de análise de agrupamentos k-médias e componentes principais. Os resultados evidenciaram que, dos três grupos formados, dois se destacaram quanto aos valores médios das DEPs. A importância desses dois grupos de touros foi confirmada pela análise de componentes principais, que associou a eles valores superiores de DEPs diretas de peso e perímetro escrotal. A quantidade da variabilidade original retida pelos dois primeiros componentes principais foi de 70,22%. Estes procedimentos mostraram-se eficientes e constituíram importantes ferramentas para classificar touros, discriminar variáveis, bem como resumir informações multivariadas, podendo ser usados como auxílio valioso na seleção de reprodutores para uso nos programas de melhoramento genético.The data set is from 234 Nelore bulls which participate of a progeny test in the period from 1996 to 2003. The expected progeny diference (EPDs) of seven economic traits, weights at 120 e 210 days of age, maternal effect (DMPP120 and DMPP210), weights and scrotal circumferences at 365 e 450 days of age, direct effects (DDP365, DDP450, DDPE365 and DDPE450), and age at first calving (DDIPP), were used in order to classify these animals in three groups and verify which EPDs showed greatest discriminating power in forming the groups. The statistical applied techniques were: k-means clusters analysis and principal components analysis. From the three groups formed, two of them stood out in relation to values of the EPDs means. Evidence of the importance of these two groups was observed in the principal component analysis that associate to them higher values of direct EPDs of weight and scrotal circumference. The two first principal components accounted for 70.22% of the total original variability. Carbon antisite clusters in SiC: a possible pathway to the D_{II} center
The photoluminescence center D_{II} is a persistent intrinsic defect which is
common in all SiC polytypes. Its fingerprints are the characteristic phonon
replicas in luminescence spectra. We perform ab-initio calculations of
vibrational spectra for various defect complexes and find that carbon antisite
clusters exhibit vibrational modes in the frequency range of the D_{II}
spectrum. The clusters possess very high binding energies which guarantee their
thermal stability--a known feature of the D_{II} center. The di-carbon antisite
(C_{2})_{Si} (two carbon atoms sharing a silicon site) is an important building

Retinoic acid and cAMP inhibit rat hepatocellular carcinoma cell proliferation and enhance cell differentiation
Hepatocellular carcinoma (HCC) is the third highest cause of cancer death worldwide. In general, the disease is diagnosed at an advanced stage when potentially curative therapies are no longer feasible. For this reason, it is very important to develop new therapeutic approaches. Retinoic acid (RA) is a natural derivative of vitamin A that regulates important biological processes including cell proliferation and differentiation. In vitro studies have shown that RA is effective in inhibiting growth of HCC cells; however, responsiveness to treatment varies among different HCC cell lines. The objective of the present study was to determine if the combined use of RA (0.1 µM) and cAMP (1 mM), an important second messenger, improves the responsiveness of HCC cells to RA treatment. We evaluated the proliferative behavior of an HCC cell line (HTC) and the expression profile of genes related to cancer signaling pathway (ERK and GSK-3β) and liver differentiation (E-cadherin, connexin 26 (Cx26), and Cx32). RA and cAMP were effective in inhibiting the proliferation of HTC cells independently of combined use. However, when a mixture of RA and cAMP was used, the signals concerning the degree of cell differentiation were increased. As demonstrated by Western blot, the treatment increased E-cadherin, Cx26, Cx32 and Ser9-GSK-3β (inactive form) expression while the expression of Cx43, Tyr216-GSK-3β (active form) and phosphorylated ERK decreased. Furthermore, telomerase activity was inhibited along treatment. Taken together, the results showed that the combined use of RA and cAMP is more effective in inducing differentiation of HTC cells
Potent and Broad-Spectrum Antimicrobial Activity of Analogs from the Scorpion Peptide Stigmurin
Scorpion venom constitutes a rich source of biologically active compounds with high potential for therapeutic and biotechnological applications that can be used as prototypes for the design of new drugs. The aim of this study was to characterize the structural conformation, evaluate the antimicrobial activity, and gain insight into the possible action mechanism underlying it, for two new analog peptides of the scorpion peptide Stigmurin, named StigA25 and StigA31. The amino acid substitutions in the native sequence for lysine residues resulted in peptides with higher positive net charge and hydrophobicity, with an increase in the theoretical helical content. StigA25 and StigA31 showed the capacity to modify their structural conformation according to the environment, and were stable to pH and temperature variation-results similar to the native peptide. Both analog peptides demonstrated broad-spectrum antimicrobial activity in vitro, showing an effect superior to that of the native peptide, being non-hemolytic at the biologically active concentrations. Therefore, this study demonstrates the therapeutic potential of the analog peptides from Stigmurin and the promising approach of rational drug design based on scorpion venom peptide to obtain new anti-infective agents.publishersversionpublishe
Kinin B1 receptor controls maternal adiponectin levels and influences offspring weight gain
Given the importance of the kinin B1 receptor in insulin and leptin hormonal regulation, which in turn is crucial in maternal adaptations to ensure nutrient supply to the fetus, we investigated the role of this receptor in maternal metabolism and fetoplacental development. Wild-type and kinin B1 receptor-deficient (B1KO) female mice were mated with male mice of the opposite genotype. Consequently, the entire litter was heterozygous for kinin B1 receptor, ensuring that there would be no influence of offspring genotype on the maternal phenotype. Maternal kinin B1 receptor blockade reduces adiponectin secretion by adipose tissue ex vivo, consistent with lower adiponectin levels in pregnant B1KO mice. Furthermore, fasting insulinemia also increased, which was associated with placental insulin resistance, reduced placental glycogen accumulation, and heavier offspring. Therefore, we propose the combination of chronic hyperinsulinemia and reduced adiponectin secretion in B1KO female mice create a maternal obesogenic environment that results in heavier pups
